Kadar logam Hg di lingkungan perairan berada pada konsentrasi rendah, yaitu kisaran satuan µg/L. Dengan konsentrasi yang rendah tersebut, maka diperlukan tahapan analisis yang sesuai untuk memaksimalkan hasil analisis. Tahapan analisis logam ini termasuk di dalamnya adalah proses digesti. Digesti adalah proses perombakkan matriks sampel dari senyawa-senyawa yang berbentuk logam organik menjadi logam anorganik yang bebas dengan menggunakan asam kuat baik tunggal maupun campuran. Digesti merupakan tahapan paling penting dalam analisis logam, karena dapat mempengaruhi konsentrasi logam yang akan teranalisis. Digesti ini dipengaruhi oleh reagen, suhu dan waktu. Penelitian ini bertujuan untuk menentukan kondisi digesti terbaik berdasarkan penggunaan reagen, suhu dan waktu digesti dalam sampel sedimen. Sampling dilakukan di Sungai Cikijing Desa Cimanggung pada koordinat 6,9567 oLU; 107,8207 oBT. Sedimen yang digunakan, didestruksi dengan tiga reagen berbeda, yaitu asam nitrat, akuaregia, serta campuran asam nitrat dan hidrogen peroksida. Variasi suhu yang digunakan adalah 25, 60, 85, dan 100 ℃. Untuk variasi waktu yaitu 24, 48, 72, 96, dan 120 menit. Kadar Hg dalam sedimen diuji menggunakan instrumen VGA-AAS. Berdasarkan hasil analisis, digesti dengan reagen akuaregia pada suhu 100 ℃ selama 120 menit memberikan hasil tertinggi dengan kadar Hg sebesar 0,4722 µg/g. Dengan linearitas (R2) sebesar 0,9990, akurasi sebesar 60,55%, presisi sebesar 0,333%, batas deteksi (LOD) sebesar 0,8622 dan batas kuantisasi (LOQ) sebesar 2,8745 yang menyatakan bahwa metode ini dapat diterima (valid).Kata Kunci: optimasi; digesti; Hg; sedimen; VGA-AAS
