The occurrence of multiple introduction events, or sudden emergence from a host jump, of forest pathogens may be an important factor in successful establishment in a novel environment or on a new host; however, few studies have focused on the introduction and emergence of fungal pathogens in forest ecosystems. While Ophiognomonia clavigignenti-juglandacearum (Oc-j), the butternut canker fungus, has caused range-wide mortality of butternut trees in North America since its first observation in 1967, the history of its emergence and spread across the United States and Canada remains unresolved. Using 17 single nucleotide polymorphic loci, we investigated the genetic population structure of 101 isolates of Oc-j from across North America. Clustering analysis revealed that the Oc-j population in North America is made up of three differentiated genetic clusters of isolates, and these genetic clusters were found to have a strong clonal structure. These results, in combination with the geographic distribution of the populations, suggest that Oc-j was introduced or has emerged in North America on more than one occasion, and these clonal lineages have since proliferated across much of the range of butternut. No evidence of genetic recombination was observed in the linkage analysis, and conservation of the distinct genetic clusters in regions where isolates from two or more genetic clusters are present, would indicate a very minimal or non-existent role of sexual recombination in populations of Oc-j in North America.
Antecedentes y Objetivos: Los modelos de nicho ecológico se han usado para entender de qué manera las especies pueden cambiar o persistir en un espacio ambiental y geográfico a lo largo del tiempo. Los objetivos del presente estudio fueron i) modelar el nicho ecológico de P. montezumae y P. pseudostrobus en México, ii) evaluar y comparar la similaridad y equivalencia de la idoneidad del nicho entre las áreas de distribución de las dos especies, y iii) proponer zonas geográficas potenciales para la delimitación de áreas productoras de semillas y conservación para cada especie.Métodos: Se usaron datos geográficos para predecir la distribución de especies y climáticos topográficos y edáficos como predictores ambientales, fueron modelados con base en el algoritmo de Máxima Entropía. Se estimaron parámetros de similaridad (I) y equivalencia (D) para conocer el grado de traslape del nicho. Para la selección de áreas semilleras se usaron los datos ambientales que definieron la distribución de cada especie y dasométricos del Inventario Nacional Forestal y de Suelos (INFyS).Resultados clave: Los modelos muestran que las dos especies presentan zonas muy similares de alta idoneidad, distribuidas en la Faja Volcánica Transmexicana y en la Sierra Madre del Sur. Las mayores contribuciones al modelo de Pinus pseudostrobus fueron las variables temperatura promedio anual (41.3%) y precipitación total anual (14.3%), mientras que el suelo (31.1%) y la altitud (29%) fueron determinantes para el modelo de P. montezumae. El traslape de nicho presenta un valor observado de equivalencia (D=0.430) menor al de similaridad (I=0.610), sugiriendo que las especies tienen un nicho similar, pero que este no es equivalente en su totalidad, el cual podría estar en proceso de diversificación.Conclusiones: La idoneidad del nicho ecológico es similar para las dos especies de pino, pero no es equivalente al presentar variables ambientales diferenciales. Estos resultados tienen implicaciones para la selección potencial de áreas productoras de semillas y conservación para cada especie.
Métodos: El estudio se realizó en siete poblaciones representativas de su distribución natural en México, con muestras de acículas de 112 árboles analizadas con cinco microsatélites de núcleo. Los datos se evaluaron con parámetros de la genética de poblaciones y métodos de aproximación Bayesiana. Resultados clave: La heterocigosidad promedio esperada (He=0.557) fue más alta que la observada (Ho=0.375). Las poblaciones no se encuentran en equilibro Hardy-Weinberg, con una endogamia significativa (FIS=0.259) y tamaños efectivos reducidos (Ne entre 375-425); están estructuradas en tres grupos genéticos (FST=0.158), con valores altos del índice Mc (0.186-0.283), sugiriendo eventos de declinamiento demográfico ancestral (entre 11,910 y 23,820 años atrás), asociados con los cambios climáticos del Pleistoceno. Conclusiones: El estudio demuestra que P. remota contiene una variación genética considerable, pero estructurada espacialmente y asociada a cuellos de botella ancestrales. Además, se confirma que P. catarinae es su sinónimo. La especie ha sido capaz de subsistir y adaptarse a condiciones ambientales locales. Con este conocimiento se plantean estrategias para la conservación de las poblaciones remanentes de la especie.
La conservación de los hábitats terrestres asociados a cuencas hidrográficas tiene como componente intrínseco la conservación de los recursos genéticos de las especies que conforman la cubierta vegetal de estos sistemas. El objetivo del presente estudio fue evaluar los niveles de variación y estructura genética de las poblaciones de pino distribuidas en la cuenca del río Cupatitzio, en Michoacán, con el uso de cuatro microsatélites de núcleo. El estudio incluye ocho poblaciones de cuatro especies de pino distribuidas en las zonas altas, media y baja de la cuenca. Los resultados indican que las poblaciones SL3 de P. pseudostrobus y RB6 de P. douglasiana contienen los valores más altos de variación genética (HE =0.674 y HE= 0.615, respectivamente). Las poblaciones presentan importantes niveles de endogamia (FIS entre 0.057-0.544) y una diferenciación genética significativa (FST entre 0.094-0.152), la cual se asocia de manera moderada con la distribución geográfica de las poblaciones (r= 0.443) y se agrupa de acuerdo con las especies. La parte alta de la Reserva, en Quinceo y la Tzaráracua, presenta los niveles más bajos de variación genética y los mayores niveles de endogamia, por lo que se recomienda hacer actividades de restauración en estas localidades. Así mismo, se sugieren actividades de conservación in situ en San Lorenzo y la parte baja de la Reserva, ya que ambas poblaciones presentan los tamaños efectivos más grandes y son representativas del acervo genético de los bosques de pino en la cuenca del Cupatitzio.
