The prevalence of psychiatric disorders has increased in recent years. Among existing mental disorders, major depressive disorder (MDD) has emerged as one of the leading causes of disability worldwide, affecting individuals throughout their lives. Currently, MDD affects 15% of adults in the Americas. Over the past 50 years, pharmacotherapy, psychotherapy, and brain stimulation have been used to treat MDD. The most common approach is still pharmacotherapy; however, studies show that about 40% of patients are refractory to existing treatments. Although the monoamine hypothesis has been widely accepted as a molecular mechanism to explain the etiology of depression, its relationship with other biochemical phenomena remains only partially understood. This is the case of the link between MDD and inflammation, mitochondrial dysfunction, and oxidative stress. Studies have found that depressive patients usually exhibit altered inflammatory markers, mitochondrial membrane depolarization, oxidized mitochondrial DNA, and thus high levels of both central and peripheral reactive oxygen species (ROS). The effect of antidepressants on these events remains unclear. Nevertheless, the effects of ROS on the brain are well known, including lipid peroxidation of neuronal membranes, accumulation of peroxidation products in neurons, protein and DNA damage, reduced antioxidant defenses, apoptosis induction, and neuroinflammation. Antioxidants such as ascorbic acid, tocopherols, and coenzyme Q have shown promise in some depressive patients, but without consensus on their efficacy. Hence, this paper provides a review of MDD and its association with inflammation, mitochondrial dysfunction, and oxidative stress and is aimed at thoroughly discussing the putative links between these events, which may contribute to the design and development of new therapeutic approaches for patients.
Doenças neurodegenerativas (DN) são desordens altamente debilitantes relacionadas ao envelhecimento, e a disfunção mitocondrial, estresse oxidativo e nitrosativo (EON) têm sido associados com as manifestações clínicas desse grupo de patologias. As mitocôndrias se destacam como organelas cruciais na relação entre a neurodegeneração e a neuroinflamação, e nesse sentido, os polifenóis são considerados promissores enquanto fármacos que têm como alvo as mitocôndrias. Compostos fenólicos são capazes de regular as mitocôndrias através do controle de seu estado redox, função e sistema apoptótico. Neste trabalho investigou-se o potencial neuroprotetor dos extratos de Araucaria angustifolia (AAE) e Camellia sinensis (GT), bem como de seis compostos isolados (resveratrol, ácido gálico, ácido elágico, catequina, epicatequina e proantocianidinas) em um modelo de neurodegeneração usando ácido quinolínico (AQ). Células gliais U87-MG foram pré-tratadas com AAE ou GT ou um dos compostos fenólicos por 1 hora e, então, expostas ao AQ por 24 horas. As células expostas ao AQ apresentaram diminuição da viabilidade, produção exacerbada de espécies reativas de oxigênio (EROs), redução do potencial de membrana mitocondrial e aumento da resposta inflamatória. Apesar dos resultados similares encontrados com o GT e o AAE, este último se destacou por ser capaz de prevenir todos os parâmetros testados neste trabalho. Em conclusão, sugere-se que o AAE apresenta-se como um agente em potencial para a prevenção de DN relacionadas com a disfunção mitocondrial associada ao EON.
Anticarsia gemmatalis is one of the most important pests in world soybean crop. The most common intervention is the application of agrochemicals, such as novaluron and glyphosate. Among biological control agents, much attention has been drawn to entomopathogenic fungi, as Metarhizium rileyi. Here, we examined the changes that occur in the immune system (total and differential hemocyte count), secondary effects (caterpillar morphology), and oxidative metabolism after the caterpillars were exposed to M. rileyi, novaluron or glyphosate. M. rileyi was able to induce changes in the width, length, and weight of A. gemmatalis pupae, along with an increased in the number of defense cells. Novaluron prompt changes the insect’s immunity, and glyphosate caused milder immunological effects. However, it caused significant secondary effects including malformations in pupae and adults, and an increase in nitric oxide (NO) levels. Mortality observed when treating insects with novaluron and malformations due to glyphosate treatments did not occur due to oxidative stress. However, when insects were exposed to M. rileyi, we verified significantly increased levels of NO and concluded that these insects died due to oxidative stress. Our data provide evidence that contributes to better understanding the mechanism of herbicide-fungus interaction in the management of Anticarsia gemmatalis.
A depressão é considerada um problema de saúde pública e é uma das principais causas de incapacidade profissional ou social em todo o mundo. Antioxidantes são substâncias conhecidas por seus efeitos preventivos ou terapêuticos em várias doenças, incluindo a depressão. Esses benefícios podem ser explicados pela capacidade inerente dessas moléculas de modular as vias de sinalização celular. O objetivo deste estudo foi avaliar os efeitos neuroprotetores do resveratrol em um modelo in vitro de depressão. Estudos experimentais foram realizados com células gliais U-87 MG derivadas da linhagem do cérebro humano. Diferentes concentrações de resveratrol foram utilizadas na presença ou ausência de quinurenina, um metabólito do aminoácido triptofano necessário para a síntese de serotonina. A viabilidade celular foi avaliada determinando a capacidade das células para reduzir o composto MTT (brometo de 3- [4,5-dimetiltiazol 2-il] -2,5-difeniltetrazolina), além da avaliação morfológica celular. Para avaliar o nível de estresse oxidativo, as células gliais tratadas foram submetidas ao teste TBARS para quantificar a peroxidação lipídica. Doses de quinurenina de 100 e 200 µM foram citotóxicas, reduzindo a viabilidade das células gliais e alterando a morfologia celular. Essa redução foi acompanhada por aumento do dano oxidativo aos lipídios. O resveratrol (10 µM) se mostrou preventivo na manutenção da viabilidade das células gliais quando tratadas com quinurenina 100 µM. Os resultados poderão colaborar para o entendimento dos mecanismos envolvidos na depressão, contribuindo com o novo enfoque da capacidade dos polifenóis, compostos naturalmente encontrados na dieta, em tornarem-se agentes terapêuticos adjuvantes para o tratamento de distúrbios neuropsiquiátricos.
