The objective of this study was to investigate the use of chloroquine (CLQ) as an antiviral agent against dengue. Chloroquine, an amine acidotropic drug known to affect intracellular exocytic pathways by increasing endosomal pH, was used in the in vitro treatment of U937 cells infected with dengue virus type 2 (DENV-2). Viral replication was assessed by quantification of virus produced through detection of copy numbers of DENV-2 RNA, plaque assay and indirect immunofluorescence. qRT-PCR and plaque assays were used to quantify the DENV-2 load in infected U937 cells after CLQ treatment. It was found that a dose of 50 mg/mL of CLQ was not toxic to the cells and resulted in significantly less virus production in infected U937 cells than occurred in untreated cells. In the present work, CLQ was effective against DENV-2 replication in U937 cells, and also caused a statistically significant reduction in expression of proinflammatory cytokines. The present study indicates that CLQ may be used to reduce viral yield in U937 cells.
Hantavirus Pulmonary Syndrome is an, often fatal, emerging zoonotic disease in the Americas caused by hantaviruses (family: Hantaviridae). In Brazil, hantavirus routine diagnosis is based on serology (IgM-ELISA) while RT-PCR is often used to confirm acute infection. A Semi-nested RT-PCR and an internally controlled RT-qPCR assays were developed for detection and quantification of four hantaviruses strains circulating in the Brazilian Amazon: Anajatuba (ANAJV) and Castelo dos Sonhos (CASV) strains of Andes virus (ANDV) species; and Rio Mamoré (RIOMV) and Laguna Negra (LNV) strains of LNV species. A consensus region in the N gene of these hantaviruses was used to design the primer sets and a hydrolysis probe. In vitro transcribed RNA was diluted in standards with known concentration. MS2 bacteriophage RNA was detected together with hantavirus RNA as an exogenous control in a duplex reaction. RT-qPCR efficiency was around 100% and the limit of detection was 0.9 copies/μL of RNA for RT-qPCR and 10 copies/μL of RNA for Semi-nested RT-PCR. There was no amplification of either negative samples or samples positive to other pathogens. To assess the protocol for clinical sensitivity, specificity and general accuracy values, both assays were used to test two groups of samples: one comprising patients with disease (n = 50) and other containing samples from healthy individuals (n = 50), according to IgM-ELISA results. A third group of samples (n = 27) infected with other pathogens were tested for specificity analysis. RT-qPCR was more sensitive than semi-nested RT-PCR, being able to detect three samples undetected by conventional RT-PCR. RT-qPCR clinical sensitivity, specificity and general accuracy values were 92.5%, 100% and 97.63%, respectively. Thus, the assays developed in this study were able to detect the four Brazilian Amazon hantaviruses with good specificity and sensitivity, and may become powerful tools in diagnostic, surveillance and research applications of these and possibly other hantaviruses.
Introdução: O SARS-CoV-2, Betacoronavirus da família Coronaviridae, é o agente etiológico da covid-19 e teve a sua origem em dezembro de 2019 na província de Wuhan (China) causando um surto de síndrome respiratória aguda grave em alguns indivíduos que logo se disseminou por todo o mundo, caracterizando a atual pandemia. Tão logo a doença foi descoberta, diversas vacinas começaram a ser desenvolvidas, e em dezembro de 2020 a primeira delas já estava disponível. Em 17 de janeiro de 2021, iniciou-se a vacinação contra a covid-19 no Brasil e atualmente dispomos de quatro tipos de imunizantes aprovados e administrados em todo o território nacional. Objetivo: Este trabalho tem por objetivo analisar a vacinação da covid-19 no Brasil no ano de 2021 por estado e por região. Material e métodos: Trata-se de um estudo transversal descritivo no qual os dados foram consultados e coletados em fontes públicas. A quantidade de doses administradas foi obtida no site Ministério da Saúde (foi considerado a população com o esquema vacinal completo - duas doses ou dose única) e o dado populacional foi obtido do Instituto Brasileiro de Geografia e Estatísticas (ano 2021). Os dados foram plotados em planilha do programa Excel e para obter a porcentagem da população vacinada foi realizado um cálculo simples: Número de indivíduos vacinados / População local x 100%. Resultados: Até 09 de dezembro de 2021, o Brasil possui 139.436.786 pessoas com o esquema vacinal completo, o que corresponde a 65% da população. A região com o maior número de imunizados é a Sudeste (72%), seguida da região Sul (70%), Centro-Oeste (61%), Nordeste (59%) e Norte (50%). O estado que mais vacinou foi o de São Paulo com 79% de sua população vacinada, enquanto o estado que menos vacinou foi o Amapá, com apenas 36%. Conclusão: A vacinação contra a covid-19 no Brasil segue em ritmo desigual tanto por estado quanto por região e representa um verdadeiro desafio para o fim da pandemia no nosso país e no mundo.
Introdução: O SARS-CoV-2, Betacoronavirus da família Coronaviridae, é um coronavírus emergente que teve a sua origem no final de 2019 na China e logo se disseminou pelo mundo, causando a atual pandemia. A sua velocidade de disseminação foi acompanhada pelo surgimento de variantes e atualmente a Organização Mundial de Saúde (OMS) considera as seguintes variantes de preocupação (VOC): Gama, Alfa, Beta, Delta e Ômicron. Objetivo: Descrever o monitoramento das variantes de preocupação identificadas no Brasil através de caracterização genômica durante o ano de 2021. Material e Métodos: Trata-se de um estudo transversal descritivo no qual os dados foram coletados de Boletins Epidemiológicos divulgados pelo Ministério da Saúde e correspondem as semanas epidemiológicas (SE) 01 a 51 de 2021. Resultados: Até a SE 51 de 2021, o Brasil identificou 48.813 registros de covid-19 causado por VOC. As cinco VOC estão presentes no território brasileiro com predominância da Delta (50%) e Gama (48,5%), seguidas das VOC Alfa (1,1%), Ômicron (0,36%) e Beta (0,01%). Mais da metade das VOC identificadas foram da região Sudeste (56,1%), as demais foram provenientes das regiões Centro-Oeste e Nordeste – ambas as regiões com 13%, Norte (9,5%) e Sul (8,4%). Em relação aos estados, os cinco que mais tiveram amostras sequenciadas foram: São Paulo (28,9%), Rio de Janeiro (13,8%), Minas Gerais (11,2), Amazonas (5,57%) e Santa Catarina (5,36%). Conclusão: O monitoramento das VOC é essencial para o entendimento do curso epidemiológico da covid-19 no mundo. Por isso, o fortalecimento da rede laboratorial no Brasil é tão importante para que essa identificação possa ocorrer de forma homogênea entre os estados e com maior proporcionalidade em relação ao número total de casos notificados de covid-19.
Introdução: As arboviroses são doenças virais transmitidas por artrópodes. O Brasil, país tropical com aproximadamente metade do seu território constituído por florestas tropicais, apresenta as condições ideais para a múltipla circulação de arbovírus, sendo a dengue, chikungunya, zika e febre amarela silvestre endêmicas no nosso país. Objetivo: O objetivo deste trabalho é realizar um levantamento epidemiológico das arboviroses urbanas (dengue, chikungunya e zika) circulantes no Brasil referente ao ano de 2021. Material e métodos: Os dados foram coletados de Boletins Epidemiológicos divulgados pelo Ministério da Saúde e do Sinan Online e correspondem as semanas epidemiológicas 01 a 52 de 2021. Resultados: Durante o ano de 2021 foram notificados 544.460 casos prováveis de dengue, 96.288 casos prováveis de chikungunya e 6.483 casos prováveis de zika. As regiões com maior incidência de casos foram a região Centro-Oeste para a dengue e região Nordeste tanto para chikungunya quanto para zika. O número casos de dengue e zika apresentaram uma diminuição de 43% e 12%, respectivamente, em relação ao número de casos registrados no ano anterior (2020), diferentemente do número de casos de chikungunya, que apresentam um aumento de quase 33%. Em relação ao número de óbitos já foram confirmados 240 por dengue e 14 por chikungunya, e nenhum registro ainda por zika. Conclusão: A dengue continua sendo a arbovirose mais importante e predominante no nosso país e o diagnóstico diferencial é uma ferramenta fundamental para o monitoramento e investigação dos casos, já que este tripé de arboviroses é uma ameaça constante à saúde pública no nosso país.
