Nosemosis bal arılarında (Epis mellifera) sıkça görülen küresel bir hastalıktır. Hastalığın etkeni fungal mikroorganizmalar olan Nosema apis ve Nosema ceranae'dir. Bu çalışmada Siirt ve Şanlıurfa'da bal verimi düşük ve zayıf (popülasyonu düşük) arı kolonilerinde Nosema spp. sporlarının varlığının mikroskobik olarak muayene edilmesi ve pozitif bulunan örneklerden, multipleks PCR (Polimeraz zincir reaksiyonu) ile etkenin tür düzeyinde tespiti amaçlandı. Her iki ilde toplam 8 arılıkta 44 kovandan alınan 880 arı numunesi mikroskobik olarak spor varlığı yönünden incelenmiş ve 19'unda (%43,18) nosema sporları tespit edilmiştir. Multipleks PCR ile bu sporların tamamının N. ceranae olduğu belirlendi. Sonuç olarak, nosemosis'in bölgede arıcılık işletmelerinde önemli bir problem olduğu, bu nedenle nosemosis yönünden gerekli tedbirlerin alınması gerektiği kanısına varıldı.
Dermatophytes infect tissues containing keratin in humans and animals, causing dermatophytosis infection. Trichophyton verrucosum is the most common agent of bovine dermatophytosis cases. Trichophytosis causes big economic lossess throughout the world and also threatens human health by being a zoonosis. T. verrucosum is usually isolated from cattle. Cattle are the natural reservoirs of this agent. The aim of this study is to isolate disease-causing dermatophytes in cattle and to carry out molecular separation of Internal Transcribed Spacer (ITS) regions of the isolated T. verrucosum strains by PCR-Restriction Fragment Lenght Polymorphism (PCR-RFLP). For this purpose, 90 samples were taken from the cattle with dermatophytosis for cultural examination. As a result of the culture of these samples, 35 (38.8%) T. verrucosum were isolated and identified. DNA isolation of these strains was made and amplification of ITS regions was performed. It was only one RFLP profile was found according to the results of RFLP analysis of T. verrucosum strains using MvaI and HinfI enzymes. At the end of study, it was founded that the isolated T. verrucosum strains showed a single profile by PCR-RFLP analysis and PCR-RFLP was a useful tool for the molecular characterization of the strains. İt was also concluded that PCR-RFLPs of strains from different regions or even from different countries might be necessary in order to detect different profiles of the tested samples.
Bu çalışmada amaç, Mycoplasma gallisepticum (MG), Mycoplasma synovia (MS) ve Mycoplasma meleagridis (MM) etkenlerini bir multipleks PCR yöntemiyle tek seferde tanımlamak ve bu yöntemin rutin teşhiste kullanılabilirliğini araştırmaktır. Çalışmada laboratuvara getirilen ve klinik belirtileri mikoplazma infeksiyonları yönünden şüpheli, tavuk ve hindilerden aseptik şartlarda alınan toplam 100 svab örneği kullanıldı. Bu örnekler Frey's besiyerinde inkübe edildi ve buradan DNA ekstraksiyonu yapıldı. Yapılan multipleks PCR testi sonucunda tavuklarda (n=50) MG ve MS için sırasıyla 9 (%18) ve 5 (%10) pozitiflik saptandı. 2 tavukta (%4) hem MG hem de MS beraber pozitif bulundu. Tavuk numunelerinin hiçbirinde MM için spesifik bant oluşumu tespit edilmedi. Hindilerde ise (n=50) MG, MS ve MM için pozitif hayvan sayısı sırasıyla 6 (%12), 4 (%8) ve 3 (%6) olarak tespit edildi. MG ve MS bantlarının beraber tespit edildiği hindi sayısı 12 (%24) iken, 1 hindide (%2) MG ve MM, 3 hindide ise (%6) MM ve MS etkenleri beraber izlendi. Numunelerin hiçbirinde üç etkene ait spesifik bant oluşumu gözlenmedi. Sonuç olarak Şanlıurfa bölgesinde kanatlı türlerine ait önemli patojen mikoplazma etkenlerinin bulunduğu, multipleks PCR testi ile kısa sürede ve yüksek duyarlılıkta çok sayıda etkenin aynı anda ve daha hızlı tespitinin sağlandığı ayrıca koruma ve kontrol önlemlerinin alınmasında başarıyla kullanılabileceği sonucuna varıldı.
