Artificial insemination has arguably been the most important practice contributing to the advancement of animal production. The numerous advantages of artificial insemination are augmented when the semen is cryopreserved and conserved for long periods. Unfortunately, the utility of cryopreserved semen is limited because for most mammals, even cattle, a considerable proportion of sperm loose their fertility during freezing-thawing. In many species, this loss of fertility is substantial, rendering cryopreserved semen impractical for routine use. Our goals are to characterize the nature of the sublethal sperm damage caused by cryopreservation, and to develop a method to improve the functional fertility of thawed semen. Les nombreux avantages qu'offre cette technique sont augmentés lorsque la semence est cryoconservée et entreposée pour de longues périodes. Malheureusement, l'utilisation de la semence cryoconservée est limitée puisque chez la majorité des mammifères, incluant la vache, une large proportion des spermatozoïdes perdra leur motilité pendant le processus de congéla-tion-décongélation. Chez plusieurs espèces, cette perte de fertilité est suffisamment substantielle pour rendre impraticable l'utilisation routinière de la semence cryoconservée. Les objectifs de notre laboratoire portent sur la caractérisation des dommages sous létaux subis par les spermatozoïdes lors de la cryoconservation et le développement de méthodes permettant d'améliorer la fertilité des spermatozoïdes décongelés. Cette revue décrit notre théorie selon laquelle les spermatozoïdes seraient prématurément activés par la cryoconservation ("cryo-capacitation"). Différentes stratégies permettant de prévenir ce dommage y sont également abordées.
ABSTRACT:We hypothesized that cryopreservation and incubation in conditions that mimic the female genital tract following insemination increases the susceptibility of ram sperm DNA to denaturation. Ram sperm samples (n ϭ 12) underwent the sperm chromatin structure assay (SCSA) and semen quality tests, including motility parameters, viability, and chlortetracycline fluorescence (CTC) patterns. We also assessed correlations between SCSA variables and semen quality parameters. Analyses were performed for both fresh and cryopreserved samples at 0, 3, and 20 hours of incubation in synthetic oviductal fluid (SOF; 39ЊC, 5% CO 2 ). The SCSA variables, mean alpha t (X␣ t ) and standard deviation of alpha t (SD␣ t ), were higher because of cryopreservation (P Ͻ .05, P Ͻ .001, respectively) after 20 hours in SOF. For both fresh and frozen spermatozoa, SCSA values (X␣ t , SD␣ t , and the percentage of cells outside the main population of ␣ t [%COMP␣ t ]) increased during incubation in SOF. Motility was negatively correlated with both SD␣ t and %COM-P␣ t , ranging from Ϫ0.39 (P Ͻ .01) to Ϫ0.59 (P Ͻ .001) for both fresh and cryopreserved semen; viability also was negatively correlated with X␣ t , SD␣ t , or %COMP␣ t (Ϫ0.36; P Ͻ .05, Ϫ.40 and -.46; P Ͻ .01, respectively) in fresh semen. The %COMP␣ t was positively correlated to the percentage of CTC pattern AR (P Ͻ .001) and negatively correlated to the percentages of patterns F and B (Ϫ0.33 to Ϫ0.60, P Ͻ .05 to P Ͻ .001). Variation among ejaculates within ram was observed (P Ͻ .01). Cryopreservation clearly facilitates DNA damage in physiological conditions. The low to moderate correlations between SCSA variables and classical semen quality parameters indicate that the SCSA provides additional information to standard tests for evaluating ram sperm quality.
. 2002. Glycerol addition and conservation of fresh and cryopreserved ram spermatozoa. Can. J. Anim. Sci. 82: 347-356. Fresh extended ram semen has a short fertile lifespan whereas acceptable fertility with cryopreserved semen is achieved only by laparoscopy, which limits widespread artificial insemination in sheep. Although glycerol is considered essential for freezing spermatozoa, it is often included in extenders for short-term storage at above-freezing temperatures. To test the hypothesis that glycerol reduces the function of fresh sperm, ram semen was divided into two aliquots and diluted with commercial extenders that were identical, except that one contained 7% glycerol (n = 6). In a second experiment, ram semen was prepared for cryopreservation by a one-step dilution with a 7% glycerol extender or gradually, with a two-step protocol, to test the hypothesis that the method and time of glycerol addition affects sperm quality after freezing and thawing (n = 7). For both experiments, semen was diluted in a synthetic oviductal fluid (SOF-m) and sperm quality was assessed by computerassisted motility, viability and chlortetracycline fluorescence (CTC) patterns (an indicator of capacitation status). The presence of glycerol did not affect the quality of fresh sperm (P > 0.27). For cryopreserved sperm, the method of glycerol addition also did not affect thawed sperm. However, a decrease in sperm motility and viability, and different distribution of CTC patterns occurred due to the duration of time in extender and in SOF-m (P ≤ 0.0002). Cryo-capacitation was also observed. In conclusion, the presence of glycerol in the extender did not reduce ram sperm quality during conservation of the semen at 5°C or when it was used to completely and rapidly dilute the semen before cooling for cryopreservation. In artificial insemination, however, diluting the semen reduces the concentration of seminal plasma. To test the hypothesis that supplemental seminal plasma in extended ram semen improves conservation at 5°C, we added various concentrations of seminal plasma to semen during storage, and investigated subsequent sperm function in vitro. Semen was divided into three aliquots, extended in a commercial diluent (Triladyl) supplemented with 0, 10 or 25% (vol:vol) ovine seminal plasma and cooled to 5°C. After 8 and 24 h at 5°C, sperm were suspended in a modified synthetic oviduct fluid (SOF-m) at 39°C to mimic the female genital tract at insemination. Sperm aliquots were assessed for motility and chlortetracycline fluorescence after 0, 4 and 8 h in the SOFm. No significant differences were observed due to seminal plasma supplementation during conservation at 5°C or incubation in SOF-m at 39°C. However, decreased sperm motility and fewer non-capacitated sperm were observed concomitant with an augmentation of capacitated and acrosome-reacted cells during incubation in SOF-m. Therefore, the hypothesis that diluent supplementation with homologous seminal plasma improves ram sperm conservation or subsequent sperm function was not supported. Pour vérifier l'hypothèse qu'un supplément de plasma séminal améliorerait la conservation du sperme de bélier allongé à 5°C, les auteurs ont ajouté à ce dernier une quantité variable de plasma séminal pendant le stockage et examiné subséquemment la motilité des spermatozoïdes in vitro. Le sperme a été divisé en trois parties, allongé avec un diluant commercial (Triladyl) auquel on a ajouté 0, 10 ou 25 % (v:v) de plasma séminal d'ovin puis a été refroidi à 5°C. Après avoir passé 8 ou 24 heures à cette tempéra-ture, le sperme a été mis en suspension dans un fluide synthétique modifié imitant celui des trompes de Fallope (SOF-m) à 39°C, de manière à reproduire les conditions dans le tractus génital de la femelle à l'insémination. Les auteurs ont évalué la motilité des spermatozoïdes et la fluorescence à la chlortétracycline des aliquotes de sperme laissés 0, 4 et 8 heures dans le SOF-m. Aucune variation sensible n'a été relevée à la suite de l'addition de plasma séminal lors de la conservation du sperme à 5°C ou son incubation dans le SOF-m à 39°C. Néanmoins, la motilité réduite des spermatozoïdes et le plus petit nombre de spermatozoïdes non capacités sont cohérents avec la hausse du nombre de cellules capacitées et de cellules avec réaction acrosomique pendant l'incubation dans le SOF-m. On en déduit que la dilution du sperme de bélier avec du plasma séminal homologue n'améliore pas sa conservation ni le fonctionnement subséquent des spermatozoïdes.
