Avaliou-se, por citometria de fluxo, a fagocitose de Staphylococcus aureus conjugados com iodeto de propídio (IP), por leucócitos circulantes obtidos de cinco fêmeas bovinas negativas no sorodiagnóstico para a Leucose Enzoótica Bovina (LEB); de cinco fêmeas infectadas, manifestando linfocitose persistente (LP); e de cinco fêmeas infectadas, porém alinfocitóticas. Observou-se que, entre as amostras dos animais soronegativos, a porcentagem média de células realizando fagocitose (12,90%) não diferiu da observada entre as células dos animais alinfocitóticos (14,70%). Contudo, ambas foram maiores (p=0,047) que aquela verificada entre as células obtidas de animais manifestando LP (7,20%). Além disso, a intensidade média de fagocitose (caracterizada pela intensidade de fluorescência do IP, em valores arbitrários), verificada em leucócitos de animais manifestando LP (17,43) foi menor (p<0,001) que a observada em leucócitos de animais alinfocitóticos (29,50), e que a observada em leucócitos de animais soronegativos (25,18), que não diferiram entre si. Assim, os resultados permitem-nos alvitrar que há alteração na função fagocítica de leucócitos circulantes em animais infectados pelo vírus da LEB, manifestando LP.
O objetivo deste trabalho foi avaliar o perfil microbiológico e celular do leite no período lactante e de involução ativa de ovelhas da raça Santa Inês. Foram avaliadas amostras lácteas de 12 ovelhas durante estes distintos períodos. Realizou-se o exame físico da mama, sendo as amostras lácteas submetidas à contagem de células somáticas (CCS), ao California Mastitis Test (CMT), ao exame microbiológico e aos testes de sensibilidade in vitro dos patógenos encontrados. Foram observados maiores escores do exame físico, CCS, CMT durante o período de involução ativa, além de uma alta persistência da infecção durante estes períodos. O período de involução ativa não se mostrou como um momento de alta susceptilidade. Os estafilococos coagulase negativa representaram o único gênero isolado das glândulas infectadas. Uma alta sensibilidade dos agentes etiológicos envolvidos frente aos diferentes antimicrobianos in vitro foi também observada.
Para realização dessa pós-graduação, muitas pessoas foram fundamentais, pois, colaboraram de diferentes formas, quer com uma palavra amiga na hora exata, quer com algum conhecimento técnico passado, mas, sem dúvida, a união de tudo me impulsionou para mais essa etapa vencida. Pesq. Vet. Bras. 32(12):1225-1229, dezembro 2012 1225 RESUMO.-A Artrite Encefalite Caprina (AEC) e a Linfadenite Caseosa (LC) possuem alta incidência e transmissibilidade em pequenos ruminantes. Como ambas possuem tropismo por monócitos-macrófagos e afetam mecanismos da resposta inata do hospedeiro, acredita-se que a AEC predispõe o animal a infecções por Corynebacteruim pseudo- Caprine arthritis encephalitis (CAE) and caseous lymphadenitis (CL) have high incidence and transmissibility in small ruminants. Since both virus have tropism for macrophages and monocytes and affect the innate immune response, it is believed that CAE can predispose the animal to infection by Corynebacteruim pseudotuberculosis, the etiological agent of CL. To conϐirm this hypothesis, we evaluated phagocytosis from the monocyte-macrophage cells from 30 Saanen goats. Goats were uniformly divided in two groups according to results of agar gel immunodiffusion test for CAE virus (CAEV). Peripheral blood mononuclear cells were isolated by density gradient centrifugation and the monocyte-macrophage cells were isolated from the mononuclear cells by their adhesion properties in plaques. Afterwards, phagocytosis of C. psudotuberculosis was performed for two hours at 37°C, 5% of CO 2 , and assessed by microscopic visualization. There was no difference in the percentage of monocyte-macrophage cells that phagocytozed C. bovis between groups (P=0.41). However, when phagocytosis rates were classiϐied according to the number of C. pseudotuberculosis phagocyted, the percentage of monocyte-macrophage cells that internalized more than 12 bacteria were higher in serologically CAEV positive animals compared to the serologically negative ones (P<0.001). Furthermore, a positive and signiϐicant correlation (r = 0.488; P = 0.006) between the percentage of monocyte-macrophage cells that internalized more than 12 bacteria and the percentage of monocyte that were carrying out phagocytosis was also encountered in serologically CAEV positive goats, however the same were not observed in serologically negative ones. These results demonstrated an alteration in the intensity of C. pseudotuberculosis phagocytosis by monocytes-macrophages from goats infected by CAEV.
