Background: Escherichia coli O157:H7 (E. coli O157:H7) have frequently been associated with food borne illness and are considered as most serious of known food borne pathogens leading to severe illnesses and high mortality rates in humans. Most of outbreaks were traced to raw meat and raw milk consumption, as well as to dairy products such as yogurt and cheese derived from raw milk. Results: Out of 200 samples examined, 40 (20%) and 7 (3.5%) of the samples were positive to E. coli and E. coli O157: H7 respectively. The highest isolation of E. coli was from cheese (40%), followed by raw milk (32%), yogurt (25.71%), beef (13.84%), and pasteurized milk (0%). Among E. coli O157:H7 isolates, the highest isolation was from raw milk (12%) followed by cheese (5.71%) and meat (3.07%). However, no E. coli O157:H7 was isolated from pasteurized milk and yogurt. Antibiotic susceptibility profile showed that E. coli was resistant for vancomycin (89.74%), ampicillin (76.92%) and streptomycin (69.23%). The analysis showed that, 92.5% of isolates showed multidrug resistance comprising 2-4 antimicrobials. Conclusion: The occurrence of E. coli O157:H7 and its multiple antibiotic resistant profiles shows a risk for public health and food safety as well as animal production. These findings stress the need for an integrated control of E. coli O157:H7 from farm production to consumption of food of animal origin.
A cross-sectional study was conducted to estimate the prevalence of Staphylococcus aureus in raw milk and swab, to assess the antimicrobial susceptibility profile, milk handling practice, and its associated risk factors in selected dairy farms of Mukaturi and Sululta Town, Oromia Region, Ethiopia. A total of 247 samples collected from dairy farms were examined using standard microbiological techniques. The antimicrobial susceptibility profiles of the isolates were also investigated. The possible risk factors for Staphylococcus aureus contaminations in milk were evaluated through a structured questionnaire. Overall, 16.6% (n= 41) of the samples were positive for S. aureus. The prevalence of S. aureus was 15.3% from udder milk and 25%, 20%, and 10% from milkers’ hand, milking bucket, and drying towel swab, respectively. The prevalence of Staphylococcus aureus in milk showed statistically significant variation with respect to age (p≤ 0.001), parity (P≤ 0.001), drainage condition of milking area (P=0.035), study sites (P=0.035), and management system (P=0.035). Majority of the isolates were found resistant to penicillin G (97.6%) and amoxicillin (43.9%). According to this study, 12/28(42.9%) Staphylococcus aureus positive raw milk samples had 104-105cfu/ml S. aureus count, which is above the recommended level for human consumption. 47.1% of milking persons store milk at room temperature temporarily (between 6 and 12 hrs) till transport to collection center with no means of cooling aid. From consumers 25.6% had no health risk associated with raw milk consumption or aware of milk borne disease associated with drinking raw milk. Thus, 60.5% of milk users had habit of raw milk consumption. The study revealed a prevalence of S. aureus, poor milk handling practices, raw milk consumption behavior in study area. Proper handling and hygiene decrease milk contamination by S. aureus and make it safe for human consumption.
Background External infections involving the skin and wound are the most frequent complications affecting humans and animals. Medicinal plants play great roles in the treatment of skin and wound infections. This study was aimed to evaluate the in vitro antibacterial activity of crude methanolic extracts of nine medicinal plants. Methods Agar well diffusion and broth dilution methods were used to determine the antibacterial activity of nine Ethiopian plants against four bacterial species including Staphylococcus aureus, Pseudomonas aeruginosa, Escherichia coli, and Klebsiella pneumoniae. Results Among the tested plants, seven (Cissus quadrangularis, Commelina benghalensis, Euphorbia heterophylla, Euphorbia prostrate, Momordica schimperiana, Trianthema spp., and Solanum incanum) were found to exhibit considerable antibacterial activity against at least one of the test bacteria. The extracts of C. quadrangularis, E. heterophylla, and E. prostrata had a wide spectrum of antibacterial activities against test bacterial strains while the extracts of Grewia villosa and Schinus molle did not show any inhibitory activity. Clinical isolate and laboratory strain of S. aureus showed the highest susceptibility to highest concentration (780 mg/mL) of E. prostrata with a zone of inhibition of 21.0mm and 22.3mm, respectively. Conclusion This study indicates clear evidence supporting the traditional use of seven plants in treating skin and wound infections related to bacteria.

Background: Bacillus cereus is a spore-forming bacterium that is widely distributed in the environment and can be detected in a variety of raw milk and milk products especially in vegetative form, which exposed directly in contact with the soil and produces toxins which caused food borne illness and considered as a significant public health hazard. A cross-sectional study was carried out to determine the prevalence and load of Bacillus cereus vegetative cells in raw bulk milk sold in market places at Bate, Haramaya and Aweday Towns, Eastern Ethiopia as well as to assess the in-vitro antimicrobial susceptibility of isolates. A total of 103 raw milk samples were collected and examined for the presence of Bacillus cereus by using Bacillus cereus selective agar media. The isolated presumptive colonies were further analyzed using standard microbiological methods to confirm the presence of Bacillus cereus. Spreading plate method was employed to count the bacteria from milk samples based on 10 fold serial dilutions. For antimicrobial susceptibility test, the disc diffusion test was employed using commercially available 10 antimicrobial discs. Results: The overall prevalence's of Bacillus cereus in raw milk samples was 38.8%. Milk collected from Aweday market showed higher occurrence (46.9%) than from Bate (40%) and Haramaya town (31.7%).
