Sleep and wakefulness are no longer to be considered as discrete states. During wakefulness brain regions can enter a sleep-like state (off-periods) in response to a prolonged period of activity (local use-dependent sleep). Similarly, during nonREM sleep the slow-wave activity, the hallmark of sleep plasticity, increases locally in brain regions previously involved in a learning task. Recent studies have demonstrated that behavioral performance may be impaired by off-periods in wake in task-related regions. However, the relation between off-periods in wake, related performance errors and learning is still untested in humans. Here, by employing high density electroencephalographic (hd-EEG) recordings, we investigated local use-dependent sleep in wake, asking participants to repeat continuously two intensive spatial navigation tasks. Critically, one task relied on previous map learning (Wayfinding) while the other did not (Control). Behaviorally awake participants, who were not sleep deprived, showed progressive increments of delta activity only during the learning-based spatial navigation task. As shown by source localization, delta activity was mainly localized in the left parietal and bilateral frontal cortices, all regions known to be engaged in spatial navigation tasks. Moreover, during the Wayfinding task, these increments of delta power were specifically associated with errors, whose probability of occurrence was significantly higher compared to the Control task. Unlike the Wayfinding task, during the Control task neither delta activity nor the number of errors increased progressively. Furthermore, during the Wayfinding task, both the number and the amplitude of individual delta waves, as indexes of neuronal silence in wake (off-periods), were significantly higher during errors than hits. Finally, a path analysis linked the use of the spatial navigation circuits undergone to learning plasticity to off periods in wake. In conclusion, local sleep regulation in wakefulness, associated with performance failures, could be functionally linked to learning-related cortical plasticity.

Variation of the magnitude of posterior alpha rhythm (8-12 Hz) has functional and behavioural effects in sensory processing and cognitive performances. Electrical brain activity, as revealed by electroencephalography (EEG), can be represented by a sequence of microstates of about 40-120 ms duration, in which distributed neural pools are synchronously active and generate stable spatial potential topographies on the scalp. Microstate dynamics may reflect transitions between global states characterized by selective inhibition of specific intra-cortical regions, mediated by alpha activity. We investigated the intra-subject and inter-subject relationship between microstate features and alpha band. High-density EEG signals were acquired in 29 healthy subjects during ten minutes of eyes closed rest. Individual EEG signal epochs were classified into four groups depending on the amount of occipital alpha power, and microstate metrics (duration, coverage and frequency of occurrence) were calculated and compared across groups. Correlations between alpha power and microstate metrics between individuals were also performed. To assess if microstate parameter variations are specific for the alpha band, the same analysis was also performed for theta and beta bands, as well as for global field power. We observed an increase in the metrics of microstate, previously associated to the visual system, with the level of intra-subject amplitude alpha oscillations, together with lower coverage of microstate associated with executive attention network and a higher frequency of microstate associated with task negative network. Other modulation effects of broad-band EEG power level on microstate metrics were observed. These effects are not specific for the alpha band, since they can equally be attributed to fluctuations in other frequency bands. We can interpret our results as a regulation mechanism mediated by posterior alpha level, dynamically interacting with other frequency bands, responsible for the switching between active areas.The dynamics of human brain signals uncover organized fluctuations both at rest and during task or sensory input. Time-of-day modulations affect both performance on a wide range of cognitive tasks and electrical activity of the brain, as recorded by electroencephalography (EEG). The aim of this work was to identify fluctuations of fractal properties of EEG time series due to circadian rhythms. In twenty-one healthy volunteers (all males, age between 20 and 30 years, chronotype: neutral type) high density EEG recordings at rest in open and closed eyes conditions were acquired in 4 times of the day (8.00 a.m., 11.30 a.m., 2.30 p.m., 7.00 p.m.). A vigilance task (Psychomotor Vigilance Test, PVT) was also performed. Detrended fluctuation Analysis (DFA) of envelope of alpha, beta and theta rhythms was performed, as well as Highuchi fractal dimension (HFD) of the whole band EEG. Our results evidenced circadian fluctuations of fractal features of EEG at rest in both eyes closed and eyes open conditions. Lower values of DFA exponent were found in the time T1 in closed eyes condition, likely effect of the sleep inertia. An alpha DFA exponent reduction was found also in central sensory-motor areas at time T3, the day time in which the sleepiness can be present. In eyes open condition, HFD lowered during the day. In eyes closed condition, an HFD increase was observed in central and frontal regions at time T2, the time in which alertness reaches its maximum and homeostatic sleep pressure is low. Complexity and the persistence of temporal correlations of brain rhythms change during daytime, parallel to changes in alertness and performance.

Aftereffects have been documented for a variety of perceptual categories spanning from body gender to facial emotion, thus becoming an important tool in the study of high-level vision and its neural bases. We examined whether the perceived valence of a complex scene is subject to aftereffects, by observing the participants’ evaluation of the valence of a test picture preceded by a different picture. For this study, we employed an adaptation paradigm with positive and negative images used as adapters, and positive, negative, and neutral images used as tests. Our results show that adaptation to complex emotional pictures induces assimilative aftereffects: participants judged neutral tests more positively following positive adapters and more negatively following negative adapters. This depended on the prolonged adaptation phase (10 s), as the results of a second experiment, in which adapters lasted for 500 ms, did not show aftereffects. In addition, the results show that assimilative aftereffects of negative and positive adapters also manifested themselves on non-neutral (negative and positive) targets, providing evidence that the global emotional content of complex pictures is suitable to induce assimilative aftereffects.

In post-COVID-19 syndrome (PCS), neurocognitive symptoms and fatigue are often associated with alterations in electroencephalographic (EEG) activity. The present study investigates the brain source activity at rest in PCS patients (PCS-pts) perceiving cognitive deficits and fatigue. A total of 18 PCS-pts and 18 healthy controls (HCs) were enrolled. A Montreal Cognitive Assessment (MoCA), Perceived Cognitive Difficulties Scale (PDCS) and Fatigue Severity Scale (FSS) were administered for assessing the symptoms’ severity. Brain activity at rest, both with open (OE) and closed eyes (CE), was recorded by high-density EEG (Hd-EEG) and localized by source estimation. Compared to HCs, PCS-pts exhibited worse performance in executive functions, language and memory, and reported higher levels of fatigue. At resting OE state, PCS-pts showed lower delta source activity over brain regions known to be associated with executive processes, and these changes were negatively associated with PDCS scores. Consistent with recent literature data, our findings could indicate a dysfunction in the neuronal networks involved in executive functions in PCS-pts complaining of fatigue and cognitive impairment.

Unihemispheric sleep is an aspect of the cerebral lateralization of certain species of birds. During sleep, domestic chicks (Gallus gallus) show brief periods of monocular-unihemispheric sleep (Mo-Un sleep): one eye is open and the connected hemisphere is awaken while the other eye remains shut and the connected hemisphere sleeps. The time spent in Mo-Un sleep was investigated following a brief monocular deprivation (MD) in chicks hatched from eggs incubated in darkness and reared in light (D-L), incubated in light and reared in light (L-L) and incubated in darkness and reared in darkness (D-D). The right eye occluded for 12 h in half of chicks and the left eye in the other half. At the end of MD, the Mo-Un sleep was recorded. The effect of MD (total time and time bias) prevailed in determining the pattern of Mo-Un sleep. Chicks showed more time sleeping with the eye/hemisphere that was in control of visual behaviour during MD and opened more time the eye and awake the hemisphere visually deprived. The Mo-Un pattern was not influenced by incubation, rearing, symmetry/asymmetry of visual pathways and imprinting thereby indicating that Mo-Un sleep pattern depends only on the kind of visual experience during wakefulness.
