For many years, there has been uncertainty concerning the reason for Hsp70 translocation to the nucleus and nucleolus. Herein, we propose that Hsp70 translocates to the nucleus and nucleoli in order to participate in pathways related to the protection of the nucleoplasmic DNA or ribosomal DNA from single-strand breaks. The absence of Hsp70 in HeLa cells, via Hsp70 gene silencing (knockdown), indicated the essential role of Hsp70 in DNA integrity. Therefore, HeLa Hsp70 depleted cells were very sensitive in heat treatment and their DNA breaks were multiple compared to that of control HeLa cells. The molecular mechanism with which Hsp70 performs its role at the level of nucleus and nucleolus during stress was examined. Hsp70 co-localizes with PARP1 in the nucleus/nucleoli as was observed in confocal studies and binds to the BCRT domain of PARP1 as was revealed with protein-protein interaction assays. The aim of this study was to investigate the potential protective effect of the Hsp70 protein in the cardiac dysfunction induced by doxorubicin (DOX) and the mechanisms of its action. For this purpose, we used both wild-type mice (F1/F1) and Hsp70-transgenic mice (Tg/Tg) overexpressing human HSP70. Both types were subjected to chronic DOX administration (3 mg/kg intraperitoneally every week for 10 weeks, with an interval from weeks 4 to 6). Primary cell cultures isolated from embryos of these mice were also studied. During DOX administration, the mortality rate as well as weight reduction were lower in Tg/Tg compared to F1/F1 mice (P < 0.05). In vivo cardiac function assessment by transthoracic echocardiography showed that the reduction in left ventricular systolic function observed after DOX administration was lower in Tg/Tg mice (P < 0.05). The study in primary embryonic cell lines showed that the apoptosis after incubation with DOX was reduced in cells overexpressing Hsp70 (Tg/Tg), while the apoptotic pathway that was activated by DOX administration involved activated protein factors such as p53, Bax, caspase-9, caspase-3, and PARP-1. In myocardial protein extracts from identical mice with DOX-induced heart failure, the particular activated apoptotic pathway was confirmed, while the presence of Hsp70 appeared to inhibit the apoptotic pathway upstream of the p53 activation. Our results, in this DOX-induced heart failure model, indicate that Hsp70 overexpression in Tg/Tg transgenic mice provides protection from myocardial damage via an Hsp70-block in p53 activation, thus reducing the subsequent apoptotic mechanism.
PurposeThe purpose of this study is to empirically examine the efficiency of Greek fixed-odds (offline) betting market as offered by OPAP for the period 2016–2019.Design/methodology/approachUsing a four-year data sample of OPAP's opening and closing odds for football matches from all over the world and applying linear probability and probit models, the market efficiency is examined and the existence of possible anomalies is investigated.FindingsThe main findings of research suggest that although the odds are dominated primarily by favorite-longshot bias and secondarily by draw bias, this mispricing cannot prove profitable. However, the opening odds, the margin levels and the market structure provide information that is not fully captured by the closing odds, giving bettors profit opportunities. Thus, findings show that the semi-strong market efficiency is questionable. Finally, competition reduces commissions leading to more efficient odds.Practical implicationsThe conclusions of this study are useful for football betting market and, particularly, for government authorities, bookmakers and bettors. Findings can be extended in future research to prediction tasks.Originality/valueTo the best of the authors’ knowledge, this is the first study about the Greek football betting market. The contribution to the literature lies on the one hand in the examination of a monopolistic land-based betting market, which is being squeezed and threatened by the more competitive online betting market, and on the other hand in the simultaneous examination of the opening and closing odds.
Ένας μεγάλος κύκλος φτάνει στο τέλος του! Ήταν Δεκέμβριος του 2003 (9 χρόνια πριν), όταν ο φίλος και συνάδελφος Γιώργος Μαρκόπουλος έχοντας ήδη ξεκινήσει πρακτική στο εργαστήριο Γενικής Βιολογίας, ήρθε ένα πρωινό λέγοντάς μου ότι ο μετέπειτα επιβλέπων καθηγητής μου κ. Αγγελίδης Χαράλαμπος ζητούσε κάποιο άτομο για πρακτική. Αμέσως ανταποκρίθηκα θετικά, δεδομένου του γεγονότος ότι αρέσκομαι ιδιαιτέρως σε τεχνικά και εργαστηριακά ζητήματα και ήρθα σε συννενόηση με τον κ. Αγγελίδη, οπότε ξεκίνησα πρακτική στο εργαστήριό του. Μετά την πρακτική, το 2005, ήρθε η ώρα της διπλωματικής μου εργασίας για το πτυχίο του τμήματος Βιολογικών Εφαρμογών & Τεχνολογιών στο οποίο φοιτούσα. Τελειώνοντας τη διπλωματική μου εργασία, ο κ. Αγγελίδης μου πρότεινε να συνεχίσουμε τη συνεργασία μας στα πλαίσια μιας διδακτορικής διατριβής. Χωρίς σκέψη, απάντησα θετικά. Έτσι, φτάνουμε στο 2012, εννέα χρόνια μετά και ήρθε η μεγάλη ώρα να υποστηρίξω την μακρόχρονη και κοπιώδη εργασία μου στα πλαίσια της διατριβής αυτής.Τα χρόνια αυτά στο εργαστήριο ήταν γεμάτα εμπειρίες, πότε ευχάριστες, πότε δυσάρεστες, αλλά όταν σε ευχαριστεί το περιβάλλον και αυτό που κάνεις, ξεχνάς τα πάντα και σου μένουν μόνο καλές αναμνήσεις. Επιπλέον, μέσα στα χρόνια αυτά, γνώρισα πολλούς συναδέλφους και έκανα αρκετές φιλίες. Δυστυχώς όμως, ολοκληρώνω τον κύκλο αυτό με μια πίκρα στο στόμα, δεδομένης της σημερινής κατάστασης του Πανεπιστημίου, το οποίο μετά λύπης βλέπω καθημερινά να βυθίζεται λόγω της σημερινής κατάστασης!
