Effect of water dispersible single-walled carbon nanotubes on the medullary cardiovascular control was studied in normotensive (control) and spontaneously hypertensive rats using physiological and biochemical approaches. We have shown for the first time the possible contribution of nitric oxide (NO) to single-walled carbon nanotube-induced effects in the cardiovascular nuclei of the medulla oblongata (the lateral reticular nucleus and nucleus ambiguous). The data of biochemical experiments indicate that single-walled carbon nanotubes added to the medulla oblongata homogenates induce an enhancement of constitutive nitric oxide-synthase (cNOS) activity, which is mostly neuronal nitric oxide-synthase (nNOS) in the brain, and an increase in the level of nitrite (NO -2 ), a stable nitric oxide metabolite which serves nitric oxide bioavailability marker. In physiological experiments, it has been revealed that effects of single-walled carbon nanotube injection in the cardiovascular nuclei under study are attenuated after preliminary nNOS inhibiting. An analysis of single-walled carbon nanotube-induced changes in the levels of malonic dialdehyde, an oxidative stress marker, as well as the levels of reactive nitrogen species (pools of nitrate anions), and the activity of Ca 2+ -independent inducible nitric oxide synthase (iNOS) has indicated that single-walled carbon nanotubes used in the study do not promote oxidative and nitrosative stress in control and spontaneously hypertensive rats.
Процеси вільнорадикального окислення є важливою ланкою патогенезу ревматоїдного артриту. Активні форми оксигену та нітрогену за нормальних фі-зіологічних умов постійно утворюються в клітинах у невеликих кількостях під час окисного фосфорилювання. Вони виконують функції передачі клітинних сигна-лів, знищення патологічних чинників, регуляції апоптозу, проліферації та бага-тьох інших функцій клітин. Надмірна продукція активних форм оксигену за пато-логічних чи стресових умов призводить до пошкодження білків, ліпідів, нуклеїно-вих кислот і матричних компонентів клітини, зокрема, синовіальної мембрани і позасуглобових тканин. Мета роботи -визначити зміни пулів активних форм оксигену та нітрогену в сироватці крові щурів за експериментального карагенін-і колаген-індукованого запального артриту. Метод дослідження активних форм оксигену та нітрогену -спектрофотометричний. Результати досліджень: швид-кість генерації активних форм оксигену (О 2•-, •OH) і стаціо нарні пули H 2 O 2 в цир-куляції у групі експериментальних тварин з колагеновим артритом була вищою, ніж у групі тварин з карагеніновим запальним процесом. У цій же групі швидкість генерації активних форм нітрогену de novo, зокрема активність cNOS та, особли-во, iNOS, як і стаціонарні пули активних форм нітрогену ( NO 2 − , ) також є ви-щими. Висновок: у групі тварин із колагеновим артритом, який є аналогом ревма-тоїдного артриту у людини, продукти оксидативного та нітрозативного стресів активніше сприяють формуванню патологічних змін у тканинах, порівняно з кара-геніновою моделлю імунозапального процесу.Ключові слова: ревматоїдний артрит, колагеновий артрит, карагеніновий артрит, активні форми оксигену, активні форми нітрогену.
