Uterine infections are a major reproductive problem in livestock. We conducted two experiments to investigate factors that may modulate uterine responses to infectious bacteria. In Exp. 1, ewes received intrauterine inoculations of either saline or bacteria (75 x 10(7) cfu of Actinomyces pyogenes and 35 x 10(7) cfu of Escherichia coli) on either d 0 or 7 of the estrous cycle. Vena caval samples containing uteroovarian blood were collected twice daily from 12 h before until 6 d after inoculation. Only ewes inoculated with bacteria on d 7 developed infections. Basal (4.8 vs .4 pmol), lipopolysaccharide-stimulated (14.2 vs 6.1 pmol), and concanavalin A-stimulated (65.8 vs 21.6 pmol) blastogenesis (i.e., [3H]thymidine incorporation) of vena caval lymphocytes was greater (P < or = .002) for ewes inoculated with bacteria or saline on d 0 rather than on d 7. The number (per 100 white blood cells) of lymphocytes was greater (41.3 vs 30.8, P < .001) and that of neutrophils was less (42.5 vs 51.6, P < .001) in ewes inoculated on d 0 rather than d 7. Bacteria increased (P < .05) vena caval PGF(2 alpha) but not PGE2 concentrations. In Exp. 2, two protein fractions (molecular weights of > or = 100 kDa and approximately 12.7 kDa) from chromatography of uterine flushings collected on d 0 or 7, or 18 d after ovariectomy on d 0 or 7, modulated phytohemagglutinin-stimulated blastogenesis; the heavier fraction from d 0 had a stimulatory component, but the major effects of the fractions were inhibitory. The differences in immune function and regulation between d 0 and 7 probably explain how the uterus of follicular phase ewes was able to prevent the development of an infection.
Background Recent emerging evidence supports the role of miR-196a2 in various human diseases. However, its role in type 1 diabetes mellitus (T1DM) is still underestimated. We aimed, for the first time, to investigate the expression of miR-196a2 in T1DM and the association of miR-196a2 (rs11614913) polymorphism with susceptibility of T1DM in a sample of patients from Cairo, Egypt. Methods The study included 150 patients and 150 healthy subjects. Evaluation of rs11614913 genotypes and miR-196a2 expression was done using the allelic discrimination and quantitative reverse transcriptase polymerase chain reaction (PCR) method, respectively. Results The Hardy-Weinberg equilibrium of single nucleotide polymorphism(SNP) was detected among controls (p = 0.2). Our results revealed that the TT genotype was more frequent in patients (22.6%) than controls (10%) while the CC genotype was more frequent in controls (47.3%) than patients (39.3%) (p = 0.01). The frequency of the T allele was significantly higher in patients than in controls (41.7 vs. 31.3%), while the C allele was more frequent in controls (p = 0.008). After adjustment for traditional risk factors, the association of the TT genotype with T1DM remained significant (TT vs. CC, odds ration [OR] = 3.2, 95% confidence interval [CI]: 1.4–7.4, p = 0.005). Power analysis of the data yielded a statistical power of 80% for the miR-196a2 rs11614913 with T1DM. Relative expression of miR-196a2 showed significant decrease in patients compared to controls (median = 0.09, 0.5, interquartile range [IQR] = 0.03–1.6, 0.1–2.1). However, miR-196a2 expression showed no significant difference between different rs11614913 genotypes (p = 0.5). Conclusions Our findings demonstrated that miR-196a rs11614913 is associated with T1DM and decreased expression of miR-196a2 may play a role in pathogenesis of T1DM.
BACKGROUND:Obesity is a multi-factorial chronic disorder. A considerable number of studies have been performed to figure out whether there is an association between obesity and polymorphisms of gene IL-6 (174G/C), but the results are equivocal.AIM:This study aimed to find out whether the IL-6 (174G/C) gene was associated with the risk of developing obesity in Egyptian children.SUBJECTS AND METHODS:The study included 149 children and adolescents with age ranged between 9.5 – 18 years. Eighty-five of them were obese which BMIZ-score is > 2, and sixty-four children with BMIZ-score ≤ 2 served as control group. Serum level of IL-6 and genetic analysis for IL-6 (174G/C) gene polymorphism were done.RESULTS:Obese children had significantly higher serum levels of IL-6 as compared to those of control children (P = 0.003). A high percentage of IL-6 polymorphism GC was found in obese subjects (93.7%), while the control group had a higher percentage of IL-6 polymorphism GG (70.6 %).CONCLUSION:Our study showed that carriers of the C allele for the IL-6 (174G/C) polymorphism have higher BMI. As the G174C polymorphism is likely to affect IL-6 expression and its physiological regulation; consequently this polymorphism may affect adiposity.
Obesity is complex heterogeneous disease controlled by genes, environmental factors, and their interaction. Genetic factors account for 40–90% of the body mass index variations. Body mass index (BMI) of children correlates more closely with maternal than paternal BMI. So, this studu was aimed to investigate the role of leptin receptor LEPR Gln223Arg, the uncoupling protein 2 (UCP2 G 866 A) and insulin receptor gene (INSR exon 17) polymorphisms in the pathogenesis of obesity. A cross-sectional study executed on 130 children and their obese mothers; classified into 2 groups according to their BMI. The 2 groups were evaluated regarding the anthropometry. Restriction fragment length analysis for LEPR Gln223Arg, UCP2 -866 G/A and INSR exon 17 polymorphisms were applied. It was reported that increased risk of obesity was found in LEPR AG + AA genotype and the A allele. Significant statistical difference was detected only in female children. Concerning UCP2, the AG followed by the GG genotype was the most frequent in all groups and the G allele was the mostly present in obese mothers and obese male children but with no statistical significance. There was difference in the INSR genotype and alleles between groups, but this difference was not statistically significant. This study concluded that the LEPR Gln223Arg, UCP2 G 866 A and INSR exon 17 polymorphisms are related to obesity in Egyptian population. Further researches on larger population are recommended to ascertain the implications of LEPR, UCP2 and INSR polymorphisms in obesity.
