Breast cancer is a common malignancy in women. Acquisition of drug resistance is one of the main obstacles encountered in breast cancer therapy. Long non-coding RNA (lncRNA) has been demonstrated to play vital roles in both development and tumorigenesis. However, the relationship between lncRNAs and the development of chemoresistance is not well established. In the present study, the high expression of lncRNA H19 was identified as a powerful factor associated with paclitaxel (PTX) resistance in ERα-positive breast cancer cells, but not in ERα-negative breast cancer cells. LncRNA H19 attenuated cell apoptosis in response to PTX treatment by inhibiting transcription of pro-apoptotic genes BIK and NOXA. H19 was further confirmed to suppress the promoter activity of BIK by recruiting EZH2 and by trimethylating the histone H3 at lysine 27. Interestingly, our data showed that lncRNA H19 was one of the downstream target molecules of ERα. Altered ERα expression may therefore change H19 levels to modulate the apoptosis response to chemotherapy in breast cancer cells. Our data suggest that the ERα-H19-BIK signaling axis plays an important role in promoting chemoresistance.
Photons can excite the collective and single-particle excitations in metals; the collective plasmonic excitations are of keen interest in physics, chemistry, optics, and nanotechnology because they enhance coupling of the electromagnetic energy and can drive nonlinear processes in electronic materials, particularly where their dielectric function ε(ω) approaches zero. We investigate the nonlinear angle-resolved two-photon photoemission (2PP) spectroscopy of Ag(111) surface through the ε(ω) near-zero region. In addition to the Einsteinian single-particle photoemission, the 2PP spectra report unequivocal signatures of nonlocal dielectric, plasmonically enhanced, excitation processes.
Light interacting with metals elicits an ultrafast coherent many-body screening response on sub-to few-femtosecond time-scales, which makes its experimental observation challenging. Here, we describe the coherent two-dimensional (2D) multi-photon photoemission study of the Shockley surface state (SS) of Ag(111) as a benchmark for spectroscopy of the coherent nonlinear response of metals to an optical field in the perturbative regime. Employing interferometrically time-resolved multi-photon photoemission spectroscopy (ITR-mPP), we correlate the coherent polarizations and populations excited in the sample with final photoelectron distributions where the interaction terminates. By measuring the non-resonant 3-and 4-photon photoemission of the SS state, as well as its replica structures in the above-threshold photoemission (ATP), we record the coherent response of the Ag(111) surface by 2D photoemission spectroscopy and relate it to its band structure. We interpret the mPP process by an optical Bloch equation (OBE) model, which reproduces the main features of the surface state coherent polarization dynamics recorded in ITR-mPP experiments: The spectroscopic components of the 2D photoelectron spectra are shown to depend on the nonlinear orders of the coherent photoemission process m as well as on the induced coherence n.
