Solanum tuberosum L. DNA sequences containing simple sequence repeat (SSR) motifs were extracted from the EMBL database, cDNA and selectively enriched small-insert DNA libraries. Enrichment was achieved using either triplex affinity capture or single-strand hybridisation selection. One hundred and twelve primer pairs which successfully amplified products of the correct size from potato DNA were ultimately designed and synthesised. Ninety-eight of these revealed length polymorphisms in a panel of four diploid and two tetraploid clones, in agreement with the high information content of this class of markers which has been found in other species. All of the markers were assigned a quality score of 1-5 based on their potential usefulness. Eighty-nine loci from 65 of the primer pairs were located on two genetic linkage maps of potato by segregation analysis of the amplified alleles. Fifty-two of the SSRs were clearly single locus. The maps were aligned using 23 SSR primer pairs and 13 RFLP loci mapped in both populations. The markers described constitute a class which should replace Restriction Fragment Length Polymorphisms (RFLP) as the markers of choice for future genetic studies in potato. The sequences of the primers, together with other information on these markers are provided.
Only limited information on nonsuicidal deliberate self-harm behavior among nonclinical populations is available, and it is unclear whether coping and emotional difficulties remain among those with a past history of self-harm behavior. The purpose of this study is to examine emotions and coping strategies among three nonclinical groups with a recent, past, and no history of nonsuicidal deliberate self-harm behavior. College students completed self-report measures of self-harm behavior, suicidal thoughts, emotional dispositions, and coping strategies. Contrary to expectations, there were few differences in coping strategies among the three groups ( p > .0033). Those with a recent history (n = 23; in the last 12 months) and past history (n = 32; more than 12 months ago) of self-harm behavior reported greater levels of negative emotion (e.g., hostility, guilt, sadness) than those who have never self-harmed (n = 161; p < .0045). This indicates that although self-harm behavior had discontinued (>12 months ago), negative emotion differences were present, and both recent and past self-harmers merit concern in managing their negative emotions to lower their risk for future difficulties.
In eukaryotes, tandem arrays of simple-sequence repeat sequences can find applications as highly variable and multi-allelic PCR-based genetic markers. In hexaploid bread wheat, a large-genome inbreeding species with low levels of RFLP, di- and trinucleotide tandem repeats were found in 22 published gene sequences, two of which were converted to PCR-based markers. These were shown to be genome-specific and displayed high levels of variation. These characteristics make them especially suitable for intervarietal breeding applications.
