INTRODUÇÃO: A inflamação é caracterizada pela coordenada ativação de várias vias de sinalização que regulam a expressão de mediadores tanto pró como anti-inflamatórios, acredita-se que outros polissacarídeos de espécies do gênero Caesalpinia, como o polissacarídeo extraído da semente da C. ferrea, possa apresentar ação anti-inflamatória e antinociceptiva no sistema gastrointestinal. OBJETIVO: Analisar a ação anti-inflamatória do polissacarídeo extraído da semente da C. ferrea sobre modelos de edema de para induzido por serotonina e sobre a migração leucocitária. MÉTODOS: Foram realizados experimentos in vivo, onde foram utilizados 6 animais por grupo (camundongos Balb/C machos). RESULTADOS: A dose de 10mg/kg (i.p.) do PLS extraído da C. ferrea foi capaz de reduzir significativamente (p< 0,05) o volume da pata e a migração de leucocitária, uma vez que a migração de neutrófilos e macrófagos correspondem a terceira etapa vascular da inflamação, sendo assim, benéfica a redução do número de leucócitos. CONCLUSÃO: O PLS extraído da C. ferrea administrado na dose de 10mg/kg tem possível potencial efeito anti-inflamatório.
INTRODUÇÃO: A inflamação pode ser dividida em aguda ou crônica conforme o tempo de duração da inflamação. Uma espécie nativa e endêmica no território brasileiro, popularmente conhecida como “pau ferro” ou “jucá”, a Libidibia férrea é muito utilizada na medicina popular, acredita-se que o polissacarídeo extraído da sua semente, possa apresentar ação anti-inflamatória no sistema gastrointestinal. OBJETIVO: Avaliar a atividade anti-inflamatória do polissacarídeo extraído da semente da C. ferrea sobre modelos de edema de pata induzido por Carragenina, sobre a concentração da enzima mioloperoxidase (MPO) e malondealdeido (MDA). MÉTODOS: Foram realizados experimentos in vivo, onde foram utilizados 6 animais por grupo (camundongos Balb/C machos). RESULTADOS: O PLS da C. ferrea mostrou uma diminuição significativa do edema de pata e nos níveis de MPO e MDA. CONCLUSÃO: Verifica-se que o PLS extraído da C. ferrea administrado na dose de 10mg/kg apresentou potencial anti-inflamatório, sendo capaz de reduzir o edema provocado pela carragenina, os níveis da enzima Mieloperoxidase (MPO) e ácido malondialdeido (MDA) no local da inflamação.
INTRODUÇÃO: As Doenças Inflamatórias Intestinais são um grupo de distúrbios inflamatórios crônicos que acometem o trato gastrointestinal. Neste grupo insere-se a Retocolite Ulcerativa, caracterizada pela inflamação da mucosa do cólon e reto. Suas abordagens terapêuticas objetivam reduzir danos ocasionados pelo processo inflamatório e seus sintomas, entretanto, essas terapêuticas possuem diversos efeitos adversos sendo necessário buscar medicamentos com menos efeitos secundários. A espécie Pilocarpus microphyllus do Jaborandi, é uma planta que possui diversos alcaloides com propriedades farmacológicas, entre elas a pilocarpina, um alcaloide colinérgico. Entre os resíduos industriais gerados pela sua produção encontra-se a epiisopiloturina (EPI), um alcaloide imidazólico com propriedades antiparasitárias e anti-inflamatórias. O receptor muscarínico M1 possui ação anti-inflamatória intestinal quando estimulado, e assim levantou-se a hipótese que a epiisopiloturina, estruturalmente semelhante ao agonista muscarínico pilocarpina, tenha seu mecanismo de ação por essa via. OBJETIVO: Avaliar a participação do receptor muscarínico M1 na atividade anti-inflamatória da epiisopiloturina através de parâmetros inflamatórios morfológicos. MÉTODOS: Para a avaliação da atividade anti-inflamatória da EPI, os animais receberam a substância nas doses 0,01, 0,1 e 1,0 (mg/kg, i.p.), 17 horas após a indução da retocolite ulcerativa. A avaliação do receptor muscarínico M1 na reposta anti-inflamatória da EPI foi testada pelo bloqueio da ação de epiisopiloturina com o antagonista seletivo pirenzepina (10 mg/kg, i.p.). Foram utilizados parâmetros morfológicos (peso úmido, escores macroscópicos e microscópicos de lesão) na avaliação da atividade anti-inflamatória. RESULTADOS: A EPI na dose 0,1 mg/kg foi capaz de reverter os parâmetros morfológicos de inflamação, diminuindo o peso úmido e as lesões macro e micro do tecido. Os animais que receberam epiisopiloturina juntamente com o antagonista pirenzepina apresentaram possível bloqueio dos efeitos anti-inflamatórios. CONCLUSÃO: Infere-se que a ação anti-inflamatória da epiisopiloturina se dá possivelmente pelo receptor muscarínico M1, pois, após o bloqueio deste receptor, a epiisopiloturina teve seu efeito anti-inflamatório bloqueado.
INTRODUÇÃO: Os tratamentos convencionais da colite ulcerativa apresentam diversos efeitos colaterais, fazendo-se necessário o desenvolvimento de alternativas terapêuticas, como as moléculas advindas de plantas medicinais. Um exemplo é o alcalóide epiisopiloturina da espécie Pilocarpus microphyllus, que já mostrou atividade anti-inflamatória em estudos anteriores. Entretanto esse al-calóide apresenta baixa absorção após a administração oral, podendo ser melhorado por meio de nanoformulações. OBJETIVO: O presente trabalho visa estudar de forma comparativa o efeito da epiisopiloturina nanoencapsulada com a goma do cajueiro e da epiisopiloturina livre sobre os parâ-metros inflamatórios da colite induzida por TNBS em ratos. MÉTODOS: Foram produzidas nano-partículas a base de goma do cajueiro modificada pelo método de nanoprecipitação. A atividade anti-inflamatória, foi analisada sobre os níveis de MDA, GSH e MPO, no cólon de ratos com colite induzida experimentalmente por TNBS. RESULTADOS E DISCUSSÃO: Os resultados encontra-dos mostram que epiisopiloturina nanoencapsulada com a goma do cajueiro mostrou maior ativida-de anti-inflamatória que a sua forma livre ao reduzir parâmetros oxidativos como MPO e MDA e aumentar os níveis de GSH. CONCLUSÃO: Diante dos resultados até aqui obtidos conclui-se que a epiisopiloturina nanoencapsulada melhora a atividade anti-inflamatória em relação a sua forma livre, tornando-a uma possível promissora para a produção de um novo fitoterápico para o tratamen-to da colite.
