Abstract:The Acanthaceae family is an important source of therapeutic drugs, and the ethnopharmacological knowledge of this family requires urgent documentation as several of its species are near extinction. Justicia is the largest genus of Acanthaceae, with approximately 600 species. The present work provides a review addressing the chemistry and pharmacology of the genus Justicia. In addition, the biological activities of compounds isolated from the genus are also covered. The chemical and pharmacological information in the present work may inspire new biomedical applications for the species of Justicia, considering atom economy, the synthesis of environmentally benign products without producing toxic by-products, the use of renewable sources of raw materials, and the search for processes with maximal efficiency of energy.

Species from genus Aspidosperma (Apocynaceae) are popularly employed to treat various diseases. This genus is characterized by the occurrence of indole alkaloids. Taking into account the various biological activities attributed to these alkaloids, the description of the chemical diversity in genus Aspidosperma is important. A review of simple carbolinic alkaloids isolated from species of various genera was published in 1979. In 1987, it was published another one dealing with the relationships between the chemical structures of the indole alkaloids and the evolution of Aspidosperma species. This work updates the information about the indole alkaloids isolated from Aspidosperma species.
The genus Aristolochia, an important source of physiologically active compounds that belong to different chemical classes, is the subject of research in numerous pharmacological and chemical studies. This genus contains a large number of terpenoid compounds, particularly diterpenes. This work presents a compilation of the 13C-NMR data of 57 diterpenoids described between 1981 and 2007 which were isolated from Aristolochia species. The compounds are arranged skeletonwise in each section, according to their structures, i.e., clerodane, labdane, and kaurane derivatives. A brief discussion on the 13C chemical shifts of these diterpenes is also included.
(+)-Bergenin (1) was isolated from Sacoglottis uchi, a species of vegetable found in the Amazon region and popularly used for the treatment of several hepatic problems. The structure of 1 was fully characterized using IR, GC-MS and NMR (1D and 2D) analyses. This phytoconstituent has been used as an oriental folk medicine for the treatment of many diseases and shows antihepatotoxic properties. Tests with beta-carotene, DPPH and a heterogeneous Fenton system were carried out, confirming the antioxidant activity of 1. Theoretical calculations were performed to investigate the formation of the radical derivatives of 1 using *H, *OH, *CH(3), and *CCl(3) as initiator radicals. DFT thermodynamic calculations showed that the methoxyl group (O-6-CH(3)) is the most favorable site for radical attack. Frontier molecular orbital analysis showed that nucleophilic radical attack is favored on the aromatic ring of 1 where the LUMO is localized, with antibonding character with respect to the O-6-CH(3) bond. The possibilities of attack at other sites on 1 were investigated in detail in order to understand the regiospecificity of this reaction.

Most of the snakebite incidents in the Amazon region involve Bothrops atrox, whose venom presents the most potent edematogenic and necrotic activities in the genus. This work describes the studies of isolation of the chemical constituents and antiedematogenic activity of the species Peltodon radicans (Lamiaceae), which is used in the treatment of snakebites and scorpion stings in the region. The extracts presented aliphatic hydrocarbons, 3β-OH,β-amirin (1), 3β-OH,α-amirin (2), β-sitosterol (3), stigmasterol (4), ursolic acid (5), 2α,3β,19α-trihydroxy-urs-12-en-28-oic acid (tormentic acid, 6), methyl 3β-hydroxy,28-methyl-ursolate (7), sitosterol-3-O-β-D-glucopyranoside (8), and stigmasterol-3-O-β-D-glucopyranoside (9). The flower extracts presented the higher antiedematogenic activity. This is the first report on the study of the flowers, stem, and roots of this plant. INTRODUÇÃOSegundo a Organização Mundial da Saúde, anualmente cerca de 2,5 milhões de pessoas sofrem acidentes ofídicos no mundo, com 125 mil mortes.1 Esses acidentes representam um enorme problema de saúde pública, especialmente em países tropicais.2-4 Na África, são registrados cerca de 500 mil acidentados por ano, com 40% dos casos hospitalizados e 20.000 óbitos.5 Na Ásia há entre 25 e 35 mil óbitos por ano.6 O Ministério da Saúde do Brasil registra entre 19 e 22 mil acidentes ofídicos por ano, com aproximadamente 0,45% de letalidade.7 Quando informado o gênero da serpente, Bothrops foi responsável por 90,5% dos acidentes ofídicos, envolvendo 0,31% de óbito.Bothrops atrox (jararaca) é encontrada vastamente na região Norte do Brasil, 8,9 habitando ambientes úmidos de zonas rurais ou periféricas das grandes cidades, onde haja facilidade para proliferação de roedores.10-13 Na Amazônia essa espécie ocorre em altas densidades, principalmente em florestas, apresentando hábitos noturnos. Os jovens ingerem presas exotérmicas e os adultos, presas endotérmicas, ocorrendo portanto variação ontogenética para a peçonha dessa espécie.14-16 Os venenos de B. atrox são constituídos principalmente por proteínas com atividade enzimática, apresentando variações tanto inter quanto intraespecíficas. Despite the fact that boranes are frequently used in amide reductions, the reaction mechanisms of the involved are note well known. This work presents the results of a bibliographic search on probable amide reduction mechanisms and an analysis of the existing literature. Steric and electronic effects were considered in light of reactivity since it could be concluded that the formation of intermediates and products depends mainly on the substitution patterns of both the boron and nitrogen atoms. Otherwise, results described in the literature for the reactions of boranes, sodium borohydride, lithium aluminum hydride, alkylboranes or haloboranes with others functional groups such as carboxylic acids, esters, ketones and alkenes were analysed with the aim to obtain something about the N-substituted amide reactions employing boranes.Keywords: amides; reactivity of boranes; reduction. INTRODUÇÃOAmidas estão envolvidas em vários processos químicos e bioló-gicos e têm sido empregadas em diversas classes de medicamentos como hipnóticos (zolpidem, zopiclone e acecarbromal), anticonvulsantes (carbamazepina, felbamato e progabida), tranqüilizan-tes (metilprilona e glutetimida), relaxantes musculares (alildietilacetamida, valnoctamida e oxanamida) e antivirais (AZT e ribavirin) 1-3 . Além disto, as amidas são empregadas também na síntese de agentes radiofarmacêuticos em medicina nuclear 4 .De um modo geral, as propriedades químicas e biológicas das amidas estão relacionadas com a ordem da ligação carbono-nitrogê-nio, a qual expressa a deslocalização eletrônica entre os heteroátomos, sendo influenciada por fatores estéricos e eletrônicos dos grupos substituintes presentes tanto no nitrogênio quanto no carbono carbonílico. Devido à deslocalização eletrônica, essa ligação (denominada também de ligação peptídica) apresenta um alto grau de rigidez, com barreiras de energia rotacional variando entre 50 e 100 kJ/mol 5,6 . A rigidez da ligação carbono-nitrogênio proporciona uma planaridade ao grupo amido em peptídeos e proteínas, a qual, somada aos efeitos de ligações de hidrogênio intramoleculares, determina formas estruturais helicoidais (Figura 1).Nos processos químicos, as reduções de amidas têm sido muito empregadas no desenvolvimento de metodologias para formação de aminas, substâncias também importantes quimicamente 8 . Os procedimentos utilizados para reduções de amidas podem ser agrupados em quatro tipos: reações eletroquímicas; reações com metais; reduções nucleofílicas e reduções eletrofílicas.As reações eletroquímicas e aquelas envolvendo metais são dependentes das condições de reação 9,10 , podendo levar à formação competitiva entre aminas, aldeídos ou álcoois [11][12][13][14][15][16][17] . As reduções nucleofílicas, empregando principalmente boro-hidreto de sódio (NaBH 4 ), hidreto de lítio e alumínio (LiAlH 4 ) e seus derivados 18-28 , proporcionam, além de aminas, a formação de outros produtos de reação. A reação entre metilamina e acetaldeído em pentano a 230 K leva à formação das aldiminas Ee Z-, sendo o isômero Z-o produto principal e gerado sob controle cinético. O isômero E-é mais estável do que o Z-por 3.3 kcal/mol (∆G*). Os cálculos ab initio mostraram que a etapa bimolecular de adição ocorre por um processo concertado, com formação da ligação C-N e transferência de próton através de um estado de transição de quatro centros. O intermediário zwiteriônico, frequentemente invocado neste tipo de reação, não existe como um mínimo na superfície de energia potencial. Entretanto, a energia livre de ativação para o mecanismo concertado é muito alta, e não é capaz de explicar a cinética deste processo. O mesmo acorre com a etapa de eliminação de água para formar a imina, ou seja, possui uma barreira de ativação muito alta. Estes resultados implicam que tanto a etapa de adição, como a de eliminação, devem ocorrer por um mecanismo de catálise.O isômero Z-pode se interconverter para a forma E-através de uma inversão sobre o nitrogênio sp 2 . A 298.15 K, a energia livre de ativação para este processo foi calculada como sendo 27.0 kcal/mol, o que leva a um tempo de vida de quatro meses para este isômero.The reaction of methylamine with acetaldehyde in pentane at 230 K leads to formation of both E-and Z-aldimines, the Z-isomer being the main product and generated upon kinetic control. The E-form is more stable than the Z-form by 3.3 kcal/mol (∆G*). Ab initio calculations have indicated that a tetrahedral zwitterion intermediate is not formed, and the bimolecular process, forming the aminoalcohol intermediate, occurs through a four center transition state, with concerted formation of C-N bond and proton transfer. However, the predicted ∆G* ≠ is very high, indicating that the true mechanism for this step is not bimolecular. A catalyzed pathway must be actuating. The following step, unimolecular elimination of water, also presents a very high activation free energy. Similarly, a catalyzed mechanism is needed. The isomerization from the Z-form to the E-form through inversion of the nitrogen atom has a predicted ∆G* ≠ of 27.0 kcal/mol at 298.15 K, resulting in a lifetime of four months for the Z-isomer.
