e13506 Background: Gliomas are the most common type of primary brain tumors, with a high level of recurrence and mortality. The purpose of the study was to determine the expression profile of micro-RNA targeting components of the Hedgehog, Notch, Wnt, EGFR, TGFβ, HIF1α signaling pathways according to MirTarBase, miRDB, TargetScan. Methods: The study included 30 patients (15 women and 15 men) aged 27 to 76 years with histologically verified glial brain tumors. The RT2-qPCR method in operational biopsy specimens determined the relative expression of 12 micro-RNAs: hsa-miR-17-3p, hsa-miR-20a-3p, hsa-miR-326, hsa-miR-330-3p, hsa-miR- 107, hsa-miR-143-3p, hsa-let-7a-5p, hsa-miR-146a-5p, hsa-miR-29a-3p, hsa-miR-92a-1-5p, hsa-miR-26b-3p, hsa-miR-96-5p. Hsa-miR-7-5p and hsa-miR-126-3p were used as reference micro-RNA. Results: A statistically significant increase in the expression of hsa-miR-143-3p, hsa-miR-146a-5p and hsa-miR-92a-1-5p by 2, 2.2 and 2.9 times, respectively, was found, as well as a decrease in the expression of hsa-miR-330- 3p by 4.1 times in tumor tissue of the brain relative to normal tissue (p < 0.05). Reduced expression of micro-RNA hsa-miR-330-3p may increase the activity of the VEGFA gene, which leads to intensive vascularization of the tumor. In addition, hsa-miR-330-3p is a validated negative regulator of the E2F1 gene known as a regulator of the cell cycle and tumor suppressor proteins. The TRAF6 gene is the direct validated target of hsa-miR-146a-5p, and its overexpression is associated with the patient's chemoresistance to temozolomide. Increased hsa-miR-143-3p micro-RNA expression can reduce the activity of the EGLN1 gene that also mediates the adaptation of tumor cells to hypoxic conditions. Hsa-miR-92a-1-5p micro-RNA is a negative regulator of PTEN gene expression. Conclusions: A significant decrease in hsa-miR-330-3p expression and an increase in hsa-miR-146a-5p, hsa-miR-143-3p, and hsa-miR-92a-1-5p, which was found in gliomas, can potentially be used to develop prognostic markers and therapeutic targets for targeted therapy.

Низкодифференцированные глиальные опухоли являются наиболее часто встречаемыми злокачественными опухолями головного мозга и составляют около 65% от злокачественных опухолей ЦНС. Диагностика и лечение данной патологии остается острой проблемой в нейроонкологии. Цель исследования: изучение изменений цитокинового состава в ткани различных гистологических и молекулярно-генетических подтипов глиальных опухолей. В исследовании приняли участие 49 пациентов. В основную группу вошли 34 пациента с впервые выявленной супратенториальной глиальной опухолью. В контрольную группу вошли 15 пациентов с впервые выявленной супратенториальной менингиомой. Во время операции у пациентов основной группы проводился забор опухолевой ткани, а у пациентов контрольной группы -прилежащего к менингиоме неизмененного мозгового вещества в функционально незначимой зоне. В основной группе диффузная астроцитома верифицирована у 4 пациентов, у 4 -анапластическая астроцитома и у 26 -глиобластома. Среди пациентов с HGG мутация гена IDH1 обнаружена у 3 пациентов. У 30 пациентов с HGG определен уровень метилирования промотора MGMT. Изучение локального цитокинового состава неизмененного мозгового вещества и опухолевой ткани осуществлялось методом ИФА. При сравнении цитокинового состава неизмененного мозгового вещества с тканью HGG имеются статистически достоверные отличия (p<0,05) в содержании ИЛ-2, ИЛ-4, ИЛ-6, ИЛ-8, ИЛ-10, ФНО-α. У больных с HGG и наличием мутации в генах IDH1 тканевой уровень ИЛ-4 был в 33 раза выше, чем у больных без мутации, а уровни ИЛ-6 и ИЛ-8, напротив, были ниже в 33 и 249 раз соответственно. При уровне метилирования промотора MGMT >10% отмечено статистически достоверное повышение локального уровня ИЛ-1β. Изучение локального цитокинового состава HGG наряду с гистологическими и молекулярно-генетическими аберрациями может служить предиктором течения заболевания и чувствительности к химиотерапии препаратами алкилирующей группы.
Purpose of the study. This research was devoted to study of mRNA and miRNA expression patterns in glioglastomas using The Cancer Genome Atlas (TCGA) data, to search for genetic determinants that determine the prognosis of patient survival and to create of interaction networks for glioblastomas.Materials and methods. Based on the data of the open TCGA database groups of glioblastomas and conventionally normal brain tissue samples were formed. Survival gene and miRNA expression data were extracted for each sample. After the data stratification by groups the differential expression analysis and search the genes affecting patient survival was carried out. The enrichment analysis by functional affiliation and an interactome analysis were performed.Results. A total of 156 glioblastoma samples with mRNA sequencing data, 571 samples with microarray microRNA analysis data, and 15 control samples were analyzed. Networks of mRNA-miRNA interactions were built and expression profiles of genes and miRNAs characteristic of glioblastomas were developed. We have determined the genes which aberrant level is associated with survival and shown the pairwise DEG and DE of microRNA correlations.Conclusion. The microRNA-mRNA regulatory pairs identified for glioblastomas can stimulate the development of new therapeutic approaches based on subtype-specific regulatory mechanisms of oncogenesis.
