Use of the crustacean Artemia franciscana for alternative biotests of the second generation was studied, and possible experimental design and applications of such tests outlined. In addition to the classical use in ecotoxicology, the test can be used in pharmacology as well, or to monitor the effects of ionizing radiation in co-exposure with some chemical compounds. The synergistic effect of co-exposure of PCB (DELOR 103), cadmium chloride and potassium dichromate with beta 89 Sr irradiation was shown. We also demonstrated the anti-oxidative and pro-oxidative effects of the ascorbic acid in dependence on its concentration. Use of the pharmaco-toxicological screening in search for the novel inhibitors of cyclin-dependent kinases was demonstrated as well, showing that Artemia franciscana may be used as a suitable biosensor instead of the expensive tests on higher vertebrates.
Cíl: Zhodnotit vliv saunování a celotělové kryoterapie na 24hodinový profi l krevního tlaku a srdeční frekvence a porovnat jejich hodnoty před působením tepla nebo chladu a bezprostředně po jejich působení. Metodika: Bylo provedeno sedmidenní ambulantní monitorování krevního tlaku (TK) a srdeční frekvence (SF). Celkově bylo sledováno 20 dobrovolníků (deset mužů a deset žen, průměrný věk 24,60 ± 1,93 roku). Průměrná hmotnost probandů byla 70,45 ± 9,90 kg, výška 1,74 ± 7,71 m, průměrná hodnota indexu tělesné hmotnosti (BMI) činila 23,07 ± 1,66 kg/m 2. Všichni probandi byli nekuřáci. Každý proband absolvoval během jednoho dne dvě saunové procedury ve fi nské sauně a s odstupem minimálně 48 hodin absolvoval jednorázový vstup do celotělové kryokomory. TK a SF byly změřeny vždy před vstupem do sauny nebo do kryokomory a ihned po jejich opuštění. V rámci sedmidenního ambulantního monitorování byly sledovány hodnoty 0-24 hodin a 24-48 hodin po působení fyzikální terapie a tyto naměřené hodnoty byly mezi sebou vzájemně srovnány. Výsledky: Průměrná sedmidenní hodnota TK byla u 20 dobrovolníků 122 ± 1,21/71 ± 0.82 mm Hg a průměrná sedmidenní hodnota SF byla 73 ± 1,12/min. Po sauně byl prokázán statisticky významný pokles systolického krevního tlaku (STK) o 9 mm Hg a diastolického krevního tlaku (DTK) o 13 mm Hg. Po kryoterapii byl prokázán statisticky významný vzestup systolického krevního tlaku (STK) o 8 mm Hg a pokles srdeční frekvence (SF) o 8/min. Závěry: Studie potvrzuje vliv saunování a kryoterapie na kardiovaskulární aparát zdravých jedinců. Bezprostředně po opuštění sauny se STK a DTK snižuje a SF se zvyšuje. Všechny parametry se v dalších 48 hodinách vracejí ke svým standardním klidovým hodnotám. Bezprostředně po vystavení se chladu v kryokomoře se STK i DTK zvyšují a SF klesá. Všechny parametry se v dalších 48 hodinách vracejí ke svým původním klidovým hodnotám.
Objective: Poor health outcomes associated with shift work include increased risks of diabetes mellitus, dyslipidemia, hypertension, heart disease, peptic ulcer disease, depression and cancer. The aim of the study was to compare the 7-day/24-h blood pressure ambulatory monitoring in healthy subjects and nurses working in day and night work shifts. Design and method: We examined 297 healthy subjects and 6 women (age 33 ± 2 years, body weight 70 ± 21 kg, mean height 165 ± 5 cm) and 4 men (age 28 ± 7 years, body weight 93 ± 11 kg, mean height 185 ± 5 cm), in days with day work, days with night work and free days, using Medical Instruments TM2431 (A and D, Japan). Results: The mean (±SD) of sleep hours in the week in 10 nurses during 7-day/24-h blood pressure monitoring was 37.75 ± 2.75 h per week. The mean of 7-day/24-h ambulatory blood pressure monitoring was SBP 126 ± 7.6 mmHg, of DBP 76 ± 5 mmHg, of HR 74 ± 5 bpm. Contrary to healthy subjects, in all shift workers we have found the disturbences in circadian rhythm according the day and night shifts. The seven day/24-hour systolic blood pressure profiles vary from 110 to 140 mmHg and we can not use reference values for 24-h ambulatory blood pressure monitoring, while there is impairment in circadian rhythm because of irregular night work. The similar condition is valid for diastolic blood pressure. Longitudinal monitoring could be implemented more generally to assess inter-individual responds to shift work. Conclusions: Seven day/24-h ambulatory blood pressure monitoring in night shift workers showed impairment of circadian rhythm and BP vary according to the work depending on different working shifts, and decrease of sleep hours. Further work is also needed to determine whether there may be shift work schedules that are less detrimental than others.
