Various studies have shown that novel cytoprotective gasotransmitter hydrogen sulfide (H2S) has cytoprotective and preventive effects in the gastrointestinal system (Wallace J., 2010‐2012). However, the mechanism of H2S on oesophageal barrier remains unclear. Therefore, the aim of present study was to examine the protective role of endogenous H2S in oeosphageal mucosal (OM) lesions related to stress‐associated injury and postprandial hyperglycemia (PHG) which we recently recognized as a serious trigger in the oesophageal injury genesis (Hrytcevych N., 2011‐2013). Aim: To determine whether or not H2S attenuates oesophageal mucosal cytoprotection in rats with nonerosive oeosphagitis and long term PHG, and explored the underlying mechanisms may be involved. Methods: Rats without and with 3,5 h water immersion and restraint stress (WIRS) and 28 days of PHG received i.p. of vehicle, cystathionine gama‐lyase (CSE) inhibitor, DL‐propargylglycine (PAG, 25 mg/kg i.p.), CBS inhibitor, beta‐cyano‐alanine (BCA, 50 mg/kg i.p.), or H2S donor NaHS (100 mlmol/kg) prior to WIRS. Scoring lesion index (LI), vascular index (VI) by OM histomorphology, IL‐10, IL‐17 by ELISA. Results: WIRS & PHG induced non‐erosive injury which characterized by rise LI and VI vs control. PAG or BCA pretreatment enhanced EM lesions escalating LI and VI in twice, drop IL‐10 and rise IL‐17. NaHS mediated decrease of OM lesions via decreased LI, VI and increase IL‐10 and decrease IL‐17 scores vs control. Conclusion: These results indicate that endogenous H2S plays oesophagoprotective role by inhibiting inflammation, oedeme and vasodilator effect, highlighting the therapeutic potential for H2S donor in postprandial hyperglycemic stress‐associated conditions.
Metaflammation (metabolic inflammation) is low‐grade chronic inflammatory status linking to nutrient excess. Visceral adipocytes in mesenterium (AM) related to high fructose diet (HFD) are sources for metaflammation process, seriously threatening human health. Novel pathogenesis of AM is still unclear and there are no specific effective drugs for treatment of AM. Recently hydrogen sulfide (H2S) demonstrated potent vasodilatation actions which is promised for AM that have reduced vascularization Therefore, the aim of this study was designed to investigate the effects and underlying mechanisms of H2S on the prevention and treatment of experimental rats with AM during HFD. Methods The study was done on rats with standard diet (SD, control group) and 28 days HFD, and acute water immersion restraint stress (WRS) was induced for evaluation AM adaptive response and testing therapeutic potential of exogenous H2S donor NaHS (100 μmol/kg/day) and the hybrid H2S‐Aspirin (ATB 340, H2S‐ASA, 17,5 mg/kg/day) vs conventional aspirin (ASA, 10 mg/kg/day) on AM were evaluated by damage index by electronic microscopic study, and glucose level, activity of Cystathionine gamma‐lyase (CSE), Cystathionine beta‐synthase (CBS), Thiosulfate‐dithiol sulfurtransferase (TST) and Carbonyl group (CG) level in blood were examined biochemically (Institutional Ethical Committee Approval: 23/04/2018 № 4). Results In HSD group with NaHS treatment protected the endothelial and sub‐endothelial structures in mesenterium destructions were absent, the nucleus of endothelial cells it was observed an uneven condensation of chromatin. Effect of hybrid H2S‐aspirin characterized as a protective on AM against HFD and WRS‐induced injury, exhibited a downward trend in the inflammatory infiltration, lowering the level CG, which may indicate a reduction in oxidative stress under the influence of hybrid H2S‐aspirin. ATB‐340 showed an increase CSE, CBS and TST activities vs ASA pretreatment (p<0.05). The use of aspirin and NaHS has shown an increase on 67% CSE and CBS activities in HSD rats vs saline group. Conclusion These findings suggested that exogenous H2S prevented and ameliorated AM metflammation via reduction of oxidative damage, and provided a theoretical basis for effective tool for prevention of HFD‐related AM against long‐term postprandial hyperglycemia. Beneficial effects of L‐tryptophan (L‐Try) have already been described by many activities in kynurenine and serotonin synthesis, redox reactions in all living cells which play key role for cancer invasion and proliferation. Recently we revealed that postprandial hyperglycemia (PPH) which characterized by hyperglycemic spikes leads to nitro‐oxidative stress and may predispose to foregut disorders. Nevertheless, the effect of L‐Try on PPH is unknown. In this study, we elaluated effect of L‐Try on NO/NOS expression in OM during PPH and abnormal PG/COX activity.Rats were used with without/with L‐Try treatment (400 mg/kg) and indomethacin (Indo, 5 mg/kg) during PPH (Kozar, 2009); oesophagus (OM) and lower oesophageal sphincter (LOS) lesions were investigated by histology score index (HIS); esophageal NO2, cNOS and iNOS via bioanalysis.PPH caused destructive lesions in the OM and LOS, accompanied by the up‐regulation of iNOS and down‐regulation of cNOS expressions, excessive NO2‐ while Indo significantly aggravated severity of these lesions; L‐Try prevented ulcerogenic response to PPH with potent up‐regulation of cNOS but did not affect synthesis NO2‐.These results suggest that L‐Try prevents L‐Try‐induced OM and LOS damage but oesophagoprotective‐promoting effect may be associated with the stimulation of endothelial metabolism, but not through an amelioration of PG/COX activity.
Existing treatment of erosive and nonerosive esophagitis (NEE) which incidence has risen dramatically in recent years are mostly based on potent acid suppression. Modern data revealed that the prolonged acid suppression is associated with dismotility in foregut, extraesophageal disorders, abnormal microbiota, and induction selective apoptosis for cancer. We hypothesized that endothelial metabolism is key in multifactorial esophageal mucosa (EM) defense and studied 5‐HT/MT specific effects on EM endothelial metabolism during NEE associated to stress model (WRS) and 28 days of postprandial hyperglycemia (PPH) in rats was studied without/with 5‐HT (100 mg/kg) and MT (20 mg/kg) via VEGF, EGF, IL‐1β, TNFα synthesis by ELISA; EM injury via histological score index (HSI); functional reorganization of microangioarchitecture by mannose specific labeling.ResultsNNE constantly increased HSI in 150% in binary PPH and WRS; 5‐HT/MT decreased HIS 38% & 57%, respectively; potent MT and 5‐HT modulation vascular tone were through VEGF, TNFα synthesis better then 5‐HT. Changes in alfa‐DMan>;D‐Gal EM expression confirmed key role of endothelial metabolism in NEE and MT decreased expression of endothelial dysfunction. We concluded that MT has potent esophagoprotective effect then 5‐HT via decreasing endothelial permeability, thus may be alternative to the acid‐orientated approach.
Біологічні науки 194Український журнал медицини, біології та спорту -Том 3, № 4 (13)З використанням панелі з 6 лектинів різної вуг-леводної специфічності (LABA, RCA, WGA, CNFA, CCRA, GNA) досліджували вплив натрієвої солі глутамінової кислоти на гістотопографію вуглевод-них детермінант структурних компонентів слизової оболонки язика. Встановлено, що глутамат-індукована перебудова глікокон'югатів супроводжу-валась повною або частковою редукцією терміна-льних залишків αLFuc, βDGal, DGlcNАc, NeuNАc та GalNAcβ1-˃4GlcNAc, (LacdiNAc) у складі цитоплаз-ми смакових рецепторних клітин та підтримуваль-них епітеліоцитів смакових бруньок, у поєднанні з накопиченням рецепторів лектинів RCA, WGA та CNFA у поверхневому шарі епітелію слизової обо-лонки язика. Зміна вуглеводного профілю під дією натрієвої солі глутамінової кислоти ймовірно спри-чиняє зміну проникності плазмолеми смакових ре-цепторних клітин, що призводить до зміни смакової чутливості. Виявлена редукція рецепторів лектинів LABA, RCA, WGA та CNFA у клітинному складі смакової ямки за умов глутамат-індукованого ожи-ріння може бути пов'язана зі зниженням секреції глікопротеїнів підтримувальними епітеліоцитами.Ключові слова: ожиріння, язик, глікокон'югати, лектини.Зв'язок роботи з науковими програмами, планами, темами. За даними ВООЗ більше мільярда осіб на планеті мають надмірну масу тіла. На сьогодні проблема ожиріння набула статусу неінфекційної пандемії [2,3]. За даними епідеміологічних дослід-жень, що були проведені в Україні, більш ніж поло-вина дорослого населення має надмірну масу тіла, що безумовно пов'язано з низьким рівнем фізичної активності, особливостями харчової поведінки, зростанням в раціоні висококалорійних продуктів і легкозасвоюваних вуглеводів, а також широким і безконтрольним використанням в харчовій промис-ловості та в мережі підприємств громадського хар-чування харчових добавок, перш за все, глутамату натрію [5,7,10]. Стимулюючи закінчення смакових нервів, глутамінова кислота та її солі підсилюють смакові відчуття, в результаті чого з'являється від-чуття задоволення. Відомо, що глутамат натрію у вигляді харчової добавки викликає порушення об-міну речовин та може бути однією з причин розвит-ку ожиріння i захворювання травного тракту [6]. Незважаючи на значне число робіт, присвячених даній проблемі, єдиної думки щодо безпечної дози поширеної харчової добавки -глутамату натрію немає [8,9,16].
