Hsp12 of S. cerevisiae is upregulated several 100-fold in response to stress. Our phenotypic analysis showed that this protein is important for survival of a variety of stress conditions, including high temperature. In the absence of Hsp12, we observed changes in cell morphology under stress conditions. Surprisingly, in the cell, Hsp12 exists both as a soluble cytosolic protein and associated to the plasma membrane. The in vitro analysis revealed that Hsp12, unlike all other Hsps studied so far, is completely unfolded; however, in the presence of certain lipids, it adopts a helical structure. The presence of Hsp12 does not alter the overall lipid composition of the plasma membrane but increases membrane stability.

Hybrid lipid/polymer giant unilamellar vesicles: eff ects of incorporated biocompatible PIB-PEO block copolymers on vesicle properties

The interaction of the surfactant octyl glucoside (OG) with dimyristoylphosphatidylcholine (DMPC), dipalmitoylphosphatidylcholine (DPPC), distearoylphosphatidylcholine (DSPC), and soy bean phosphatidylcholine (soy bean PC) was studied using high-sensitivity titration calorimetry. We determined the partition coefficient of OG between water and lipid bilayers and the transfer enthalpy of the surfactant by addition of lipid vesicles to OG monomers or vice versa. Comparison with the micellization enthalpy of the surfactant gives information on differences in the hydrophobic environment of OG in a liquid-crystalline bilayer or a micelle. The average partition coefficient P in mole fraction units for x(e) approximately 0.12-0.2 decreases slightly from 4152 at 27 degrees C to 3479 at 70 degrees C for DMPC and from 4260 to 3879 for soy bean PC, respectively. The transfer enthalpy deltaH(T) of OG into lipid vesicles is positive at 27 degrees C and negative at 70 degrees C. Its temperature dependence is larger for the incorporation of OG into DMPC than into soy bean PC vesicles. It is concluded that OG in DMPC vesicles is better shielded from water than in soy bean PC vesicles or in micelles. Titration calorimetry was also used to determine the phase boundaries of the coexistence region of mixed vesicles and mixed micelles in the systems OG/DMPC, OG/DPPC, OG/DSPC, and OG/soy bean PC vesicles at 70 degrees C in the liquid-crystalline phase. DMPC and soy bean PC solubilization was also studied at 27 degrees C to investigate the effect of temperature. The effective surfactant to lipid ratios at saturation, R(e)(sat), for all PCs studied are in the range between 1.33-1.72 and the ratios at complete solubilization, R(e)(sol), are between 1.79-3.06. At 70 degrees C, the R(e)(sat) values decrease with increasing chain length of the saturated PC. The ratios depend also slightly on temperature and the degree of unsaturation of the fatty acyl chains. For the OG/soy bean PC system, the coexistence range for mixed vesicles and mixed micelles is larger than for the corresponding PCs with saturated chains.

The folding of amyloid beta (1-40) peptide into beta-sheet-containing fibrils is thought to play a causative role in Alzheimer's disease. Because of its amphiphilic character, the peptide can interact with phospholipid membranes. Langmuir monolayers of negatively charged DPPS, DPPG, and DMPG, and also of zwitterionic DPPC and DMPC, have been used to study the influence of the peptide on the lipid packing and, vice versa, the influence of phospholipid monolayers on the peptide secondary structure by infrared reflection absorption spectroscopy and grazing incidence X-ray diffraction. The peptide adsorbs at the air/water (buffer) interface, and also inserts into uncompressed phospholipid monolayers. When adsorbed at the interface, the peptide adopts a beta-sheet conformation, with the long axis of these beta-sheets oriented almost parallel to the surface. If the lipid exhibits a condensed monolayer phase, then compression of the complex monolayer with the inserted peptide leads to the squeezing out of the peptide at higher surface pressures (above 30 mN m(-1)). The peptide desorbs completely from zwitterionic monolayers and negatively charged DPPG and DPPS monolayers on buffer, but remains adsorbed in the beta-sheet conformation at negatively charged monolayers on water. This can be explained in terms of electrostatic interactions with the lipid head groups. It also remains adsorbed at, or penetrating into, disordered anionic monolayers on buffer. Additionally, the peptide does not influence the condensed monolayer structure at physiological pH and modest ionic strength.

We report extended pH- and temperature-induced preparation procedures and explore the materials and molecular properties of different types of hydrogels made from human and bovine serum albumin, the major transport protein in the blood of mammals. We describe the diverse range of properties of these hydrogels at three levels: (1) their viscoelastic (macroscopic) behavior, (2) protein secondary structure changes during the gelation process (via ATR-FTIR spectroscopy), and (3) the hydrogel fatty acid (FA) binding capacity and derive from this the generalized tertiary structure through CW EPR spectroscopy. We describe the possibility of preparing hydrogels from serum albumin under mild conditions such as low temperatures (notably below albumin's denaturation temperature) and neutral pH value. As such, the proteins retain most of their native secondary structure. We find that all the combined data indicate a two-stage gelation process that is studied in detail. We summarize these findings and the explored dependences of the gels on pH, temperature, concentration, and incubation time by proposing phase diagrams for both HSA and BSA gel-states. As such, it has become possible to prepare gels that have the desired nanoscopic and macroscopic properties, which can, in future, be tested for, e.g., drug delivery applications.
