La antracnosis es la enfermedad más limitante en cultivos de mango en el departamento del Magdalena, Colombia, pues causa pérdidas de 40 a 50 % de la cosecha. A nivel mundial, se registran pérdidas en campo hasta de 51 % y en poscosecha, de 20 a 30 %. El agente causal, Colletotrichum spp., es un hemibiótrofo que inicialmente no produce síntomas en su hospedante, pero después se torna necrotrófico. El endofitismo es un estilo de vida del hongo cuyo papel en la patogenia es aún incierto. En este contexto, se propuso la caracterización de aislamientos endofíticos de Colletotrichum spp. a partir de hojas y ramas de mango cultivar Azúcar de un cultivo ubicado en Ciénaga. Se determinó la prevalencia de los aislamientos en hojas y ramas de tres estratos del árbol mediante tres protocolos de extracción: 1) en cámara de cloro, 2) con hipoclorito de sodio al 4 %, y 3) con hipoclorito de sodio al 2 %. Se describió la morfometría (colonia, conidios y apresorios), se hizo la prueba de patogenia en los frutos y la identificación molecular del aislamiento más virulento. Se obtuvieron nueve cepas endofíticas, ocho de hojas y una de ramas. La mejor extracción (18,2 %) se logró con el hipoclorito de sodio al 2 %; la prevalencia fue mayor en el estrato superior (10 % de las muestras) y en las hojas (8,9 % de las muestras). Las cepas endofíticas indujeron lesiones de 0,65 a 19,7 mm de diámetro y la más virulenta (HP3A-4) se identificó molecularmente como C. tropicale, siendo este el primer registro en Colombia del hongo como endófito patogénico en el mango. Se concluyó que es posible encontrar Colletotrichum como endófito en hojas y ramas de mango y que puede causar infección al inocularse en los frutos, evidenciando así una gran probabilidad de que actúe como un inóculo oculto para el desarrollo de la antracnosis en el mango. © 2019. Colletotrichum, a genus of Ascomycota fungi with a diversity of species grouped into several species complexes, or clades, is associated with diseases known as "Anthracnose". It affects significantly different tropical and subtropical fruit species. Infections occur in the field and postharvest; quiescent stages are mainly responsible for postharvest losses. An analysis of this pre-infective stage is made based on consulted papers. The infective process is modulated by the length of the quiescent period of the fungus during the vegetative or pre-productive stages of trees. Quiescent stage is determined by the host's biochemical responses and the pathogen's activity. Once the pathogen is activated, it develops an infectious necrotrophic process. Colletotrichum quiescence has been studied mainly in subtropical fruits and horticultural species, and the studies highlight the role of volatile compounds, metabolites and enzymes involved in the length and breaking of quiescence, as well as the differential responses according to the phenological stage and the genotype of the plant. Transcriptomic and proteomic analysis of the host-fungus interaction have revealed the role of genes in the occurrence and breaking of quiescence. Knowledge about the quiescence of Colletotrichum in tropical fruits is necessary to improve management efficiency. Detection and induction of quiescent infections has been studied and genomics has allowed to understand the occurrence of quiescence in the infective process; however, in crops in the tropics, such as mango, an important crop in Colombia, questions about associated species and biology of their quiescence are still unanswered.
La antracnosis, asociada al género fitopatógeno Colletotrichum, es una de las enfermedades más limitantes de mango en el mundo al generar daños en campo y poscosecha. El patógeno es hemibiótrofo y además puede ser endófito asintomático; asimismo, algunas especies presentan quiescencia. El papel de estos estados biológicos como fuente de inóculo es aún incierto. Se evaluó la patogenicidad de 14 aislamientos endófitos-quiescentes de Colletotrichum obtenidos de estudios previos y de un aislamiento extraído de un fruto sintomático de mango cv. Azúcar. Al describir morfométricamente (colonia) los aislamientos en cultivos monospóricos y compararlos por su tasa de crecimiento, densidad de inóculo, conidias y apresorios, se encontraron diferencias estadísticas para las primeras tres variables. Asimismo, se evaluó la patogenicidad de los morfotipos en hojas y ramas de mango cv. Azúcar mediante inoculaciones con atomizador en árboles de dos años en condiciones de trópico seco, se describieron síntomas y se evaluaron variables biológicas (número, diámetro y tasa de crecimiento de lesiones). En hojas, todos los morfotipos indujeron lesiones, pero solo hubo diferencias en la tasa de crecimiento; en ramas, se observaron diferencias altamente significativas entre morfotipos en el número y diámetro de lesiones, y diferencias en la tasa de crecimiento de estas, aunque hubo morfotipos que no indujeron daños. Hay alta variabilidad morfológica y biológica entre morfotipos de Colletotrichum, y los aislamientos endofíticos son potencialmente infectivos sobre hojas y ramas de mango cv. Azúcar.
Epidemiological analyzes of foliar diseases associated with Colletotrichum spp. in Enterolobium cyclocarpum and Platymiscium pinnatum were performed under field conditions and without any type of intervention. At the Universidad del Magdalena (Santa Marta, Colombia), four trees for each species and four equidistant monitoring sites per tree were established. The incidence and severity were recorded for 33 weeks (March to November 2016), including two follow-up periods: dry and rainy season. Disease development curves were elaborated. Moreover, the development rate (r) and the area under the disease progress curve (AUDPC) were calculated for each follow-up period. The effect of the meteorological variables was statistically analyzed by correlation and multiple regression. In E. cyclocarpum, the highest incidence and severity were recorded between September and November with 100 and 19.6%, respectively, showing a positive correlation with relative humidity and negative with average temperature, solar radiation and wind speed. In P. pinnatum, the maximum values of incidence and severity were observed between March and April with 68.9 and 1.3%, respectively. However, correlation analyzes did not support their relationship with the environmental factors. The r values during the dry months were 0.136 and 0.107 units week-1 and the AUDPCs were calculated at 51 and 4 units week-1 for E. cyclocarpum and P. pinnatum, respectively. In the rainy months, the r values were 0.187 and 0.016 units week-1 and the AUDPCs were 186 and 2 units week-1, respectively. In conclusion, the development of the disease varies according to the forest species, time of year and some meteorological variables.
