Microvesicles are plasma membrane-derived vesicles released into the extracellular environment by a variety of cell types. Originally characterized from platelets, microvesicles are a normal constituent of human plasma, where they play an important role in maintaining hematostasis. Microvesicles have been shown to transfer proteins and RNA from cell to cell and they are also believed to play a role in intercellular communication. We characterized the RNA and protein content of embryonic stem cell microvesicles and show that they can be engineered to carry exogenously expressed mRNA and protein such as green fluorescent protein (GFP). We demonstrate that these engineered microvesicles dock and fuse with other embryonic stem cells, transferring their GFP. Additionally, we show that embryonic stem cells microvesicles contain abundant microRNA and that they can transfer a subset of microRNAs to mouse embryonic fibroblasts in vitro. Since microRNAs are short (21–24 nt), naturally occurring RNAs that regulate protein translation, our findings open up the intriguing possibility that stem cells can alter the expression of genes in neighboring cells by transferring microRNAs contained in microvesicles. Embryonic stem cell microvesicles may be useful therapeutic tools for transferring mRNA, microRNAs, protein, and siRNA to cells and may be important mediators of signaling within stem cell niches.

Na(+)-activated potassium channels (K(Na)) have been identified in cardiomyocytes and neurons where they may provide protection against ischemia. We now report that K(Na) is encoded by the rSlo2 gene (also called Slack), the mammalian ortholog of slo-2 in C. elegans. rSlo2, heterologously expressed, shares many properties of native K(Na) including activation by intracellular Na(+), high conductance, and prominent subconductance states. In addition to activation by Na(+), we report that rSLO-2 channels are cooperatively activated by intracellular Cl(-), similar to C. elegans SLO-2 channels. Since intracellular Na(+) and Cl(-) both rise in oxygen-deprived cells, coactivation may more effectively trigger the activity of rSLO-2 channels in ischemia. In C. elegans, mutational and physiological analysis revealed that the SLO-2 current is a major component of the delayed rectifier. We demonstrate in C. elegans that slo-2 mutants are hypersensitive to hypoxia, suggesting a conserved role for the slo-2 gene subfamily.

Potassium channels have evolved to play specialized roles in both excitable and inexcitable tissues. Here we describe the cloning and expression of Slo3, a novel potassium channel abundantly expressed in mammalian spermatocytes. Slo3 represents a new and unique type of potassium channel regulated by both intracellular pH and membrane voltage. Reverse transcription-polymerase chain reaction, Northern analysis, and in situ hybridization show that Slo3 is primarily expressed in testis in both mouse and human. Because of its sensitivity to both pH and voltage, Slo3 could be involved in sperm capacitation and/or the acrosome reaction, essential steps in fertilization where changes in both intracellular pH and membrane potential are known to occur. The protein sequence of mSlo3 (the mouse Slo3 homologue) is similar to Slo1, the large conductance, calcium-and voltage-gated potassium channel. These results suggest that Slo channels comprise a multigene family, defined by a combination of sensitivity to voltage and a variety of intracellular factors. Northern analysis from human testis indicates that a Slo3 homologue is present in humans and conserved with regard to sequence, transcript size, and tissue distribution. Because of its high testisspecific expression, pharmacological agents that target human Slo3 channels may be useful in both the study of fertilization as well as in the control or enhancement of fertility.

Both intracellular calcium and voltage activate Slo1, a high-conductance potassium channel, linking calcium with electrical excitability. Using molecular techniques, we created a calcium-insensitive variant of this channel gated by voltage alone. Calcium sensitivity was restored by adding back small portions of the carboxyl (C)-terminal 'tail' domain. Two separate regions of the tail independently conferred different degrees of calcium sensitivity; together, they restored essentially wild-type calcium dependence. These results suggest that, in the absence of calcium, the Slo1 tail inhibits voltage-dependent gating, and that calcium removes this inhibition. Slo1 may have evolved from an ancestral voltage-sensitive potassium channel represented by the core; the tail may represent the more recent addition of a calcium-dependent modulatory domain.

The gating of different potassium channels depends on many diverse factors. We now report a unique example of a K+ channel with a Cl - dependence. The slo-2 gene was cloned from Caenorhabditis elegans and is widely expressed in both neurons and muscles; it was highly abundant, as suggested by its high representation in the C. elegans EST database. SLO-2, like its paralogue, SLO-1, was also dependent on Ca 2+. We show by site-directed mutagenesis that its requirements for both Cl- and Ca2+ are synergistic and associated with the same functional domain. SLO-2's dependence on Cl - implies that intracellular Cl- homeostasis may be important in regulating cellular excitability through this unusual K+ channel.

Submacular choroidal thickness, as measured by SD-OCT EDI, is significantly reduced in patients with RP, but did not correlate with visual acuity or retinal thickness in RP patients. Further research is needed to understand better  the pathophysiological significance of the choroidal alterations present in RP.

PURPOSE. To demonstrate a novel algorithm for macular hole (MH) segmentation and volumetric analysis. METHODS.A computer algorithm was developed for automated MH segmentation in spectral-domain optical coherence tomography (SD-OCT). Algorithm validation was performed by trained graders with performance characterized by absolute accuracy and intraclass correlation coefficient. A retrospective case series of 56 eyes of 55 patients with idiopathic MHs analyzed using the custom algorithm to measure MH volume, base area/diameter, top area/diameter, minimum diameter, and height-to-base diameter ratio. Five eyes were excluded due to poor signal quality (1), motion artifact (1), and failure of surgical closure (3) for a final cohort of 51 eyes. Preoperative MH measurements were correlated with clinical MH stage, baseline, and 6-month postoperative best-corrected Snellen visual acuity (BCVA).RESULTS. The algorithm achieved 96% absolute accuracy and an intraclass correlation of 0.994 compared to trained graders. In univariate analysis, MH volume, base area, base diameter, top area, top diameter, minimum diameter, and MH height were significantly correlated to baseline BCVA (P value from 0.0003-0.011). Volume, base area, base diameter, and height-to-base diameter ratio were significantly correlated to 6-month postoperative BCVA (P value from <0.0001-0.029). In multivariate analysis, only base area (P < 0.0001) and volume (P ¼ 0.0028) were significant predictors of 6-month postoperative BCVA.CONCLUSIONS. The computerized segmentation algorithm enables rapid volumetric analysis of MH geometry and correlates with baseline and postoperative visual function. The DISCOVER iOCT study demonstrated both generalized feasibility and usefulness based on the surgeon-reported impact on surgical decision making. This large-scale study confirmed similar findings from other studies on the potential value and impact of iOCT on ophthalmic surgery.
