Сбои в работе системы фибри-нолиза могут приводить к состояниям, угрожающим жизни, -тромбозам и неконтролируемым кровотечениям. Наблюдается стремительный прогресс в понимании молекулярных механизмов работы системы фибринолиза, детально описаны все основные участники и реакции процес-са, тем не менее вопросы регуляции и оценки состояния системы фибринолиза изучены слабо. В данном обзоре представлен анализ накопленной информации о работе системы фибриноли-за и сопутствующей патологии. Подробно рассмотрены методы оценки и диагностики состояния системы фибринолиза. Fibrinolysis is an important biochemical process responsible for the dissolution of fibrin clots, which appear during the blood coagulation process. Disorders in fibrinolysis system may result in life-threatening conditions like thrombosis and bleedings. At this time, we know a lot about the molecular mechanism of fibrinolysis, all reactions are described in details, and yet its regulation and evaluation of fibrinolysis state are still unclear. In this review we analyze the information about how fibrinolysis functions and what disorders of this system can be met. We describe in details the methods used to evaluate the state of fibrinolysis system. При этом запускается множество процессов: одни отвечают за формирование тромбо-цитарного агрегата, другие обеспечивают образова-ние фибринового сгустка, третьи вызывают растворе-ние тромба. Отдельные белки могут принимать участи во всех процессах, обеспечивая взаимную регуля-цию различных этапов свертывания. В данном обзоре мы остановимся на функционировании и регуляции финального этапа существования тромба -фибри-нолизе.Нарушения в системе фибринолиза могут быть связаны с увеличением риска развития венозного тромбоза [1], но не во всех работах такая закономер-ность была обнаружена [2]. В ряде случаев, таких как инфаркт миокарда, тромбоз глубоких вен, легочная эмболия, инсульт, возникает необходимость экстрен-ного удаления тромба, препятствующего кровотоку. Для этого фибринолитическую систему организ-ма приводят в гиперактивное состояние при помощи таких препаратов, как тканевый активатор плазми-ногена, стрептокиназа [3] или урокиназный актива-тор плазминогена [4]. Fibrinolysis is a cascade of proteolytic reactions occurring in blood and soft tissues, which functions to disintegrate fibrin clots when they are no more needed. In order to elucidate its regulation in space and time, fibrinolysis was investigated using an in vitro reaction-diffusion experimental model of blood clot formation and dissolution. Clotting was activated by a surface with immobilized tissue factor in a thin layer of recalcified blood plasma supplemented with tissue plasminogen activator (TPA), urokinase plasminogen activator or streptokinase. Formation and dissolution of fibrin clot was monitored by videomicroscopy. Computer systems biology model of clot formation and lysis was developed for data analysis and experimental planning. Fibrin clot front propagated in space from tissue factor, followed by a front of clot dissolution propagating from the same source. Velocity of lysis front propagation linearly depended on the velocity clotting front propagation (correlation r2 = 0.91). Computer model revealed that fibrin formation was indeed the rate-limiting step in the fibrinolysis front propagation. The phenomenon of two fronts which switched the state of blood plasma from liquid to solid and then back to liquid did not depend on the fibrinolysis activator. Interestingly, TPA at high concentrations began to increase lysis onset time and to decrease lysis propagation velocity, presumably due to plasminogen depletion. Spatially non-uniform lysis occurred simultaneously with clot formation and detached the clot from the procoagulant surface. These patterns of spatial fibrinolysis provide insights into its regulation and might explain clinical phenomena associated with thrombolytic therapy.
