-The objective of this work was to evaluate the total and thermotolerant coliform densities in the oyster culture water of Cananeia, SP, Brazil, correlating these densities with environmental variables and tidal variations. Superficial water samples were collected in two tide conditions (spring and neap) from three areas of Cananéia municipality (Mandira, Itapitangui and Cooperostra). The three studied areas showed good conditions for the culture regarding coliform densities. The two tidal conditions differed significantly as to total coliform concentration; however, the same procedure was not performed for thermotolerant coliforms. No correlation was observed between water temperature, pH, and concentrations of total and thermotolerant coliforms. Coliform density was positively correlated with rainfall and negatively correlated with salinity. Spring and neap tides differed significantly as to coliform number. Simple diagnosis of environmental conditions of the crop fields is insufficient to assess water quality of shellfish cultivation. A continuous monitoring program of planted areas is necessary both for the assessment of water quality potential for marine culture and for ensuring safe consumption of seafood, besides constituting an important tool to understand the relationships between contamination and the involved environmental variables.Index terms: Crassostrea, contamination, microbiology, principal component analysis, total and thermotolerant coliforms.
Densidade de coliformes em águas de cultivo de ostras e sua relação com fatores ambientaisResumo -O objetivo deste trabalho foi avaliar as densidades de coliformes totais e termotolerantes, em água de cultivo de ostra de Cananeia, SP, e correlacionar estas densidades com variáveis ambientais e as variações das marés. Foram coletadas amostras de água superficial em duas condições de maré (sizígia e quadratura) de três áreas do Município de Cananéia (Mandira, Itapitangui e Cooperostra). As três áreas estudadas apresentaram boas condições para cultivo, quanto à densidade de coliformes. Diferenças significativas foram registradas entre as condições de marés quanto à concentração total de coliformes; no entanto, o mesmo procedimento não se aplicou para coliformes termotolerantes. Nenhuma correlação foi observada entre a temperatura da água, o pH e as concentrações de coliformes total e termotolerantes. As densidades de coliformes correlacionaram-se positivamente com a pluviosidade e negativamente com a salinidade. O número de coliformes foi significativamente diferente nas marés de sizígia e de quadratura. O simples diagnóstico das condições ambientais é insuficiente para avaliar a qualidade da água de cultivo de ostras. This study sought to evaluate the microbiological quality of water from the various locations where specimens of the oysters Crassostrea sp. were There was a significant difference between the locations analyzed as to the density of total and thermotolerant coliforms in water. After analysis of the variations of bacterial counting with regard to seasonal mean, autumn presented the highest mean numbers of coliforms in oysters, and most data were collected in April. There was a positive correlation between the coliforms found in water and those in the oyster.
Antimicrobial resistance of marine heterotrophic bacteria to different antimicrobials agents were evaluated in seawater, dry and wet sands from three marine recreational beaches with different pollution levels. In all studied beaches, the greatest frequencies of resistance were found in relation to penicillin. On Gonzaguinha, the most polluted beach, 72.3% of all isolated strains showed simple resistance, whilst 8.33% had multiple resistance. The values found on Ilha Porchat beach, were 70.8% and 6.9% for simple and multiple resistances, respectively. On Guaraú, the less polluted beach, only 35.3% of isolated strains had simple resistance. Multiple resistance was not observed. While samples from Gonzaguinha and Ilha Porchat beach showed isolated strains resistant to seven and six different antimicrobial agents, respectively, samples from Guaraú beach were resistant only to penicillin and erytromicin. The positive correlations obtained between the degree of seawater contamination and frequency and variability of bacterial resistance indicate that polluted marine recreational waters and sands are sources of resistant bacteria contributing thus, to the dissemination of bacterial resistance.
Heavy metals influence the population size, diversity, and metabolic activity of bacteria. In turn, bacteria can develop heavy metal resistance mechanisms, and this can be used in bioremediation of contaminated areas. The purpose of the present study was to understand how heavy metals concentration influence on diversity and distribution of heavy metal-resistant bacteria in Araça Bay, São Sebastião, on the São Paulo coast of Brazil. The hypothesis is that activities that contribute for heavy metal disposal and the increase of metals concentrations in environment can influence in density, diversity, and distribution of heavy metal-resistant bacteria. Only 12 % of the isolated bacteria were sensitive to all of the metals tested. We observed that the highest percentage of resistant strains were in areas closest to the São Sebastião channel, where port activity occurs and have bigger heavy metals concentrations. Bacterial isolated were most resistant to Cr, followed by Zn, Cd, and Cu. Few strains resisted to Cd levels greater than 200 mg L(-1). In respect to Cr, 36 % of the strains were able to grow in the presence of as much as 3200 mg L(-1). Few strains were able to grow at concentrations of Zn and Cu as high as 1600 mg L(-1), and none grew at the highest concentration of 3200 mg L(-1). Bacillus sp. was most frequently isolated and may be the dominant genus in heavy metal-polluted areas. Staphylococcus sp., Planococcus maritimus, and Vibrio aginolyticus were also isolated, suggesting their potential in bioremediation of contaminated sites.
A B S T R A C TUrban development in coastal areas is intense and leads to the increase of sewage outfall and other negative impacts as consequences. Thus, stringent regulations establishing limits to the microbiological contamination of water and seafood are needed. The objective of this study is to evaluate the usefulness of Enterococci and Thermotolerant Coliform densities in the flesh of mussels Perna perna as an alternative tool for monitoring the microbiological quality of coastal waters. The study also considers allometric relations applied to clearance rates to understand rates of bacterial concentration. Bacterial loads obtained in mussels' flesh were from 50 to 4,300 times greater than in the water sampled in the vicinity of the mussels and some were considered inappropriate for consumption even when the water presented no restrictions. The mean clearance rate obtained for Enterococci retention was 317.7 ml h -1 and this rate (CR) is related to the mussels´ size (L) by the equation CR = 28.3229L1.6421 . The results showed that bacterial densities in the mussels' tissues may reflect chronic contamination of the environment and that clearance rates are important for taking the best decision in situations where, for example, it is desired to introduce mussels for aquaculture and the evaluation of the water concerned is required.
R E S U M OO desenvolvimento urbano em áreas costeiras é intenso e o aumento das descargas de esgoto é uma das conseqüências. Assim, leis específicas que regulem limites para a contaminação microbiológica das águas e de alimentos de origem marinha são necessárias. O objetivo deste estudo é avaliar a utilização de Enterococos e coliformes termotolerantes na carne de mexilhões Perna perna como alternativa para o monitoramento da qualidade microbiológica de águas costeiras. O estudo também considera relações alométricas aplicadas às taxas de "clearance" para entender a concentração de bactérias pelo molusco. As densidades bacterianas obtidas no molusco foram de 50 a 4300 vezes maiores do que nas amostras de água coletadas próximas dos moluscos e alguns foram considerados impróprios para consumo, mesmo quando as águas não mostraram restrições. A taxa de "clearance" média obtida para retenção de Enterococos foi 317.7 ml h -1 e essas taxas (CR) puderam ser relacionadas com o tamanho dos mexilhões (L) pela equação CR = 28.3229L 1.6421 . Os resultados mostram que as densidades bacterianas nos tecidos dos mexilhões podem refletir a contaminação crônica do ambiente e que as taxas de "clearance" devem ser consideradas para a tomada de decisão em situações onde, por exemplo, se deseja implantar mexilhões para cultivo.
