The recent outbreak of Zika virus (ZIKV) disease caused an enormous number of infections in Central and South America, and the unusual increase in the number of infants born with microcephaly associated with ZIKV infection aroused global concern. Here, we developed a reverse transcription loop-mediated isothermal amplification (RT-LAMP) assay using a portable device for the detection of ZIKV. The assay specifically detected ZIKV strains of both Asian and African genotypes without cross-reactivity with other arboviruses, including Dengue and Chikungunya viruses. The assay detected viral RNA at 14.5 TCID50/mL in virus-spiked serum or urine samples within 15 min, although it was slightly less sensitive than reference real time RT-PCR assay. We then evaluated the utility of this assay as a molecular diagnostic test using 90 plasma or serum samples and 99 urine samples collected from 120 suspected cases of arbovirus infection in the states of Paraíba and Pernambuco, Brazil in 2016. The results of this assay were consistent with those of the reference RT-PCR test. This portable RT-LAMP assay was highly specific for ZIKV, and enable rapid diagnosis of the virus infection. Our results provide new insights into ZIKV molecular diagnostics and may improve preparedness for future outbreaks.
Objective To assess the influence of bariatric surgery on transcription factor 7-like 2 ( TCF7L2 ) expression and its association with body mass index (BMI) and Type 2 diabetes mellitus (T2DM). Methods Prospective study performed between 2016 and 2018, where 26 obese patients undergoing bariatric surgery were divided into two subgroups: diabetics and non-diabetics. The RNAs were extracted from peripheral blood samples that were obtained from each patient in two different moments: before surgery and after 12 months of follow-up. The relative expression of TCF7L2 was determined according to the delta-Ct method. Results The linear regression model of BMI x delta-Ct showed a positive correlation (p = 0.037). In the subgroups, an inversely proportional relationship was found between delta-Ct and BMI in the diabetic group and a directly proportional relationship in the non-diabetic group (p>0.05 in both). In the postoperative period, the regression model was similar to the preoperative, except when analyzing the subgroups, where diabetic patients showed a directly proportional relationship (p>0.05). The relative expression of TCF7L2 showed an average of 1.16 ± 0.91, CI-95% 0.79–1.53. There was an increase in relative expression of 48% in the non-diabetic group (p = 0.021), and a decrease of 27% in the T2DM group (p>0.05) in the postoperative. There was a positive correlation between a greater decrease in BMI and increased relative expression (p = 0.027). Conclusion Our results showed that generally, the TCF7L2 expression increase with a decrease in BMI, however, for patients with T2DM, it exhibits an inverse pattern, which is normalized one year after bariatric surgery.
As Comissões de Infecção Hospitalar são reconhecido como espaços importantes para a diminuição e controle das doenças considerada nosocomial, que trazem maior índice de mortalidade e aumento no tempo de internação. Nesses cenários que os profissionais enfermeiros aparecem como gestores em potencial nos processos de educação permanente, e gestão de pessoas. O presente estudo teve como objetivo identificar e mapear acerca da gestão e competências dos profissionais enfermeiros nas Comissões de Infecção Hospitalar. O método utilizado foi a revisão integrativa, realizada nos meses de julho a outubro de 2020, mediante busca de estudos em bases de dados, revistas, catálogos de teses e dissertações nacionais e internacionais, além de sites de instituições brasileiras de saúde. Incluem-se pesquisas publicadas na íntegra na língua portuguesa, espanhola ou inglesa; que tinham como objeto de estudo a temática investigada, além de manuais e protocolos. Como resultados foram encontrados 16 artigos que corresponderam ao escopo da pesquisa, identificou-se que o profissional enfermeiro é solicitado principalmente no que diz respeito ao processo de educação continuada nas instituições hospitalares. Foi possível identificar que o perfil gerencial que é discutido desde a formação profissional tem impacto positivo quando os enfermeiros ocupam esses espaços.
A prevalência de parasitas intestinais na população está relacionada a precariedade sanitária e aos maus hábitos higiênicos. O presente estudo visa fazer detecção, tanto quantitativamente quanto qualitativamente, de agentes enteroparasitários em alfaces cruas do tipo crespa (Lactuca Sativa) adquiridas em mercados públicos em bairros da região de Recife - PE, utilizando o IDHM como indicador socioeconômico para fazer uma comparação. Foram analisadas 12 amostras de alface crespa (Lactuca sativa) coletadas em seis bairros diferentes e foi utilizado o método de sedimentação de 24 horas seguido de centrifugação, para a pesquisa de parasitas intestinais. Foram detectadas, ao todo, 324 agentes enteroparasitários e entre eles destacam-se: Giardia lamblia (28,4%), Strongyloides stercoralis (24,8%) e Entamoeba Coli (19%). Todos os mercados analisados apresentaram mais de um tipo de enteroparasitas, porém não foi possível confirmar se há uma correlação entre os achados enteroparasitários com o índice socioeconômico IDHM.
