A banana é a segunda fruta mais consumida no mundo, porém do campo até o mercado consumidor algo em torno de 40 % é perdido devido entre outras causas as doenças pós-colheita e a mais significativa é a antracnose. Diante da necessidade do conhecimento de fatores ambientais que condicionam estas perdas, o trabalho objetivou avaliar métodos de inoculação (com discos de BDA e estruturas do patógeno e suspensão de conídios com e sem ferimento) e a influência da temperatura (10,15,20,25 e 30 ºC) e do período de molhamento (0, 12, 24 e 36 h) sobre o desenvolvimento de Colletotrichum musae em banana. As frutas foram inoculadas com 17 isolados de C. musae onde todos mostraram-se patogênicos quando inoculados com ferimento independentemente do tipo de inóculo utilizado. No experimento envolvendo temperatura e período de molhamento, utilizou-se três isolados de C. musae, MAG2, SFV1 e FSA, que se comportaram como mais agressivo, intermediário e pouco agressivo, respectivamente. As temperaturas em torno de 20, 25 e 30 ºC e os períodos de molhamento testados favoreceram um maior desenvolvimento de lesões, sendo as maiores lesões observadas em temperaturas ao redor de 25 e 30 ºC, com redução à medida que ocorria uma diminuição da temperatura para todos os isolados testados. A temperatura em torno de 15 ºC proporcionou o menor desenvolvimento da doença.,
O objetivo dessa pesquisa foi verificar o efeito de produtos alternativos em mangas inoculadas com Lasiodiplodia theobromae e a influência desses sobre o crescimento micelial do fitopatógeno. Mangas foram tratadas com fosfato de potássio (FP) (50, 100 e 150), ácido hidroxidobenzóico (AH) (5, 10 e 15), cloreto de cálcio (CC) (0,13, 0,26 e 0,39) (em milimolar), Luz ultravioleta (LUV) (10; 20 e 30 min.), extratos de alho (EA), melão-de-são-caetano (EM), casca de manga (ECM) (25, 50 e 75%) e água (testemunha). Inoculou-se 10m de suspensão de 10(6) conídios/mL de L. theobromae imediatamente (T1), 12 horas (T2) e 24 horas, após os tratamentos (T3). No T1, os melhores tratamentos foram FP (50 mmol) e CC (0,13 mmol). No T2 foram CC (0,13 mmol), AH (5 mmol), EA (25%) e FP (100 mmol). No T3 não houve diferença entre os tratamentos. In vitro, os produtos foram misturados ao meio de cultura. Após solidificação, depositaram-se no centro de cada placa, estruturas do fitopatógeno. Quarenta e oito horas após, mediu-se o diâmetro da colônia. Os melhores tratamentos foram FP (100 e 150 mmol), ECM (50 e 75%) e EA (50%).
Although ecotone areas occupy a significant extent in Piauí State, there is little information about these areas, especially regarding the presence of microorganisms. Thus, this study evaluated the effect of seasonality on the occurrence of fungal genera associated with forest species in an ecotone Cerrado-Caatinga in Piauí State, Brazil. The experimental area consisted of one-hectare fragment within a legal reserve, where five plots of 20m x 20m were established and the phytosociological survey was carried out. The collection of the material (healthy leaves and leaves with disease symptoms) was performed in two periods: the dry season (June and August/2017) and the rainy season (December/2017 and February/2018), totaling four collections. In the laboratory, isolation and purification of fungi were performed. The fungal identification was based on morphological characteristics and the use of identification keys. The relative abundance of fungi was calculated, and the data were submitted to multivariate cluster analysis. In total, 225 isolates were found, related to 15 fungal genera, associated with 10 forest species. The occurrence of fungi in the area was influenced by seasonal variation, with more isolates found in the dry period, as examples of important groups, whether beneficial or phytopathogenic microorganisms are: Trichoderma spp. and Lasiodiplodia spp. It was also observed that fungi are more likely to occur in forest species like Bauhinia cheilantha and Pityrocarpa moniliformis, belonging to the Fabaceae family.
This study aimed to assess the potential of 19 isolates of Trichoderma spp. originating from a Cerrado-Caatinga ecotone in control of Colletotrichum truncatum, in soybean. In vitro tests (inhibitory effect of volatile and fixed thermostable metabolites) were carried out to assess the percentage of mycelial growth of the pathogen and in vivo tests (in soybean seeds) were performed to assess the percentage of germination, germination rate index, dry matter, and health of seeds. Regarding the pathogen growth under the effect of volatile and fixed thermostable metabolites, the lowest values were provided, respectively, by the isolates UFPIT03 (49.25%) and UFPIT13 (38.38%). No significant differences were observed in the in vivo tests regarding the action of volatile metabolites of the different isolates. In relation to the effect of fixed metabolites, the isolates UFPIT11 and UFPIT19 presented the best results, with the highest means of seed dry matter and lower pathogen incidence. However, the other isolates also showed positive results. It is noteworthy that this is the first report of Trichoderma orientale in the biocontrol of a plant pathogen. Thus, the perspectives of using these species of Trichoderma originating from native forest fragments in the biological control of phytopathogens are evidenced.Bioprospecção de Trichoderma spp. originadas de um ecótono Cerrado-Caatinga sobre Colletotrichum truncatum, em soja RESUMO: Objetivou-se com o presente trabalho avaliar o potencial de 19 isolados de Trichoderma spp. originárias de um ecótono Cerrado-Caatinga no controle de Colletotrichum truncatum, em soja. Foram realizados testes in vitro (efeito inibitório de metabólitos voláteis e fixos termoestáveis), em que se avaliou a porcentagem de crescimento micelial do patógeno e testes in vivo (em sementes de soja), sendo avaliados a porcentagem de germinação, o índice de velocidade de germinação, a massa seca e a sanidade das sementes. Em relação ao crescimento do patógeno, sob efeito de metabólitos voláteis e fixos termoestáveis, os menores valores foram proporcionados, respectivamente, pelos isolados UFPIT03 (49,25%) e UFPIT13 (38,38%). Nos testes in vivo, não foram observadas diferenças significativas quanto a ação de metabólitos voláteis dos diferentes isolados. Em se tratando do efeito de metabólitos fixos, os isolados UFPIT11 e UFPIT19 apresentaram os melhores resultados, proporcionando as maiores médias de massa seca da semente e menor incidência do patógeno. Contudo, os demais isolados também apresentaram resultados positivos. Vale ressaltar que este é o primeiro relato de Trichoderma orientale no biocontrole de um fitopatógeno. The indiscriminate use of chemical pesticides increasingly harms the health of living beings and the environment. Thus, biological control carried out by microorganisms has gained prominence, since it consists of an environmentally friendly alternative to the use of pesticides for controlling plant diseases. Herein, we evaluated the potential role of endophytic Trichoderma strains isolated from forest species of the Cerrado-Caatinga ecotone as biological control agents of crop pathogenic fungi. Nineteen Trichoderma strains were used to assess the antagonistic activity by in vitro bioassays against the plant pathogens Colletotrichum truncatum, Lasiodiplodia theobromae, Macrophomina phaseolina, and Sclerotium delphinii isolated from soybean, cacao, fava bean, and black pepper crops, respectively. All Trichoderma strains demonstrated inhibitory activity on pathogen mycelial growth, with maximum percent inhibition of 70% against C. truncatum, 78% against L. theobromae, 78% against M. phaseolina, and 69% against S. delphinii. Crude methanol extracts (0.5 to 2.0 mg mL-1) of Trichoderma strains were able to inhibit the growth of C. truncatum, except Trichoderma sp. T3 (UFPIT06) and T. orientale (UFPIT09 and UFPIT17) at 0.5 mg mL-1, indicating that the endophytes employ a biocontrol mechanism related to antibiosis, together with multiple mechanisms. Discriminant metabolites of Trichoderma extracts were unveiled by liquid chromatography-tandem mass spectrometry-based metabolomics combined with principal component analysis (PCA), which included antifungal metabolites and molecules with other bioactivities. These results highlight the biocontrol potential of Trichoderma strains isolated from the Cerrado-Caatinga ecotone against crop pathogenic fungi, providing support for ongoing research on disease control in agriculture.
Resumo -O objetivo deste trabalho foi avaliar o efeito da radiação gama em doses baixas no controle pós-colheita da podridão por Fusicoccum em manga 'Tommy Atkins', bem como avaliar o efeito desse método sobre as características físico-químicas da fruta. Frutos aparentemente livres de doenças no estádio de maturação 1,5 foram inoculados com 10 µL de suspensão de Fusicoccum parvum a 10 6 conídios mL -1 . Após a inoculação, os frutos foram irradiados com as doses de 0,24, 0,35 e 0,45 kGy e armazenados a 13ºC, durante 15 dias, seguidos de mais seis dias em temperatura ambiente, a 25ºC. A dose mais alta de radiação gama foi eficiente em retardar o desenvolvimento da doença em razão do atraso causado na maturação das frutas. Não houve efeito significativo da radiação sobre as características físico-químicas das frutas. Fusicoccum rot in mangoes submitted to low doses of gamma irradiationAbstract -This study aimed to evaluate the effect of gamma irradiation in low doses on the postharvest control of 'Tommy Atkins' mango rot caused by Fusicoccum, and to evaluate the effect of this method on the physicochemical characteristics of the fruit. Fruits in ripening stage 1,5 and apparently free of disease were inoculated with 10 μL of a Fusicoccum parvum suspension at 10 6 conidia mL -1 . They were gamma irradiated with the doses 0.24, 0.35 and 0.45 kGy, and stored under 13°C for 15 days, followed by a further six days at room temperature, 25°C. The higher dose of gamma radiation was effective in delaying the disease development due to fruit delayed maturation. No significant effect of radiation was observed on fruit physicochemical characteristics.
