Aims: To develop a pilot-plant fermentation process for the production of the yeast Rhodotorula minuta, to be used as a biocontrol agent of mango anthracnose, using a low-cost culture medium. To develop a stable liquid formulation that preserve high viability of the yeast stored at 4°C. Methods and Results: Keeping constant the volumetric power input, a fermentation process was scaled-up from shake flasks to a 100 l bioreactor. Preharvest applications of the yeast resulted in postharvest anthracnose severity equal or lower than that observed with a chemical fungicide. Glycerol was added to the formulation as water activity reducer and xanthan gum as a viscosity-enhancing agent. Yeast initial concentration of 10 10 CFU mlresulted in 4-5 orders of magnitude decrease after 1 month of storage at 4°C, whereas when it was formulated at 10 9 CFU ml )1 , the decrease was of two orders of magnitude in 6 months. Conclusions:The fermentation process was successfully scaled-up using a low-cost culture medium. Postharvest anthracnose severity could be considerably reduced using this yeast. Formulating the yeast at 10 9 CFU ml )1and adding glycerol (20%) and xanthan (5 g l )1 ) avoided both contamination and yeast sedimentation and it was able to preserve up to 10 7 CFU ml )1 after 6 months at 4°C.Significance and Impact of the Study: The yeast R. minuta is reported as a novel antagonistic micro-organism against the pathogen Colletotrichum gloeosporioides. Pilot plant production of this yeast allowed us to conduct field tests in commercial orchards during three harvest seasons. Yeast suspensions applied to mango trees reduced the fruit anthracnose severity in levels similar or better than chemical fungicides.
En Poaceae, la semilla o propágulo comercial consiste de espiguilla o diáspora: cariópside (Cs) + brácteas accesorias: normalmente glumas, lemas, paleas y, en ocasiones, específico de especie, ramas modificadas, las cuales tienen efecto sobre la germinación, influyen el manejo requerido y por tanto la calidad biológica de la semilla. La eliminación de brácteas accesorias con o sin clasificación de Cs por tamaño es una alternativa no documentada en el establecimiento de praderas de temporal. El objetivo del estudio fue evaluar la germinación sembrando cada 30 días, Cs clasificadas de pastos, dos nativos y dos introducidos, obtenidas a partir de diásporas comerciales al inicio del experimento (Cs base -CsB) o cada 60 días (Cs testigo-CsT), durante 15 meses; similarmente, evaluar la emergencia de plántulas a partir de diásporas. Los propágulos (diásporas o Cs) se evaluaron de febrero 2010 a mayo 2011, en laboratorio. Las especies nativas e introducidas incluyeron Bouteloua gracilis, B. curtipendula, Cenchrus ciliaris y Chloris gayana, respectivamente. A partir de diásporas, se eliminaron brácteas accesorias y las Cs resultantes, se separaron con tamices, en grandes (CsG), medianos (CsM) y chicos (CsCh). Se sembraron, en base a semilla pura viable, tanto los diferentes tamaños de Cs, sobre papel húmedo, como diásporas, a 1cm de profundidad, en peat moss. El comportamiento de la germinación se analizó mediante regresión logística con la rutina Logistic de SAS. En las especies evaluadas, mayor tamaño de Cs se relacionó con mayor germinación y las covariables beneficio y tiempo de almacenamiento fueron significativas (p< 0.001). La germinación en pastos nativos tendió a disminuir con el tiempo, contrariamente a introducidos y la germinación fue mayor en relación directa con el tamaño de la Cs. La viabilidad se conserva mejor manera al mantener la Cs con brácteas accesorias; pero, en especies introducidas eliminar éstas, incrementó la germinación.
El análisis de crecimiento es una aproximación cuantitativa de descripción e interpretación del desarrollo vegetal, determinado por condiciones genético-ambientales. El objetivo fue conocer el desarrollo en 4 cultivares de sorgo forrajero (caña dulce, Silo miel, Esmeralda y Fortuna) mediante a acumulación de biomasa (BT), altura de planta (AP), nudos por planta (NNP), índice de área foliar (IAF), tasa de crecimiento del cultivo (TCC) y tasa de asimilación neta (TAN), con el fin de determinar la producción y momento óptimo de corte, bajo condiciones de secano en el Altiplano Central, Colegio de Postgraduados, Campus Montecillo, Texcoco, Estado de México, México. El diseño experimental fue bloques al azar en parcelas divididas, con arreglo factorial. Por muestreo, se tomó una planta de cada cultivar con 4 repeticiones y se midió BT, AP, NNP y IAF a 115 días después de la siembra (dds). La caña dulce presentó mayor AP (202 cm), IAF (3,8) y BT (14,35 t.ha-1); en contraste, Fortuna presentó menor AP (90 cm), BT (3,96 t.ha-1) e IAF (2,1). La caña dulce en el periodo 25-55 dds, presentó mayores TCC (24,2 gm-2d-1) y TAN (10,2 gm-2d-1); por su parte Fortuna presentó menor TCC (5 gm-2d-1) y TAN (5,51 gm-2d- 1). La BT se correlacionó positivamente con AP, IAF, TCC (p≤0,001) y TAN, con TCC (p≤0,001). El mejor cultivar para producción de forraje y condiciones del estudio fue Caña dulce; lo anterior, debido a que presentó los mejores parámetros a lo largo del ciclo del cultivo para AP, NNP, BT, IAF, TCC y TAN. La máxima acumulación de biomasa se observó a 115 dds y coincide con la etapa fenológica reproductiva en inicio de floración, con plantas de 200 cm de longitud; posteriormente, tanto TAN (0,1 gm-2d-1) como el IAF (3,9), TCC (0,3 gm-2d-1) disminuyeron considerablemente; por tanto, el momento óptimo de corte es a 115 dds en el cultivar: Caña dulce.
