An automated determination of p-acetylaminohippurate in serum is described. It is combined with an existing procedure for determining inulin to give a simultaneous method. The same manifold can also be used to determine p-aminohippurate in combination with fructose by omitting hydrolysis. The results of automated and manual determination are compared. Recoveries of p-acetylaminohippurate and p-aminohippurate added to serum were quantitative.
An automated determination for xanthurenic acid in urine was devised based upon its reaction with 2,6-dichloroquinonechlorimide. The separation of xanthurenic acid from interfering substances, such as urine chromogens and phenolic compounds other than xanthurenic acid, was performed by using Sephadex G-10. The effluents containing the xanthurenic acid were determined automatically. As a result of the high sensitivity of this procedure, concentration technics are unnecessary.
SUMMARY The results of investigations on a family after a screening programme for congenital hypothyroidism are presented. The newborn child and her father appeared to have a congenital thyroxine binding globulin deficiency. Problems regarding standard thyroid hormone tests are discussed. It was observed that the free T, index as well as a commercial free T 4 determination introduced earlier can give misleading results. The first experience with another more promising free T4 determination is also presented.
