The ovaries of the adult cochineal scale insect, Dactylopius coccus Costa (Hemiptera: Coccoidea: Dactylopiidae) are made up of more than 400 short ovarioles of the telotrophic type. The ovarioles develop asynchronously. The ovarioles consist of a germarium with six or seven nurse cells, a vitellarium with an oocyte, and pedicel. A terminal filament is lacking. A maturing oocyte was attached to the trophic core by the trophic cord during previtellogenesis and most of vitellogenesis.
RESUMEN. Debido a su importancia económica y con la finalidad de profundizar en el conocimiento sobre la biología reproductiva del gusano rojo del maguey Comadia redtenbacheri (Hammerschmidt) (Lepidoptera: Cossidae), se determinó la morfología del sistema reproductor de la hembra de esta especie, el cual presenta en general las características básicas de otros Lepidoptera, con algunas particularidades específicas. La fecundidad potencial tuvo un valor promedio de 104 ovocitos por hembra. En la base de la bursa copulatrix se presentó un pequeño divertículo o lóbulo, y dentro de ella se encontró un solo espermatóforo. La espermateca presentó tanto un utrículo como una lagena. La porción glandular de las glándulas accesorias es inusualmente larga en comparación a otros Lepidoptera.ABSTRACT. Due to the economic significance of the agave red worm Comadia redtenbacheri (Hammerschmidt) (Lepidoptera: Cossidae), and with the objective of increasing the knowledge about its reproductive biology, the morphology of the female reproductive system was studied. In general, it is similar to that of other Lepidoptera, with a few differences. The average potential fecundity was 104 oocytes per female. A small diverticulum or lobe was present in the base of the bursa copulatrix; inside the bursa, a single spermatophore was found. The spermatheca has a utriculus and a lagena. The glandular portion of the accessory glands is unusually long compared with other Lepidoptera.
Using transmission electron microscopy, light microscopy (Giemsa MayGrumwald), and the Periodic Acid-Schif (PAS) and Sudan Black B staining techniques, hemocytes in the hemolymph of adult female Dactylopius coccus were characterized. The following, in order of abundance, were found: granulocytes, plasmatocytes, prohemocytes, and oenocytoids. Granulocytes varied in size with granulations in the cytoplasm, a large quantity of mitochondria, rugose endoplasmatic reticulum, ribosomes and vesicles, central or exocentric, spherical and occasionally lobulate nucleus. Plasmatocytes were polymorphic with irregularities in the plasma membrane; cytoplasm contained mitochondria, rugose endoplasmatic reticulum and vesicles, and exocentric, spherical, or irregular nucleus. In both types of hemocytes, scant polysaccharides and lipids were found. Prohemocytes were small and spherical with homogeneous cytoplasm and large exocentric nuclei. Oenocytoids were oval or irregular with dense homogeneous cytoplasm and elongated exocentric nuclei. Se realizó la descripción ultraestructural del germario de hembras adultas de Dactylopius coccus Costa, 1829, mediante microscopía electrónica de transmisión. El germario está rodeado por una cubierta epitelial interna y por una túnica propia. Contiene además grandes células nutricias con núcleos de forma irregular, cuya membrana estaba perforada con gran cantidad de poros nucleares, observándose además uno o más nucléolos. Por otra parte, su citoplasma contenía una gran cantidad de ribosomas y mitocondrias, pero escaso retículo endoplásmico rugoso. No se observaron retículo endoplásmico liso ni complejos de Golgi. En las células nutricias de las hembras de dos días se observaron bacterias, aparentemente simbiontes, quedando por determinar su posible función en la fisiología de los ovarios. En algunas células nutricias de las hembras de diez días se observaron pequeños grupos de laminillas anulares, las cuales podrían estar relacionadas con un proceso degenerativo de dichas células. Los cordones tróficos se conformaron por una gran cantidad de microtúbulos y entre ellos se observaron numerosos ribosomas y numerosas mitocondrias, muchas de ellas de forma alargada. An ultrastructural description of adult females tropharium of Dactylopius coccus Costa, 1829, by electron microscopy transmission was realized. The tropharium is surrounded by an inner epithelial sheath and by a tunica propria. Furthermore, it contains large nurse cells with irregular-shaped nuclei and their nuclear membrane is abundantly perforated with nuclear pores; further one or more nucleoli were seen. Moreover their cytoplasm contained large numbers of ribosomes and mitochondria, but scarce rough endoplasmic reticulum. Neither smooth endoplasmic reticulum nor Golgi complex were observed. In the nurse cells of the females of two days-old, bacteria apparently symbionts were found, but their possible role in the ovarian physiology is unknown. Some nurse cells of females at ten days-old, had small groups of annular lamellae, which could be related with a degenerative process in these cells. The trophic cords were made of great number of microtubules and between them numerous ribosomes and mitochondria, many of them of elongated form.
La cochinilla Dactylopius coccus es un insecto de gran importancia industrial para la extracción del colorante rojo ácido carmínico. Se determinó el efecto de la cópula sobre la maduración del ovario. Para ello, se emplearon hembras copuladas y hembras vírgenes de tres meses de edad, contados desde la infestación de los cladodios de Opuntia ficus indica (en Montecillo, México). Se evaluaron las características morfofisiológicas del grado de madurez de sus ovariolas. La cópula no fue necesaria para que diera inicio la maduración de las ovariolas, ya que en las hembras vírgenes un bajo porcentaje de sus ovocitos lograron culminar adecuadamente la vitelogénesis y la coriogénesis; sin embargo, la cópula promovió ambos procesos, ya que aumentó notablemente la proporción de los ovocitos maduros que se producirán en las hembras copuladas. Del mismo modo, la falta de cópula en D. coccus fue un factor que indujo la degeneración de las ovariolas y, en consecuencia, la reabsorción de sus ovocitos.
