It is generally assumed that radiation-induced micronuclei (MN) in cytokinesis-blocked cells are an expression of cellular radiosensitivity. Therefore, radiosensitive cells should have a high frequency of MN and radioresistant cells should show lower levels. We have irradiated cells of a panel of 13 neuronal cell lines of widely differing radiosensitivity [human neuroblastomas: N2alpha, SHSY5Y, SK-N-SH, KELLY and SK-N-BE(2c); murine neuroblastomas: OP-6 and OP-27; human glioblastomas: G120, G60, G28, G112, G44 and G62] and compared their radiation response using the micronucleus and standard clonogenic assays. It was found that micronucleus frequency was much higher in some of the radioresistant cell lines (N2alpha, G28, G120 and G44; SF2 >/= 0.60). Evaluation of 100 patients with prostatic pathologies (70 PCa; 30 BPH) shows the uPA/PAI-1 ratios in PCa patients to be significantly higher than in BPH patients. This is fully consistent with a previous study on 62 patients (16 were PCa; 46 BPH) where the ratios were 0.055 and 0.031 for PCa and BPH patients, respectively (P = 0.0028). In older PCa patients, uPA/PAI-1 ratios tend to be higher.

Using a contingent of 62 patients of which 46 were BPH and 16 were PCa, we report definitive concentrations of uPA and PAI-1 in tumour tissue extracts and show that the uPA/PAI-1 ratio emerges as a candidate marker to distinguish between BPH and PCa.

Irradiation of human melanoma (MeWo, Be11) and squamous cell carcinoma (4451, 4197) cells induces cell cycle blocks from which the cells recover to re-enter mitosis after 40-60 h. In the TP53 mutant cell lines, MeWo and 4451, irradiation induces a G(2)-phase block, where the fraction of cells in G(2) phase reaches a maximum after 18-20 h. In the TP53 wild-type cell lines, 4197 and Be11, a G(1)- and G(2)-phase block is reached 12 and 16 h postirradiation, respectively. Addition of pentoxifylline after irradiation at the time when the number of cells in G(2) phase has reached a maximum shortens the normal recovery from G(2)-phase block to approximately 7 h. Addition of daunorubicin, melphalan and cisplatin under these conditions markedly enhanced drug toxicity. In the TP53-mutated cell lines MeWo and 4451, the survival ratio at 7 Gy measured by colony formation was 2.3-2.8, 8.6-85 and 52-74 for daunorubicin, melphalan and cisplatin, respectively. In the TP53 wild-type cell lines, the corresponding survival ratios were found to be 1.3-1.4, 2.3-3.0 and 1.2-2.6, respectively. The survival ratios are for clonogenic survival after 7 Gy and 2 mM pentoxifylline and measure the influence of drug doses that ensure 95% survival in nonirradiated controls. The results indicate that the G(2)-phase block is a crucial event in the damage response that can be manipulated to achieve a significant enhancement of drug toxicity. These effects are particularly pronounced in TP53 mutant cells and are observed at drug doses well below the clinical range.

The relationship between radiosensitivity and DNA repair was investigated in six human prostate cell lines, 1542-NPTX, BPH-1, 1542-CP(3)TX, 1532-CP(2)TX, 1535-CP(1)TX and LNCaP. Except for LNCaP, these cell lines are new and were derived from primary prostate tumours and normal non-tumourigenic prostate tissue. Cell survival was assessed by clonogenic assay. DNA damage was determined in non-synchronised cells by constant-field gel electrophoresis, and expressed as the fraction of DNA released. For initial damage, cells were embedded in agarose and irradiated on ice with 0-100 Gy (60)Co gamma-irradiation. Residual DNA damage was measured after 2 h and 20 h of repair. Radiosensitivity, given as the mean inactivation dose, was found to vary between 1.62 and 2.77 Gy. We found that radiosensitivity significantly correlates with the 2 h DNA repair component, giving a correlation coefficient of 0.92 ( P=0.009). In the cell lines examined here the 2 h repair component emerges as an indicator of radiosensitivity.
