This research aims to determine the optimum potassium diformate (KDF) dosage which was added to feed to increase the immune status of goldfish (Carassius auratus L). This study was conducted from September 31 - November 15, 2019 at the Laboratory of Aquaculture and Molecular Biotechnology Laboratory of the Faculty of Fisheries and Marine Sciences, University of Padjadjaran. The research method used is the experimental Complete Random Design (CRD) with 4 treatments and 3 replications. The treatments are A (without KDF/control), B (0.1% KDF), C (0.3% KDF) and D (0.5% KDF). The observed parameters are survival rate, total leukocyte count (white blood cells), total erythrocyte count (red blood cells) and gross clinical sign. Observations were made after 30 days of KDF and post-test challenge by Aeromonas hydrophila for 14 days. Data on the number of total leukocyte count and total erythrocyte count were analyzed using F test and Duncan's advanced test at a 95% confidence level, while the gross clinical sign data were analyzed descriptively. The results showed that the KDF 0.3% was the optimum dose and was very effective in optimizing the performance of the goldfish immune system optimally, seen from the number of white blood cells and red blood cells which experienced the highest increase of 23.48% and 9.30%. After the challenge test, fish that were given KDF 0.3% had the highest survival rate of 46.67%. In addition, the process of recovering the number of white blood cells and red blood cells can be faster than other treatments, with a value of 12.85×104 cells mm3-1 and 0.96×106 cells mm3-1, marked also by the healing of clinical symptoms in the morphology of goldfish.
This research was carried out from June to November 2017. This research aims to analyze genetic relationship of tilapia (Oreochromis niloticus) nirwana strain which is cultured in Aceh Besar (Aceh Province) and in Wanayasa (West Java Province) with RAPD-PCR technique. The DNA Isolation Activity of tilapia nirwana strain was carried out in The Biotechnology Laboratory, Building No.4, FPIK UNPAD, Indonesia and the PCR process was carried out in The Microbiology and Biotechnology Laboratory, Building No.3 FPIK UNPAD, Indonesia. The purpose of this research is to determine the level of genetic relationship of tilapia nirwana strain cultured in Aceh Besar and Wanayasa and to find out whether tilapia nirwana strain in Aceh Besar can be used as a genetic stock. The results showed that genetic relationship level of tilapia nirwana strain cultured in Aceh Besar with those cultured in Wanayasa is 67%. The existence of environmental differences results in genetic diversity between Aceh Besar and Wanayasa tilapia nirwana strain. Factors that causing grouping movement are external factors such as: Geographical isolation and habitat fragmentation as well as internal factors such as mutation, natural selection, genetic drift, and gene flow. Tilapia nirwana strain that is cultured in Aceh Besar produce polymorphic bands, so it can be used as a new genetic stock to stabilize the tilapia nirwana strain production.
Penelitian ini bertujuan untuk mengetahui pengaruh kepadatan pada pengangkutan dengan suhu rendah terhadap kadar glukosa darah dan kelulusan hidup ikan nila (Oreochromis niloticus). Penelitian dilaksanakan pada bulan April sampai Oktober 2016 di Laboratorium Basah dan Kolam Percobaan, Ciparanje Unpad. Metode penelitian yang digunakan yaitu metode eksperimental dengan menggunakan Rancangan Acak Lengkap (RAL) yang terdiri dari empat perlakuan dan empat ulangan. Perlakuan berupa kepadatan ikan sebanyak 5 ekor/L, 10 ekorLl, 15 ekor/L dengan pemingsanan, serta 10 ekor/L tanpa pemingsanan sebagai pelakuan kontrol. Pemingsanan dilakukan dengan cara penurunan suhu air menjadi 10±0,1 ºC. Pengamatan yang dilakukan terhadap kelulusan hidup setelah pengangkutan, kadar glukosa darah, kelulusan hidup setelah pemeliharaan pasca pengangkutan. Hasil penelitian menunjukkan perlakuan dengan kepadatan 15 ekor/l menunjukan hasil terbaik, yaitu kelulusan hidup setelah pengangkutan 98,33 %, kadar glukosa darah 192 mg/dL serta kelulusan hidup setelah pemeliharaan pasca pengangkutan 88,33 %.
This research was conducted from May to September 2018 at the Biotechnology Laboratory, Faculty of Fisheries and Marine Sciences, University of Padjadjaran. Fish samples were taken from Jatigede Reservoir, Sumedang, West Java. The hampala fish consisted of female hampala barb with gonadal maturity level (GML) IV, GML V, and GML VI. Ovarian histological preparations were made at the Animal Microtechnical Laboratory, Faculty of Mathematics and Natural Sciences, Universitas Padjadjaran. This research aimed to determine the stages of ovarian development of hampala barb, and determine the types of spawning. The hampala barb in the Jatigede Reservoir had five stages of oocyte development, namely chromatin nucleolus stage, perinucleolus stage, cortical alveolus stage, vitellogenesis, and oocyte maturation with multi-spawner group synchronous type of ovarian development.
West Java Province is one of the potential areas of freshwater fisheries resources development in Purwakarta District. The utilization of development resources is directed to achieve comparative advantage and competitive advantage as an effort to encourage the growth of regional economy. Purwakarta is one area that has the potential of the aquaculture potential to be developed. One type of aquaculture that has a high economic potential such as tilapia nirwarna fish (Oreochromis niloticus), but in the institutional aspects of marketing there are still constraints from producers to consumers. This research aims to analyze marketing institutional of Nila seed (Oreochromis niloticus) through Integrated Supply Chain Management approach. The type of data used in this research is primary and secondary data. The analytical tool used marketing channels and supply chain risk by testing the validity and reliability of data. The technique of taking respondents using snowball sampling with the number of respondents 30 consisting of suppliers of nirwarna tilapia fish breeders to nirwarna tilapia seed cultivators. Based on the results of the research concluded that the analysis of tilapia supply chain risk has a value of 1.0 means it has a high risk. Based on the research results of the aquaculture business in Purwakarta is divided into two types of business, namely the enlargement effort and fish hatchery business. The marketing institutional of tilapia nirwarna seeds through Integrated Supply Chain Management approach in Purwakarta district through hilirization of tilapia fishery through the stage of input production supply and downstream of tilapia fishery fishery through fish farm institution through production and distribution output stage.
Tujuan penelitian ini adalah untuk mengetahui kelangsungan hidup dan pertumbuhan larva rajungan (P. pelagicus) melalui pemberian nauplius Artemia yang diperkaya dengan minyak ikan dan minyak jagung. Penelitian ini dilakukan di Panti Pembenihan Family Portunidae, Balai Besar Perikanan Budidaya Air Payau (BBPBAP) Jepara, Jawa Tengah. Larva yang baru menetas (zoea 1) dipelihara dalam wadah plastik 20 L yang diisi 15 L air laut dengan kepadatan larva 50 ind/L. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) dengan enam perlakuan dengan tiga kali ulangan, yaitu : A (tanpa pengayaan), B (100% minyak ikan + 0% minyak jagung), C (75% minyak ikan + 25% minyak jagung), D (50% minyak ikan + 50% minyak jagung), E (25% minyak ikan + 75% minyak jagung), F (0% minyak ikan + 100% minyak jagung). Parameter utama yang diamati adalah kelangsungan hidup dan pertumbuhan panjang mutlak. Analisis statistik dilakukan dengan analisis ragam (ANOVA) dan uji jarak berganda Duncan taraf kepercayaan 95%. Hasil penelitian menunjukkan bahwa perlakuan memberikan pengaruh nyata terhadap kelangsungan hidup dan pertumbuhan larva P. pelagicus. Pemberian nauplius Artemia yang dipperkaya 75% minyak ikan dan 25% minyak jagung mempunyai kelangsungan hidup terbaik sebesar 12,89% dengan pertumbuhan panjang mutlak sebesar 2,12 mm.
