In the current study, we evaluated total phenolic content, antioxidant activity (by using β-carotene bleaching assay), and antibacterial activity of aqueous and methanol extracts of the aerial part of the Rumex crispus L. (Polygonaceae) naturally grown in the Eastern Anatolia region of Turkey. The aerial part of the plant had high total phenolic content (56.31 µg/mg DW). The antioxidant activity of aqueous and methanol extracts of the aerial part of R. crispus L., BHA, and BHT were found to be 92.35%, 95.49%, 98.16%, and 96.66%, respectively. Although the antioxidant activity of the aqueous and methanol extracts of R. crispus was lower than that of the BHA and BHT, the difference between these was not statistically significant, p < 0.05. The methanol extract possessed strong antibacterial activity against Agrobacterium tumefaciens, Bacillus cereus, Bacillus subtilis, Pseudomonas corrugate, Pseudomonas syringae pv. tomato, Salmonella typhimurium, Serratia liquefaciens, Vibrio cholerae, Yersinia frederiksenii, and Yersinia pseudotuberculosis. Therefore, the aerial part of R. crispus can be used as an effective and safe source of antioxidants and antibacterial agent.
This study was carried out to identify pathogenic bacteria and fungi on mistletoe (Viscum album L.) and investigate their potential use in biological control of this parasitic plant. For this purpose, a total of 48 fungal isolate and 193 bacterial strains were isolated from contaminated V. album during the summers [2005][2006]. The isolated bacterial strains and fungal isolates were identified by using the Sherlock Microbial Identification System (MIS; Microbial ID, Newark) and microscopic methods, respectively. The bacterial strains that induced hypersensitive reaction (HR) on tobacco (Nicotiana tabacum L.) and fungal isolates were tested for pathogenicity on young shoots of mistletoe by using injection methods. The pathogenic bacterial strains and fungal isolates were also tested for their activity against mistletoe using spray methods. Five bacterial strains (two Burkholderia cepacia, one each of Bacillus megaterium, Bacillus pumilus and Pandoraea pulminicola) were HR and pathogenicity positive when injected but none of them when sprayed on mistletoe. When fungi were injected, 32 isolates were pathogenic but only thirteen when sprayed on mistletoe. Alternaria alternata VAS¸-202, VAS¸-205, VAS¸-217 and Acremonium kiliense VA-11 fungal isolates were the most effective ones and caused strong disease symptoms on mistletoe. The present study is the first report on the efficiency of potential biocontrol agents against mistletoe in Turkey.
A b s t r a c tIn this study, we aimed to determine chemical composition and antibacterial activities of Satureja hortensis and Calamintha nepeta against to 20 phytopathogenic bacteria causing serious crop loss. The essential oils of S. hortensis and C. nepeta were isolated by the hydrodistillation method and the chemical composition of the essential oils were analyzed by GC-MS. The antibacterial properties of the essential oils were evaluated against 20 phytopathogenic bacteria through Disc diffusion assay and micro dilution assay. The results revealed that the essential oils of S. hortensis and C. nepeta have significant antibacterial activity. Furthermore, the findings of the study are valuable for future investigations focusing on the alternative natural compounds to control plant diseases. Despite the number of studies describing metal hyper-accumulating plants and their associated bacteria in various regions and countries, there is no information on rhizosphere microbial potential of the Turkish serpentine soils. This study aimed to explore the rhizosphere microbial diversity of Ni-resistant, hyper-accumulating plants grown on Ni-rich soils and their metal tolerance-resistance characteristics. One hundred ninety-one locations were visited to collect soil and plant samples from different serpentine regions of Western Turkey. Following bioavailable and total Ni analysis of collected samples, the seeds of the selected plants with higher Ni content were taken to the growth/germination test in a range of serpentine soils in a growth chamber condition. In order to investigate the rhizosphere microbial diversity, Isatis pinnatiloba and Alyssum dasycarpum which were able to germinate and grow well in the preliminary tests, were introduced to 6-month greenhouse experiment in the range of three serpentine soils with higher bioavailable Ni content. I. pinnatiloba had a better stimulatory effect on the rhizosphere microbial diversity. A total of 22 bacterial isolates were identified from different soil conditions in the end of experiment. Following microbial identification and confirmation tests, 11 isolates were found to be resistant and tolerant to the increasing concentrations of Ni, Pb, Cd and Zn in the range of 50-2,000 mg L( - 1), which was considerably higher than those indicated by earlier studies. The strains isolated and identified from the Turkish serpentine soils were the members of genera Arthrobacter, Bacillus, Microbacterium and Staphylococcus.
In this study, the antibacterial effect of Origanum rotundifolium Boiss's essential oil was evaluated against 20 plant pathogenic bacteria. The essential oil (EO) of O. rotundifolium was hydrodistillated from wild O. rotundifolium plants that grown in Eastern Anatolia region of Turkey. Then, 10 chemical compounds of EO were identified via gas chromatography/mass spectrometry (GC-MS). The main compounds were determined as thymol (40.86 %-GC), carvacrol (43.62 %), p-cymene (5.95 %) and borneol (2.49 %). According to the results of the study, the essential oil of O. rotundifolium exhibits considerable antibacterial effect against pathogenic bacteria that cause significantly crop loss.
