Sólidos magnéticos tipo Fe3O4, Fe3O4-NH2, Fe3O4@SiO2 y Fe3O4@SiO2-NH se usaron como soportes en la inmovilización de la ureasa, estos sólidos fue caracterizados por análisis de fisisorción de N2 a 77K, curvas de magnetización y FTIR. La actividad enzimática se evaluó por descomposición de la urea. Los sólidos presentan características magnéticas que les permite fácilmente ser separados. La enzima inmovilizada mejoró notablemente su actividad enzimática comparada con la enzima libre. Para los sólidos Fe3O4-NH2, y Fe3O4@SiO2-NH2 se observó una mayor cantidad de enzima acoplada.
Se han desarrollado procedimientos de inmovilización con invertasa utilizando diferentes soportes. Sin embargo, las desventajas como el uso de partículas pequeñas para inmovilizaciones de invertasa en reactores de lecho compacto se están resolviendo utilizando partículas magnéticas. En este estudio, los compuestos que contienen Fe3O4 se prepararon mediante la incorporación de una capa de polisiloxano necesaria para la adsorción física de la invertasa. Además, la magnetita funcionalizada se activó con glutaraldehído y polietilenimina (PEI) con el objetivo de realizar una inmovilización covalente. Se analizó el efecto de diferentes condiciones como la relación enzima: soporte, pH y temperatura en la conservación de la invertasa. Los resultados demostraron que la relación enzima: soporte óptima es mayor para la unión covalente que para la adsorción física. El pH ideal para la enzima inmovilizada es 5,0 y la actividad enzimática se mantiene hasta 70 ° C. Los valores de km son similares en ambos métodos de inmovilización. El análisis del efecto del pH y la termoestabilidad mostró que la actividad catalítica de la invertasa no se ve afectada en comparación con la enzima libre. La inmovilización covalente muestra una mayor eficacia en el proceso de inmovilización (Fε), menos inhibición y el doble de estabilidad. Las enzimas inmovilizadas por métodos físicos y covalentes se pueden reutilizar hasta por cuatro ciclos y se pueden eliminar del medio de reacción aplicando un campo magnético externo.
Para mejorar la actividad enzimática de las celulasas es necesario disminuir las barreras estructurales de la celulosa con distintos pretratamientos, en este trabajo se realizó el estudio del pretratamiento hidrotérmico de celulosa comercial a distintos factores de severidad, con el fin de mejorar la conversión a azucares reductores y disminuir la cristalinidad. Además, se estudió la degradación enzimática de la celulosa pretratada hidrotermicamente con celulasas comerciales previamente inmovilizadas en SiO2-NH2. Los resultados indican una mayor degradación de la celulosa tratada frente a la celulosa no tratada, a pesar que la actividad enzimática disminuye cuando las enzimas se inmovilizan. La acidez de la zeolita beta fue modificada por impregnación con cerio, lantano, grupos sulfónicos y óxidos de hierro, este último se utilizó para generar propiedades magnéticas en el material que permitan la recuperación del catalizador y reciclaje en reacción en fase líquida. Los sólidos obtenidos fueron caracterizados por DRX, espectroscopia infrarroja FT-IR, la acidez fue obtenida por titulación y la actividad catalítica fue evaluada en la hidrólisis de celulosa cristalina como porcentaje de azúcares reductores totales (ART). La cristalinidad de los sólidos impregnados con Óxidos de Ce y La no se ve afectada, caso contrario ocurre cuando se impregna con óxidos de hierro, además la acidez disminuye en ambos casos. La incorporación de grupos sulfónicos tanto a la zeolita beta precursora como a la magnetizada aumenta la acidez y mostró rendimientos del 59% y 38% respectivamente alto rendimiento de azúcares reductores totales en la reacción.
