Стремительный технический прогресс способствует появлению все новых подходов в клеточной биологии, одним из них является клеточная микрофлюидика. Применение технологий микрофлюидики открыло новые возможности по культивированию, прецизионному анализу и манипулированию как популяциями клеток, так и отдельными клетками. Основой новой технологии является микрофлюидный чип - миниатюрное устройство, содержащее систему микро- и наноканалов, полостей, мембран и других элементов. Возможность прецизионного управления пространственным расположением клеток и их микроокружением предоставляет уникальные и беспрецедентные возможности для биомоделирования in vitro фунциональных элементов органов и тканей. В данном обзоре приведены примеры построения и применения таких трехмерных микрофлюидных клеточных моделей для анализа протекающих в них физиологических и патологических процессов. Особое внимание уделено влиянию клеточного микроокружения клетки на её функционирование. Significant technological progress has brought new approaches to cell biology. Using microfluidic technologies has opened new opportunities for cultivation, analysis, and manipulation of both individual cells and their populations. The basis of the new technology is a microfluidic chip, a miniature device containing a system of micro- and nanochannels, cavities, membranes, and other elements. The precise control of spatial arrangement of cells and their microenvironment opens new prospects for in vitro biomodeling of functional elements of organs and tissues. This review shows examples for construction and application of such three-dimensional microfluidic cellular models for analysis of physiological and pathological processes. Particular attention is paid to the influence of cellular microenvironment on cell functioning.
Эндотелиальные клетки, выстилающие стенки сосудов, являются одними из важнейших регуляторных элементов кровеносной системы. Непосредственно соприкасаясь с потоком крови, эти механочувствительные клетки способны детектировать свою деформацию через ее тангенциальный компонент (сдвиг) и составляющую, направленную по нормали к поверхности (растяжение). Деформация сдвига является ключевым индуктором комплекса сигнальных путей, опосредуемых тирозинкиназами, интегринами, ионными каналами, вовлекающих также мембранные липиды, гликокаликс и другие клеточные компоненты. На фоне достаточно большого количества данных о сигнальной трансдукции, в литературе меньше внимания уделено клеточной адаптации к сдвиговой деформации и сравнительно мало информации об участии генов стрессового ответа. Гидродинамические условия в определенных зонах сосудистой системы характеризуются значительной неоднородностью, что может приводить к ослаблению обратных связей, необходимых для поддержания гомеостаза в эндотелиальных клетках. Это может способствовать развитию заболеваний, например, таких, как атеросклероз. В обзоре обсуждаются новые аспекты и концепции, связанные с ответами эндотелиоцитов на сдвиговую деформацию и основные методы анализа эффектов сдвиговой деформации in vitro . Цель исследования. Обобщение современных данных о механизмах механочувствительности и механотрансдукции эндотелия. Результаты. В обзоре изложены основные механизмы механочувствительности клеток эндотелия, пути внутриклеточной передачи сигнала, рассмотрено вовлечение механизмов стрессового ответа клеток и адаптации. Обсуждаются эксперименты по изучению молекулярных основ механотрансдукции, в том числе белков и других молекул, вовлеченных в детектирование, передачу сигнала и клеточный ответ на сдвиговую деформацию. Endothelial cells lining the walls of blood vessels are one of the most important regulatory elements of the circulatory system. These mechanosensitive cells are in a direct contact with the flow of blood and able to detect deformation through its tangential component (shear) and the component directed along the normal to the surface (tension). Shear stress is the key inducer of the complex of signaling pathways mediated by tyrosine kinases, integrins, ion channels, involving also membrane lipids, glycocalyx and other cellular components. There are large amount of data on signal transduction in the literature, but less attention is paid to cellular adaptation to shear stress and there is relatively little information on the involvement of stress response genes in that process. Hydrodynamic conditions in certain zones of the vascular system are characterized by considerable heterogeneity, which can lead to weakening of feedbacks necessary for maintaining homeostasis in endothelial cells. This can contribute to the development of diseases such as atherosclerosis. This review presents new aspects and concepts related to the responses of endotheliocytes to shear stress and, in addition, highlights the basic methods of analyzing the effects of shear stress in vitro . Purpose of the study. Generalization of modern data on mechanisms of mechanosensitivity and mechanotransduction of the endothelium. Results. The review outlines the main mechanosensitivity mechanisms of endothelial cells, the pathways of intracellular signaling, the involvement of mechanisms of cellular stress response and adaptation. There are descriptions of experiments in which the molecular basis of mechanotransduction is identified, including the determination of proteins and other molecules involved in detection, signal transduction, and cellular response to shear stress.

Objectives. The chemistry of 67Ga and 68Ga radionuclides plays a key role in nuclear medicine for applications in radiopharmaceuticals, in particular, in noninvasive in vivo molecular imaging techniques. The use of radiometals for labeling biomolecules typically requires the use of bifunctional chelators, which contain a functional group for covalent bonding with the targeting vector in addition to the polydentate fragment coordinating the metal. The aim of the present review article is to analyze the currently accumulated experimental material on the development and application of bifunctional chelators of gallium cations in medical research, as well as to identify the main requirements for the structure of the chelator and its complexes with 68Ga, which are used to create effective Gabased pharmaceutical preparations.Results. The review analyzed macrocyclic bifunctional chelators forming stable in vivo complexes with 68Ga and acyclic chelators, whose main advantage is faster complexation kinetics due to the short half-life of 68Ga. The advantages and disadvantages of both types of ligands were evaluated. In addition, a critical analysis of the binding constants and the conditions for the formation of complexes was presented. Examples of the influence of the geometry, lipophilicity, and total charge of the metal complex on the biodistribution of target radiopharmaceuticals were also given.Conclusions. Despite the progress made in the considered areas of bifunctional chelators, the problem of correlating the chemical structure of a metal-based radiopharmaceutical with its behavior in vivo remains important. Comparative studies of drugs having an identical targeting vector but containing different bifunctional chelating agents could help further elucidate the effectof metal chelate moiety on pharmacokinetics. In order to create effective bifunctional chelating agents, it is necessary to take into account such factors as the stability and inertness of the chelator and its complexes under physiological conditions, lipophilicity, complexation kinetics, chelation selectivity, combinatoriality of the basic structure, along with economic aspects, e.g., the availability of raw materials and the complexity of the synthesis scheme.

Мезенхимные стволовые фибробластоподобные клетки FRSN были культивированы в условиях воздействия постоянного потока жидкости в микрофлюидном чипе. Проведены исследования миграции клеток на разных стадиях адгезии под действием потока в различных областях чипа. Обнаружены значительные перемещения клеток в режиме «stick-slip» вдоль направления потока. The fluid flow exerts a significant effect on most cells in the body. This effect can involve cell migration under the action of shear stress or nutrient gradient. FRSN mesenchymal stem cells were cultured under the action of a constant fluid flow of low intensity in a microfluidic chip. The study of cell migration at different stages of adhesion was performed under the action of flow in different areas of the chip. Significant cell movements in a stick-slip mode along the flow direction were observed.

Для адекватного биомоделирования многоклеточных тканевых структур in vitro необходим тщательный подбор используемых в качестве подложки для клеток материалов. В работе мы исследовали адгезию эндотелиоцитоподобных клеток EA.hy926 к стеклу и полимерным материалам, часто применяемых в микрофлюидике для моделирования тканевых структур. Методом конфокальной сканирующей микроскопии было показано, что различные виды полидиметилсилоксана (ПДМС) характеризуется низкой адгезией клеток по сравнению со стеклом и специально подготовленным полистиролом (ПС). При этом обработка кислородной плазмой не оказывает существенного влияния на адгезию. Обработка коллагеном I, напротив, существенно улучшает адгезию. Белковый гель достаточно хорошо покрывает как гидрофильную поверхность стекла, так и гидрофобную поверхность полимера. Методом сканирующей зондовой микроскопии были также исследованы особенности распластывания отдельных клеток на поверхности субстрата. Cell-substrate adhesion is a key factor affecting the cell functionality and fate. Therefore, for adequate biomodeling of cellular structures in vitro, it is necessary to carefully select the materials used. In this study, we evaluated adhesion of endothelial-like cells EA.hy926 to polymeric materials and glass, which are used in microfluidics to model blood vessels. It was shown that different types of polydimethylsiloxane (PDMS) provided low cell adhesion compared to glass and specially prepared polystyrene (PS). The treatment with oxygen plasma did not exert a significant effect on adhesion. Covering with collagen I, on the contrary, significantly improved adhesion. The protein gel quite well covered both the hydrophilic surface of the glass and the hydrophobic surface of the polymer. The features of individual cells spreading on the substrate surface were also studied using scanning probe microscopy.

К настоящему моменту накоплен большой объём данных о клеточных механизмах активации продукции NO, однако сведений о динамике генерации оксида азота эндотелиальными клетками в зависимости от гидродинамических условий недостаточно. В этой связи разработка микрофлюидных систем in vitro, имитирующих кровеносное русло, и изучение в них эндотелия в сложных гидродинамических условиях является актуальной задачей. В данной работе для создания контролируемых гидродинамических условий для монослоя эндотелиоцитоподобных клеток EA.hy926 была спроектирована и разработана микрофлюидная система, имитирующая линейные участки микрососудистого русла. Методом непрямого определения содержания оксида азота (II) NO с использованием флуоресцентного зонда 4,5-диаминофлуоресцеина DAF-2 впервые получены данные об увеличении продукции NO клетками EA.hy926 при механическом стрессе, создаваемом потоком ростовой среды. Представлены расчетные гидродинамические характеристики микрофлюидной системы, а также методика измерения продукции NO. Возможность исследования функциональной активности эндотелия позволяет использовать разработанную микрофлюидную модельную систему как для изучения клеточно-автономных регуляторных свойств эндотелия при действии ряда вазоактивных фармакологических препаратов и других методов воздействия на эндотелий, так и при моделируемой дисфункции эндотелия. Endothelial cells lining vascular walls transform the flow-induced deformation of their own structures into chemical signals, one of which, nitric oxide (NO), is an important regulator of the vascular lumen diameter. By present, a large amount of data on cellular mechanisms for activation of NO production has been accumulated. However, there is insufficient information on changes in endothelial NO generation under different hydrodynamic conditions. Therefore, development of microfluidic systems that model blood vessels in vitro and using them to study the endothelium under complex hydrodynamic conditions are relevant tasks. In this study, a microfluidic system was developed to create controlled hydrodynamic conditions for a monolayer of endotheliocyte-like cells EAhy.926. This system simulates linear sections of the microvasculature. By indirect measurement of NO (II) content with a fluorescent 4,5-diaminofluorescein (DAF-2) probe, we showed an increase in the NO production by EAhy.926 cells under mechanical stress generated by the medium flow. The article presents the method for measuring NO production and the calculated hydrodynamic characteristics of the microfluidic system. The results showed that the developed microfluidic model system is promising for studying cell-autonomous regulatory properties of the endothelium both under the action of vasoactive agents and in simulated endothelial dysfunction.
