At present, aquaculture is the fastest growing sector of animal food production and holds great potential as a sustainable solution for world food security. The ability to control sex is one of the most important factors for the commercialisation and efficient propagation of fish species, due to influences on reproduction, growth and product quality. Accordingly, there is a large body of research that targets sexual development in commercially important species in an attempt to understand and control fish sex and reproductive function. In this review, we provide an introduction to sex determination and differentiation in fish, including the genetic, epigenetic and environmental factors that can influence fish sex ratios. We also summarise the major approaches used to control sex in fish and discuss their application in commercially important species. Specifically, we discuss the use of exogenous steroid hormones, chromosome ploidy, environmental manipulations, sexlinked genetic markers, selection for altered sex ratios, and transgenics and comment on the challenges associated with controlling sex in a commercial environment.

Fibropapillomatosis (FP) is a marine turtle disease recognised by benign tumours on the skin, eyes, shell, oral cavity and/or viscera. Despite being a globally distributed disease that affects an endangered species, research on FP and its likely causative agent chelonid alphaherpesvirus 5 (ChHV5) in Australia is limited. Here we present improved molecular assays developed for detection of ChHV5, in combination with a robust molecular and phylogenetic analysis of ChHV5 variants. This approach utilised a multi-gene assay to detect ChHV5 in all FP tumors sampled from 62 marine turtles found at six foraging grounds along the Great Barrier Reef. Six distinct variants of ChHV5 were identified and the distribution of these variants was associated with host foraging ground. Conversely, no association between host genetic origin and ChHV5 viral variant was found. Together this evidence supports the hypothesis that marine turtles undergo horizontal transmission of ChHV5 at foraging grounds and are unlikely to be contracting the disease at rookeries, either during mating or vertically from parent to offspring.

Epigenetics is involved in sex differentiation of gonochoristic and hermaphroditic fish species, whereby two genes dmrt1 (pro-male) and cyp19a1 (pro-female) are known to play major roles. Barramundi, Lates calcarifer, is an important tropical aquaculture species that undergo natural and permanent male to female sex change, a process for which the exact underlying molecular mechanisms are still unknown. To elucidate whether DNA methylation is involved in sex control of barramundi, a next-generation bisulfite amplicon sequencing approach was used to target 146 CpG sites within proximal promoters and first exons of seven sex-related genes (dmrt1, cyp19a1, amh, foxl2, nr5a2, sox8 and sox9) of 24 testis and 18 ovaries of captive and wild adult barramundi. Moreover, comparative expression profiles of the key dmrt1 and cyp19a1 genes were further investigated using RT-qPCR and Sanger sequencing approaches, whereas expression levels of remaining targeted genes were based on available literature for the species. Results showed that cyp19a1 and amh were more methylated in males, whereas dmrt1 and nr5a2 were more methylated in females (P < 0.001), with no gender differences found for foxl2, sox8 or sox9 genes (P > 0.05). Sex-biased promoter DNA methylation was inversely related to gene expression only for dmrt1 and nr5a2, and directly related to amh expression, whereas no differences in cyp19a1 expression were found between testes and ovaries. Notably, unique sex-specific alternative splicing of dmrt1 and cyp19a1 were discovered, whereby males lacked the full-length aromatase coding cyp19a1 mRNA due to partial or total exon splicing, and females lacked the dmrt1 exon containing the DM-domain sequence. This study advances the current knowledge aiming to elucidate the genetic mechanisms within male and female gonads of this large protandrous hermaphrodite by providing the first evidence of epigenetics and alternative splicing simultaneously affecting key genes (cyp19a1 and dmrt1) central to sex differentiation pathways.

19The deep red colour of the Giant Tiger prawn, Penaeus mondon, is highly desired 20 and fetches premium market prices. Prawn pigmentation is influenced by the 21 interaction of a range of factors, including the amount of dietary carotenoid, the 22 distribution of hypodermal pigments, and genetics. These aspects have been 23 studied in isolation, but there is limited knowledge on how these components 24 interact to influence prawn pigmentation. This study tracked the colour of 25 prawns that had been fed four different levels of dietary astaxanthin (Axn) over 26 6 weeks, and then transferred to either black or white coloured tanks. The 27 dietary influence on colour was slow and had only developed after 6-weeks.

Meat eating quality with a healthy composition hinges on intramuscular fat (IMF), fat melting point (FMP), tenderness, juiciness, flavour and omega-3 long-chain polyunsaturated fatty acids (n-3 LC-PUFA) content. These health-beneficial n-3 LC-PUFA play significant roles in optimal cardiovascular, retinal, maternal and childhood brain functions, and include alpha linolenic (ALA), eicosapentaenoic (EPA), docosahexaenoic (DHA) and docosapentaenoic (DPA) acids. The primary objective of this review was to access, retrieve, synthesise and critically appraise the published literature on the synthesis, metabolism and genetics of n-3 LC-PUFA and meat eating quality. Studies on IMF content, FMP and fatty acid composition were reviewed to identify knowledge gaps that can inform future research with Tattykeel Australian White (TAW) lambs. The TAW is a new sheep breed exclusive to MARGRA brand of lamb with an outstanding low fat melting point (28–39°C), high n-3 LC-PUFA EPA+DHA content (33–69mg/100g), marbling (3.4–8.2%), tenderness (20.0–38.5N) and overall consumer liking (7.9–8.5). However, correlations between n-3 LC-PUFA profile, stearoyl-CoA desaturase (SCD), fatty acid binding protein 4 (FABP4), fatty acid synthase (FASN), other lipogenic genes and meat quality traits present major knowledge gaps. The review also identified research opportunities in nutrition–genetics interactions aimed at a greater understanding of the genetics of n-3 LC-PUFA, feedlot finishing performance, carcass traits and eating quality in the TAW sheep. It was concluded that studies on IMF, FMP and n-3 LC-PUFA profiles in parental and progeny generations of TAW sheep will be foundational for the genetic selection of healthy lamb eating qualities and provide useful insights into their correlations with SCD, FASN and FABP4 genes.

Numerous mathematical models have been put forward to describe these patterns, but all rely on assumptions regarding the nature of the pigments and the cells from which they are secreted. We investigated pigmentation and cellular morphology of the mantle (shell-secreting organ) of the tropical abalone, Haliotis asinina, as a crucial step towards understanding the molecular mechanisms of shell patterning in this gastropod mollusc. Results: Accumulation and release of pigmented products occurs within secretory tubules in the prismatic zone of the juvenile H. asinina mantle. The colour observed within these tubules closely matches the colour deposited most recently within the shell lying directly above. The pigments are autofluorescent, and confocal microscopy reveals that multiple pigments can be present within a single tubule at any one time. Examination of mantle morphology in other gastropods reveals that mantle tubules are not restricted to abalone, but are not universal. Conclusions: The presence of a tubule-based secretory system within the abalone mantle demonstrates that pigmentation is not controlled by a simple line of cells. Instead, coordination of patterning events is likely to be modular, with signals received by individual cells being transmitted throughout the entire tubule to synchronise the accumulation and secretion of pigmented material.
