Aphelenchoides fragariae çilek bitkisinin en önemli bir zararlısı ve çilek fidesi üretiminde de en sık karşılaşılan sorunların başında gelmektedir. 2015 yılında Adana'da saksılı çilek satışı için üretim yapan seradan alınan örnekte nematod analizi yapılmıştır. Çilek yaprakları küçük parçalara ayrılarak Geliştirilmiş Baermann huni yöntemine göre nematodlar elde edilmiştir. Elde edilen nematodların dişi ve larvaların morfolojik analizine göre A. fragariae olduğu tespit edilmiştir. Bu nematodun yapraklarda suya batırılmış gibi bir simptom oluşturmuş olduğu gözlenmiştir. Akdeniz Bölgesi'nde açıkta yetişen çilek alanlarında damlama sulama sisteminin kullanılması ve düzenli yağışın olmaması ilgili nematod için ciddi bir sorun teşkil etmemektedir. Ancak, özellikle çatı yüksekliği standartlara uygun olmayan (engin olan) ve havalandırma sisteminin yetersiz olduğu sera alanlarında çilek bitkisinin yapraklarında bulunan nem tabakası nedeniyle bu zararlının potansiyel olarak sorun oluşturma olasılığı bulunmaktadır.

It is known that the nematode belong to Meloidogyne genus causes significantly crop losses in vegetables and cereals. The root-knot nematodes, M. artiellia causes damages especially cereals, legumes and cruciferous crops. Isolates of M. artiellia recovered from root and soil samples collected from wheat fields in 2012 growing season were firstly identified by using morphological and molecular aspects in Hatay province of Turkey. M. artiellia was morphologically identified by using morphological features and allometric criteria of second stage juvenile and female. Also, it was molecularly identified by using the sequences of partial mitochondrial DNA cytochrome C oxidase subunit (COI) gene region and clearly separated from other species.Key words: Meloidogyne artiellia, morphology, nucleotide polymorphism, wheat.
ÖzetKök-ur nematodları (Meloidogyne spp.) dünyada ekonomik önem sahip bitki paraziti nematodları arasındadır. Meloidogyne cinsine ait nematod türleri sebzelerde ve tahıllarda önemli oranda ürün kayıplarına neden oldukları bilinmektedir. Kök ur nematodu, M. artiellia özellikle tahıllar, baklagiller ve cruciferae familyasına ait ürünlerde zarara neden olabilmektedir. Bu çalışmadaki M. artiellia izolatları Türkiye'de ilk defa Hatay ili buğday ekim alanlarından 2012 yılı üretim sezonunda toplanan toprak ve kök örneklerinden elde edilmiş olup morfolojik ve moleküler olarak tanımlamaları yapılmıştır. M. artiellia, ikinci dönem larva ve dişi bireylere ait morfolojik özellikler ve allometrik kriterler esas alınarak tanımlanmıştır. Ayrıca, mitokondriyal DNA'nın sitokrom C oksidaz alt ünitesine (COI) ait nükleotid dizilimleri kullanılarak moleküler olarak da tanımlanmış ve diğer kök ur nematodu türlerinden bariz olarak ayrıldığı saptanmıştır.Anahtar sözcükler: Meloidogyne artiellia, morfoloji, nükleotid polimorfizmi, buğday.
Strawberry is one of the most economically important crops worldwide. Several species of plant-parasitic nematodes have been reported to be pathogenic on strawberries, among them the northern root-knot nematode (Meloidogyne hapla), which considered to be strawberry most important nematode pest worldwide. In August 2019, strawberry growers at Silifke (Mersin, Turkey) identified nematode-like symptoms on strawberry roots and infected seedlings were brought to the nematology laboratory at of Mersin University for diagnostics. Roots were separated into small pieces and nematode extraction was performed by a modified Baermann funnel method and identified under the microscope. DNA was extracted from individual nematodes using Worm Lysis Buffer(WLB (+)). The species-specific SCAR markers (JMV1, JMV2, and JMVhapla) yielded a 440 bp band specific to M. hapla. The 28S rRNA gene region, obtained using the general primers D2\D3, sequence was analysed from. The analyzed sequence was 100% identicle to M. hapla. The gene sequences were deposited into GenBank database with accession numbers MN897751 and MN895037. Both morphological and molecular diagnostic methods confirmed that the strawberry plants collected in Silifke were infested with M. hapla. To our best knowledge this is the first report of plantparasitic nematode species M. hapla infecting strawberry in Turkey. Currently, the adverse effect of RKN on strawberry production in the region is unknown to strawberry growers.
