Pros Sem Nas Masy Biodiv Indon 1: 413-417. Mastitis subklinis merupakan penyakit radang ambing yang paling banyak menimbulkan dampak ekonomi cukup besar di kalangan peternak sapi perah. Hal tersebut dikarenakan menurunnya kuantitas dan kualitas susu. Penyebab paling umum dari peradangan ambing tersebut di antaranya mikroorganisme, seperti E. coli, Streptococcus agalactiae dan Staphylococcus aureus. Penelitian ini bertujuan untuk mengisolasi dan mengidentifikasi bakteri Staphylococcus aureus sebagai salah satu bakteri patogen yang sering dijumpai pada kejadian mastitis. Pengambilan sampel susu dilakukan secara acak (random sampling) dengan terlebih dahulu melakukan California Mastitis Test (CMT). Sampel susu yang positif mastitis subklinis kemudian diisolasi menggunakan media Manitol Salt Phenol Red Agar (MSPRA). Selanjutnya dari isolat bakteri dengan koloni berwarna kuning yang mampu memfermentasi phenol red menjadi warna kuning, dilakukan pewarnaan Gram. Tampilan mikroskopik terlihat sel berbentuk bulat dan bergerombol tersebut selanjutnya dilakukan kultur bakteri menggunakan Nutrient Broth (NB) selama 24 jam. Hasil kultur kemudian diekstraksi untuk mendapatkan DNA bakteri dan dilakukan analisis PCR. Hasil menunjukkan bahwa enam isolat dari sepuluh sampel susu sapi mastitis merupakan Staphylococcus aureus. Subclinical mastitis is an inflammatory disease of the udder as the most considerable economic impact among dairy farmers, due to a decrease in the quantity and quality of milk. The most common cause of the udder inflammation includes microorganisms, such as E. coli, Streptococcus agalactiae and Staphylococcus aureus. The aim of this study was to isolate and identify the bacteria Staphylococcus aureus as one of the pathogenic bacteria that were often found in the incidence of mastitis. Milk sampling was randomly conducted by doing the California Mastitis Test (CMT). A positive sample of the subclinical mastitis milk then isolated using the Mannitol Salt Phenol Red Agar (MSPRA) media. Furthermore, from bacterial isolates with yellow colonies able to ferment phenol red into yellow, the Gram staining was conducted. Microscopic appearance looks round and clustered cells were then carried out using a bacterial culture Nutrient Broth (NB) for 24 hours. Results culture was then extracted to obtain bacterial DNA and PCR analysis. The results showed that the isolates from ten samples of cattle's were Staphylococcus aureus.
