La reacción en cadena de la polimerasa con transcripción inversa (RT- PCR) es uno de los métodos de laboratorio más precisos y ampliamente utilizados para diagnosticar la enfermedad respiratoria de coronavirus 2019 el objetivo de investigar fue evaluar la reacción en cadena de la polimerasa: variabilidad en técnicas y método diagnóstico para COVID-19”, mediante diseño cualitativo, descriptivo de revisión sistemática a través de la recopilación de fuentes de datos científicos como; Scielo, Pubmed, Scopus, Latindex,Web of science, , Science direct, Redalyc, se utilizaron artículos científicos y se utilizaron los términos “PCR”, “COVID-19” y “SARS-CoV-2”. Se obtuvieron resultados de la variabilidad de técnicas en reacción de la cadena de la polimerasa donde se encontró tipos como la RT-PCR, PDR-Ag,RT-qPCR, RT LAMP, que son pruebas diagnósticas que tienen una confiabilidad entre 90 y 98.9% en la detección del virus SARS-COV-2, uno de los métodos más utilizados es la RT.PCR por su alto nivel de eficacia y confiabilidad en la detección de gen E del virus ya que no generan falsos positivos en el diagnóstico de la enfermedad y tienen una sensibilidad del 95% y especificidad del 100% catalogándose como una de las más seguras en la detección del diagnóstico clínico para COVID 19, uno de los factores principales encontrados que dificulta la detección del virus es la recolecta de la muestra, temperatura y ambiente antes de su análisis para detectar el virus de SARS-COV-2. Se concluye que La RT-PCR es una de las técnicas principalmente utilizadas por su eficacia y confiabilidad en la detección del gen E del virus SARS-COV-2 detectándolo en pacientes asintomáticos y no generar resultados falsos positivos en el diagnóstico de la enfermedad COVID 19.
