Earliness is one of the most important adaptation traits in plant breeding. Our purpose was to identify the genome regions of bread wheat involved in the control of earliness and its three components: photoperiod sensitivity (PS), vernalization requirement (VR) and intrinsic earliness (IE). A QTL meta-analysis was carried out to examine the replicability of QTL across 13 independent studies and to propose meta-QTL (MQTL). Initial QTL were projected on a recent consensus map (2004). Quality criteria were proposed to assess the reliability of this projection. These criteria were based on the distances between markers in the QTL regions. Chromosomes of groups 2 and 5 had a greater incidence on earliness control as they carry the known, major genes Ppd and Vrn. Other chromosome regions played an intermediate role in earliness control: 4A [heading date (HD) Meta-QTL], 4B (HD MQTL), 2B (VR MQTL) and 5B (IE MQTL). Markers at this four MQTL should prove helpful in marker-assisted selection, to better control earliness.

Mineral nitrogen fertilization has improved crop yield over the last century but has also caused air and water pollution. Reduction of nitrogen inputs and maintaining high yields are therefore essential to ensure a more sustainable agriculture. Improving the nitrogen use efficiency (NUE) of crops is therefore needed. Rapeseed, Brassica napus, depends on nitrogen fertilization due to its low NUE, with the ratio of plant nitrogen content to nitrogen supplied often not exceeding 60 %. Here, we review the major phenotypic traits associated with NUE in B. napus, with special emphasis on winter oilseed rape. We discuss the genetic diversity available and potential breeding strategies. The major points are the following: (1) rapeseed seed yield elaboration is complex, with overlapping phases of nitrogen uptake and remobilization during the crop cycle; (2) traits related to nitrogen uptake, such as root length and the amount of nitrogen absorbed after flowering, and traits related to nitrogen remobilization, such as the "staygreen" phenotype, have been identified as possible levers to improve NUE in rapeseed; (3) a substantial body of studies investigating the genetic control of NUE traits have already published and potential candidate genes identified; and (4) rapeseed genetic diversity may be enriched by exploiting interpopulation genetic variation and the closely related gene pools of Brassica rapa and Brassica oleracea.

Lower market prices and environmental concerns now orientate wheat (Triticum aestivum L.) breeding programs towards low input agricultural practices, and more particularly low nitrogen (N) input management. Such programs require knowledge of the genetic determination of plant reaction to N deficiency. Our aim was to characterize the genetic basis of N use efficiency and genotype x N interactions. The detection of QTL for grain yield, grain protein yield and their components was performed on a mapping population of 222 doubled haploid lines (DH), obtained from the cross between an N stress tolerant variety and an N stress sensitive variety. Experiments on the population were carried out in seven different environments, and in each case under high (N(+)) and low (N(-)) N supplies. In total, 233 QTL were detected for traits measured in each combination of environment and N supply, for "global" interaction variables (N(+)-N(-) and N(-)/N(+)), for sensitivity to N stress and for performance under N-limited conditions which were assessed using factorial regression parameters. The 233 QTL were detected on the whole genome and clustered into 82 genome regions. The dwarfing gene (Rht-B1), the photoperiod sensitivity gene (Ppd-D1) and the awns inhibitor gene (B1) coincided with regions that contained the highest numbers of QTL. Non-interactive QTL were detected on linkage groups 3D, 4B, 5A1 and 7B2. Interactive QTL were revealed by interaction or factorial regression variables (2D2, 3D, 5A1, 5D, 6A, 6B, 7B2) or by both variables (1B, 2A1, 2A2, 2D1, 4B, 5A2, 5B). The usefulness of QTL meta-analysis and factorial regression to study QTL x N interactions and the impact of Rht-B1, Ppd-D1 and B1, are discussed.

Breeding new varieties adapted to low-input agricultural practices is of particular interest in light of current economical and environmental concerns. Improving nitrogen (N) uptake and N utilization efficiency (NUE) are two ways of producing varieties tolerant to low N input. To offer new possibilities to breeders, it is necessary to acquire more knowledge about these two processes. Knowing C and N metabolisms are linked and knowing N uptake is partly explained by root characteristics, we carried out a QTL analysis for traits associated with N uptake and NUE by using both a conceptual model of C/N plant functioning and a root architecture description. A total of 120 lines were selected according to their genotype among 241 doubled haploids derived from two varieties, one N stress tolerant and the other N stress sensitive. They were grown in hydroponic rhizotrons under N-limited nutritional conditions. Initial conditions varied among genotypes; therefore, total root length on day 1 was used to correct traits. Heritabilities ranged from 13 to 84%. Thirty-two QTL were located: 6 associated with root architecture (on chromosomes 4B, 5A, 5D and 7B), 6 associated with model efficiencies (1B, 2B, 6A, 6B, 7A, 7B and 7D) and 20 associated with state variables (1A, 1B, 2B, 4B, 5A, 5B and 6B). The effects of the dwarfing gene Rht-B1 on root traits are discussed, as well as the features of a conceptual plant functioning model, as a useful tool to assess pertinent traits for QTL detection. It is suggested that further studies that couple QTL with a functioning model and a root architecture description could serve in the search for ideotypes.

Seeds are involved in the transmission of microorganisms from one plant generation to the next and consequently act as reservoirs for the plant microbiota. The driving processes influencing seed microbiota assemblage have not been yet deciphered because of confounding factors related to environmental location, agricultural practices, and host genotype selection. Nine genotypes were chosen among a large panel of genetically diverse Brassica napus accessions. The taxonomic structure of the seed microbiota was monitored by amplification and subsequent high-throughput sequencing of gyrB and internal transcribed spacer 1 markers for two successive years on seed lots collected from self-pollinated plants. Seed germination capacities were compared between all seed lots. Although harvesting year was the main driver of seed microbiota composition, the host genotype also significantly altered the structure of seed microbial assemblages. The core microbiota of B. napus included nine fungal taxa shared between all the genotypes and years, while no bacterial taxa were conserved across all genotypes and years. The harvesting year had the major effect on seed germination but with some differences between genotypes. The study demonstrated the relative contribution of host- and environmental-filtering on the assemblage of the seed microbiota. It suggested some influence of these assemblages on seed germination.

Despite its high nitrogen absorption capacity, oilseed rape (OSR) has a low apparent nitrogen use efficiency (NUE), which makes its production highly dependent on nitrogen fertilization. Improving NUE in OSR is therefore a main target in breeding. The objectives of the present work were to determine the genomic regions (QTLs) associated with yield and to assess their stability under contrasted nitrogen nutrition regimes. One mapping population, AM, was tested in a French location for three growing seasons (2011, 2012 and 2013), under two nitrogen conditions (optimal and low). Eight yield-related traits were scored and nitrogen-responsive traits were calculated. A total of 104 QTLs were detected of which 28 controlled flowering time and 76 were related to yield and yield components. Very few genotype 9 nitrogen interactions were detected and the QTLs were highly stable between the nitrogen conditions. In contrast, only a few QTLs were stable across the years of the trial, suggesting a strong QTL 9 year interaction. Finally, eleven critical genomic regions that were stable across nitrogen conditions and/or trial years were identified. One particular region located on the A5 linkage group appears to be a promising candidate for marker assisted selection programs. The different strategies for OSR breeding using the QTLs found in the present study are discussed.

Nitrogen levels can modulate the effectiveness of clubroot resistance in an isolate- and host-specific manner. While the same QTL were detected under high and low nitrogen, their effects were altered. Clubroot, caused by Plasmodiophora brassicae, is one of the most damaging diseases of oilseed rape and is known to be affected by nitrogen fertilization. However, the genetic factors involved in clubroot resistance have not been characterized under nitrogen-limiting conditions. This study aimed to assess the variability of clubroot resistance under different nitrogen levels and to characterize the impact of nitrogen supply on genetic resistance factors. Linkage analyses and a genome-wide association study were conducted to detect QTL for clubroot resistance and evaluate their sensitivity to nitrogen. The clubroot response of a set of 92 diverse oilseed rape accessions and 108 lines derived from a cross between 'Darmor-bzh' (resistant) and 'Yudal' (susceptible) was studied in the greenhouse under high- and low-nitrogen conditions, following inoculation with the P. brassicae isolates eH and K92-16. Resistance to each isolate was controlled by a major QTL and a few small-effects QTL. While the same QTL were detected under both high and low nitrogen, their effects were altered. Clubroot resistance to isolate eH, but not K92-16, was greater under a low-N supply versus a high-N supply. New sources of resistance were found among the oilseed rape accessions under both low and high-N conditions. The results are discussed relative to the literature and from a crop improvement perspective.

Clubroot, caused by Plasmodiophora brassicae, is a worldwide disease affecting Brassica. Until now, the detection of genetic factors (QTLs) implicated in clubroot resistance has been based on estimates of disease index. However, as the amount of resting spores released in soil after club disintegration influences clubroot epidemics and resistance‐breaking dynamics, its genetic control may deserve specific attention. In a previous report, it was shown that nitrogen fertilization modulated quantitative partial resistance toward clubroot symptom development in rapeseed. The present work aimed to identify genetic factors involved in the control of resting spore production and to assess their regulation by nitrogen supply. A flow cytometer method was adapted for rapidly estimating resting spore content in a large series of samples. Linkage analysis was conducted to detect QTLs implicated in resting spore production in a Brassica napus doubled haploid progeny from the cross Darmor‐bzh × Yudal. DH lines inoculated with the P. brassicae isolate eH were grown under low‐ and high‐nitrogen supply. Under low‐nitrogen conditions, resting spore production was reduced compared to high‐nitrogen conditions, regardless of genotypes. Genetic architecture controlling resting spore production and clubroot symptom development was similar. Under high‐nitrogen conditions, resting spore production was controlled by one major QTL (C09a) and a few small‐effect QTLs. By contrast, two major QTLs (C02 and C09a) controlled resting spore production under low‐nitrogen conditions. This work highlighted a large see‐saw effect between the relative contribution of the C09a QTL (high effect under high‐nitrogen conditions) and the C02 QTL (high effect under low‐nitrogen conditions), with possible implications in resistance breeding.
