In this study, the effects of queen genotype and breeding season on the amount of mandibular pheromone in honeybees were determined. For this purpose, in three different seasons (April, June and September), Caucasian, Italian and Aegean ecotype of Anatolian honeybee queens was reared. 9-oxodec-2-enoic acid (9 ODA), 9-hydroxydec-2-enoic acid (9 HDA), methyl p-hydroxybenzoate (HOB) and 4-hydroxy-3-methyloxphenyethanol (HVA) quantities in the mandibular glands of the queens collected from nucs after
. m. ligustica) x Aegean crossbred honeybee colonies on the royal jelly production in the conditions of South Aegean region. In addition to that of the effects of number of grafted larvae on royal jelly production was also determined. The average acceptance rate (71.1±2.72% and 70.6±2.23%), and total colony yields of royal jelly (28.5±2.70 g and 26.7±4.6 g) were found similar, but amount of royal jelly in per cell (229±8.9 mg and 216±10.2 mg) were found significant (P<0.05) in Aegean ecotypes and Italian x Aegean crossbred honeybees, respectively. The differences between the colonies for the number of grafted larvae, the average acceptance rate, amount of royal jelly in per cell were found significant (P<0.05). As the number of grafted larvae increased, the rate of acceptance of colonies decreased, but the total royal jelly production of colonies were increased. Aegean ecotype was found to be suitable for royal jelly production under the conditions of South Aegean region.
Chaste tree and pine honeys were bought from three different producers (natural) and three local markets (commercial). The honey samples were stored for 30 months at room temperature. The shelf life of the samples was estimated for indices of HMF and diastase activity. The values for HMF formation and diastase deactivation fitted to a zero-order reaction. The highest rate of HMF formation was observed in the chaste tree honey as 2.674±0.013 mg/kg/month while the least value was observed in the pine honey as 0.435±0.037 mg/kg/month. The maximum diastase deactivation was determined in the natural chaste tree honey as 0.613±0.000 DU/kg/months and the minimum was 0.318±0.002 DU/kg/month for the commercial chaste tree sample. In terms of HMF, all honeys except commercial chaste tree honey, have exhibited longer shelf life than 30 months of storage. The results showed that the shelf life of the commercial honeys depend on the botanical origin, pH, electrical conductivity, temperature and storage.Keywords: Chaste tree, pine honey, shelf life, HMF, diastase. HAYIT (Vitex agnus castus) VE ÇAM BALININ RAF ÖMRÜNÜN BELİRLENMESİ ÖzHay›t ve çam ballar› üç farkl› üretici (do¤al) ve üç farkl› yerel marketten (ticari) sat›n al›nm›fl ve bal örnekleri 30 ay oda s›cakl›¤›nda depolanm›flt›r. Bal örneklerinin raf ömrü HMF miktar› ve diastaz aktivite indeksleri üzerinden tahmin edilmifltir. HMF oluflumu ve diastaz deaktivasyonu s›f›r›nc› dereceden kinetik modele uyumlu bulunmufltur. En yüksek HMF oluflum h›z› hay›t ballar›nda 2.674±0.013 mg/kg/ay olarak hesaplan›rken en düflük h›z çam bal›nda 0.435±0.037 mg/kg/ay olarak belirlenmifltir. Diastaz say›s› bak›m›ndan ise, en yüksek (0.613±0.000 DU/kg/ay) ve en düflük (0.318±0.002 DU/kg/ay) deaktivasyon h›z› do¤al ve ticari hay›t ballar›nda belirlenmifltir. Ballar›n 30 ay depolanmas› sonunda HMF bak›m›ndan ticari hay›t bal› hariç di¤er tüm ballar daha uzun raf ömrüne sahip olmufltur. Özet: Bu araştırma, Aydın yöresinde üretilen hayıt balının bazı kalite parametrelerinin belirlenerek, çam ve karışım çiçek balları ile karşılaştırılması amacıyla gerçekleştirilmiştir. Çalışmada, Aydın ili sınırları içinde üretilmiş ve yeni hasat edilmiş hayıt balı, çam balı ve yöre arıcılarının Orta Anadolu'da ürettiği karışım çiçek ballarının bazı kimyasal parametreleri belirlenmiştir. Üç bal grubunda her bir özelliğe ait veriler basit varyans analizi (ANOVA) ile değerlendirilmiştir. Tüm özelliklerin bir arada değerlendirilmesi için, çok değişkenli varyans analizi (MANOVA) ve diskriminant analizi yapılmıştır. Çalışmada, üzerinde durulan özellikler bakımından gruplar arası farklar önemli bulunmuştur (P<0.05). Hayıt, karışım çiçek ve çam ballarında sırasıyla; nem oranı %15.95±0.040, %16.86±0.341 ve %17.11±0.273; invert şeker (glikoz+früktoz) oranı %73.76±0.572, %60.92±1.195 ve %60.9±0.426; früktoz/ glikoz oranı %1.16±0.03, %1.15±0.5 ve %1.36±0.104; pH değeri 3.75±0.033, 4.08±0.015 ve 4.02±0.143; serbest asitlik (meq/ kg) 26.6±0.3, 36.03±0.534 ve 27.3±0,36; elektriksel iletkenlik (mS/cm) 0.42±0.001, 0.73±0.009 ve 1.17±0.08; diastaz sayısı 27.3±0.208 ve 23.15±4.10 ve 24.07±1.17, HMF miktarı (mg/kg) ise 4.92±0.08, 0.56±0.037 ve 0.93±0.0305 olarak belirlenmiştir. Ayrıca hayıt ve karışım çiçek ballarında prolin miktarı da sırasıyla 980±37.4 mg/kg ve 922±45.8 mg/kg olarak saptanmıştır. Araştırma verilerine uygulanan çok değişkenli varyans analizine göre, tüm gruplar arası uzaklıklar önemlidir (P<0.01). Diskriminant analizi sonucuna göre, çam balı diğer ballara en uzak grubu oluşturmuştur. Ayrıca, hayıt balı ile karışım çiçek balları da birbirinden uzakta dağılım göstermişlerdir. Araştırma sonuçları özgün markerlerinin yokluğunda da rutin kimyasal parametreler kullanılarak hayıt balının diğer ballardan ayrı küme oluşturduğunu göstermiştir.
ÖZETAydın'da 2012-2014 yılları arasında yürütülen bu araştırmanın amacı Ege Bölgesi koşullarında bireysel ve kitlesel ana arı (Apis mellifera) kışlatma başarısını karşılaştırmaktır. Ardı sıra 2 yıl Anadolu arısı Ege ekotipi (AE) ve İtalyan ırkından (It) yetiştirilen ana arılar banka kolonilerinde kitlesel ve çiftleştirme kutularında ve ruşetlerde (kontrol) bireysel kışlatılmıştır. Kitlesel kışlatma için birinci yıl AE ve It ırkından ana arılı (ABK) ve ana arısız (ASBK) banka kolonileri, ikinci yıl ise ana arısız banka kolonileri (ASBK) kullanılmıştır. Birinci yıl, her 2 ırktan (AE: 4 koloni, It: 4 koloni) 8 banka kolonisinde (ABK: 4 koloni, ASBK: 4 koloni) toplam 112 ana arı, ikinci yıl ise, 4 ASBK (AE: 2 koloni, It: 2 koloni)'de toplam 56 ana arı kitlesel kışlatılmıştır. Her 2 yılda 20 çiftleştirme kutusunda 10 It ve 10 AE ana arı bireysel kışlatılmıştır. Yaklaşık 3 ay süren her kışlatma dönemi sonunda her grupta ana arı yaşama gücü (%) saptanmıştır. Kitlesel kışlatmanın ana arıların üreme performansını etkileyip etkilemediğini test etmek için birinci yıl kitlesel ve bireysel kışlatılmış ana arılardan rasgele 30'u (banka kolonisi: 10, çiftleştirme kutusu: 10, ruşet: 10) üretim kolonilerine kabul ettirilmiştir. Ana arı yaşama gücü; banka kolonisinin ırkına (P>0.05) ve kışlatılan ana arının genotipine (P>0.05) göre değişmemiştir. Fakat banka kolonisinin tipi ana arı yaşama gücünü etkilemiştir (P<0.01). Birinci yıl, ASBK'da kışlatılan ana arıların % 64.3'ü, ABK'da kışlatılan ana arıların ise % 17.9'u kıştan çıkabilmiştir (P<0.01). Her 2 yılda da banka kolonilerinde ana arı yaşama gücü (% 64.3 ve % 71.5) çiftleştirme kutularında yaşama gücüne (% 100 ve % 100) göre daha düşük (P<0.01) bulunmuştur. Kitlesel kışlatılan ana arıların test kolonilerinde yumurtlama performansları bireysel kışlatılanlardan farklı bulunmamıştır (P>0.05).
In this study, the retinue pheromones (9-ODA, R and S HDA, HOB, HVA, methyl oleate, conyferyl alcohol, palmityl alcohol and linolenic acid) of Caucasian and Aegean Ecotype of Anatolian queens stored in Kirchain mating boxes and Reservoir colonies were determined. After the queens beginning egg lying for 102 days was collected individually in eppendorf tubes and stored at -20°C until the analysis (group A). Second group queens (10 Aegean and 10 Caucasian) collected from Kirchain mating boxes in the same day stored 25 days in the reservoir colonies (B group). Also, the queens in the last group (10 Aegean and 10 Caucasian) were collected after beginning egg lying for 352 days (C group). After all, the queens in the group B and C put individually in eppendorf tubes and stored at -20°C until the analysis. Pheromones in the head, chest and abdomen of the stored queens were identified by gas chromatography. According to results, the differences in the values of 9 ODA and 9 HDA for were statistically insignificant for queen breeding season, application groups (A, B and C) and genotypes, but differences in body parts were determined to be statistically significant. The differences between HOB and HVA values were also statistically insignificant for queen breeding season, application groups, genotype and body parts. In the study, the effects of application group, genotype and body parts on the amount of linoleic acid were determined to be statistically significant. Linolenic acid secreted higher from the abdomen than that of the head and chest, and the differences among the body parts were also statistically significant. In general, 9 ODA and 9 HDA amounts were secreted the highest in the head of the queens, HOB and HVA were secreted equally amounts in the three parts of the body, and linolenic acid was secreted the highest from the abdomen. The queens allowed egg lying for 352 days in Kirchain mating boxes were secreted 9 ODA, 9 HDA, HOB and HVA more than the queens allowed egg lying for 102 days and stored reservoir colonies.
