A genetic and physical map has been developed for the loculoascomycete Leptosphaeria maculans, a pathogen of oilseed Brassicas. The genetic map was constructed from 58 F(1) progeny and comprises 155 amplified fragment length polymorphic (AFLP) markers, three random amplified polymorphic DNA (RAPD) markers, the mating type locus and a host specificity locus conferring the ability to form lesions on Brassica juncea. Twenty-one linkage groups, 5 pairs, and 18 unlinked markers were assigned, and the genome size was 1520 cM. Pulsed field gel electrophoresis experiments showed that the parental isolates each had 16 chromosomes and a genome size of about 33.5 Mb. Attempts to anchor a large number of markers to chromosomes were hampered by difficulties in converting AFLPs into RFLP markers, and because many markers bound to every chromosome, indicating that L. maculans has a high level of dispersed repetitive sequences. This fungus displays chromosomal length polymorphisms, but in the cross examined, the linkage and physical maps were essentially congruent and there was no evidence of translocations. The host specificity locus is 18 cM from the nearest AFLP marker and is located on a chromosome sized 1.85 Mb in the virulent parent. The mating type locus is on a chromosome sized 2.6 Mb and coincident on an AFLP marker amplified from the virulent parent. The derived amino acid sequence of part of this marker has some conserved amino acids present in the High Mobility Group DNA binding domain of MAT-2 mating type genes of other ascomycetes.
SUMMARYReduced soil fertility and damage from pests and diseases have contributed to a decline in productivity of cocoa (Theobroma cacao L.) smallholdings in Sulawesi, Indonesia over the last decade. In a trial on a marginal, acidic soil in South Sulawesi, young PBC123 cocoa trees were supplied with compost, mineral fertiliser (NPK fertiliser and urea) or dolomite, alone and in combination. After 20 months, the trees supplied with compost were taller, flowered more profusely and had a five-fold higher dry bean yield than other treatments. Treatments had no impact on incidence of cocoa pod borer, Phytophthora pod rot and vascular streak dieback. All of the trees supplied with compost survived, while the control, mineral-fertiliser- and dolomite-treated trees had a mortality rate of 22–45% and symptoms of interveinal necrosis. Leaf concentrations of N, P and K were within the normal range in all treatments. In the control and mineral fertiliser treatments, leaf concentrations of Ca (0.28–0.30%) and Mg (0.11–0.15%) were deficient, but were higher in trees supplied with compost (0.78–1.21% and 0.26–0.29%, respectively). The Mg/K ratio in soil-exchangeable cations and leaves was increased three-fold by the combined compost/dolomite treatment. Supplying mineral fertiliser alone resulted in 3.3 cmol kg−1 exchangeable Al, compared to 2.2 cmol kg−1 in control soils. Since 10 kg tree−1 year−1 compost was supplied, a rate that is not practical on most cocoa smallholder farms, further investigation of cost-effective applications of organic matter in conjunction with appropriate formulations of inorganic fertilisers is recommended.
A range of Brassica species was screened for resistance to Leptosphaeria maculans, the causal agent of blackleg. The lines were assessed in 8 disease nurseries in 4 canola growing regions of Australia and in 1�glasshouse trial, with a view to identifying alternative sources of resistance to L. maculans for Australian breeding programs. Lines were screened for degree of internal and external blackleg symptoms during both the seedling and adult plant growth stages. Correlation for resistance with ranking between disease nurseries was very strong (0.41-0.98). Brassica carinata and B. nigra were the most resistant species in the disease nurseries, being even more resistant than B. juncea. The 7 European winter B. napus lines tested were significantly more resistant than the 7�Australian spring B. napus lines, with another crucifer, Sinapis alba, being intermediate in resistance between the European and Australian B. napus lines. The same ranking of lines from most to least resistant was also seen when cotyledons and stems were inoculated in the glasshouse with 2 well-characterised Australian isolates. With the exception of the B. napus susceptible control Westar, all lines had similar frequencies of seedling survival in the nurseries. However, mature plants of these lines varied significantly in their degree of resistance. This indicates that screening for seedling survival is not useful in selecting L. maculans resistant lines in Australia. The Brassica lines with the B genome, especially B. carinata, and the winter B. napus types are now being used as sources of resistance in Australian breeding programs.
