Espécimes depositados nas coleções dos museus são uma comprovação da biodiversidade do nosso planeta, assim como, os dados associados a cada um desses espécimes. Visando a ampliar o conhecimento da ictiofauna no sul do Brasil, nosso objetivo foi apresentar uma lista das espécies de peixes marinhos e de água doce, assim como a sua distribuição no sul do Brasil dos espécimes registrados no Museu Regional do Alto Uruguai (MuRAU). O material ictiológico do MuRAU foi revisado, com os nomes científicos atualizados, sendo que exemplares não catalogados foram incorporados ao acervo e identificados nas categorias possíveis. Após a revisão dos aspectos taxonômicos, realizamos manutenção e acondicionamento adequado dos exemplares. A coleção ictiológica é composta por 29 ordens, 59 famílias, 119 gêneros e 174 espécies. Até o momento 75,12% dos exemplares registrados foram identificados ao nível de espécie, 8,9% ao nível de gênero, 2,74% ao nível de família, 3,63% identificados ao nível de ordem, e 9,57% dos registros ainda não foram identificados em nenhum dos níveis mencionados. Por meio deste levantamento, obtivemos resultados promissores sobre a representação das espécies presentes na coleção e na região.
