Steroidalkohole verschiedener Struktur, deren Hydroxyfunktionen aus elektronischen und sterischen Griinden in ihrer Reaktivitat differieren und die dariiber hinaus empfindliche Gruppierungen enthalten, werden mit 2,3,4,6-Tetra-0-p~va~oyl-a-~-glucopyranosy~bromid (1) selektiv und effektiv in P-Glucoside iibergefiihrt. Dank des lenkenden Einflusses des 2-0-Pivaloyl-Substituenten wird eine Orthoesterbildung bei den Koenigs-Knorr-Reaktionen stark unterdriickt. Mit dem o-Toluoylrest als Hydroxyschutzgruppe wird diese Lenkung nur in geringem MaR erreicht.
Stereoselective Glycosylation of Steroid Alcohols Using 2,3,4,6-Tetra-O-pivaloy1-u-~-glucopyranosyl Bromide (Pivalobromoglucose) and 2,3,4,6-Tetra-O-(o-toluoyl)-u-~-glucopyranosyl BromideStructurally diverse steroid alcohols possessing hydroxylic functions of electronically and sterically different reactivity and, furthermore, including sensitive groups, are effectively and selectively converted into their P-glucosides using 2,3,4,6-tetra-O-piva~oy~-a-~-g~ucopyra-nosy1 bromide (1). The formation of orthoesters is suppressed in these Koenigs-Knorr type reactions due to the directing influence of the 2-pivaloyl substituent. Fur die meisten publizierten Steroidglycosidsynthesen wurde 2,3,4,6-Tetra-O-acetyl-a-~-glycopyranosylbromid (Acetobromglucose) (2) verwendet, wobei die Ausbeuten selten 50% iiber~teigen*~~-~). Ein Grund fur die unbefriedigenden Ausbeuten liegt in der Bildung unerwiinschter Orthoester, die bei den sterisch anspruchsvollen Steroidaglyconen uber die Acyloxonium-Ionen9) begiinstigt entstehen und bei der Aufarbeitung haufig durch Hydrolyse verloren gehen.Durch Verwendung des Pivaloyl-Restes anstelle der Acetylgruppe als Hydroxyschutzgruppe im Glycosylbromid haben wir eine Lenkung der Glycosidsynthese zum gewunschten 1,2-trans-Glycosid erreichen konnen, wobei die Bildung des
