Anthrax and botulism are dangerous infectious diseases that can be fatal unless detected and treated quickly. Fatalities from these diseases are primarily due to endopeptidase toxins secreted by the pathogens. Rapid and sensitive detection of the presence of active toxins is the key element for protection from natural outbreaks of anthrax and botulism, as well as from the threat of bioterrorism. We describe an ultrasensitive polymerase chain reaction (PCR)-based assay for detecting proteolytic activity of anthrax and botulinum toxins using composite probes consisting of covalent peptide-DNA conjugate for the detection of anthrax, and noncovalent protein-aptamer assembly to assay botulinum toxin activity. Probes immobilized on the solid-phase support are cleaved by toxins to release DNA, which is detected by real-time PCR. Both assays can detect subpicogram quantities of active toxins isolated from composite matrices. Special procedures were developed to isolate intact toxins from the matrices under mild conditions. The assay is rapid, uses proven technologies, and can be modified to detect other proteolytic and biopolymer-degrading enzymes.

Этот вид делится на четыре подвида: ssp. tularensis, holarctica, mediasiatica и novicida, которые различаются по географическому ареалу распространения, вирулентности и эпидемиологическому потенциалу. До недавнего времени на территории РФ выявлялся только подвид holarctica. Однако в 2013 г. на Алтае был обнаружен природный очаг туляремии, в котором циркулируют штаммы F. tularensis subsp. mediasiatica, выделяемые ранее только в Средней Азии. Данные лабораторных исследований свидетельствуют о способности штаммов этого подвида вызвать инфекцию у кроликов и мышей, сравнимую по тяжести с инфекцией, вызываемой штаммами голарктического подвида. Однако вирулентность и эпидемическая опасность среднеазиатского подвида F. tularensis для человека до сих пор остается неопределенной, так как случаев туляремии у человека, вызванных штаммами этого подвида, не было зарегистрировано, что может быть связано с особенностями его географического распространения-и Средняя Азия, и Горный Алтай являются крайне редконаселенными регионами. Основным фенотипическим признаком этого подвида является отсутствие активности фермента β-лактамазы, отвечающего за природную устойчивость к β-лактамным антибиотикам (пенициллинам, цефалоспоринам и карбапенемам). При этом, несмотря на отсутствие детектируемой ферментативной активности, штаммы среднеазиатского подвида сохраняют устойчивость к этим антибиотикам. В настоящей статье мы приводим данные о том, что штаммы F. tularensis subsp. mediasiatica, вопреки общепринятым представлениям, обладают β-лактамазной активностью, но при этом скорость гидролиза β-лактамов значительно снижена по сравнению со штаммами подвида holarctica. Кроме того, при снижении количества микробных клеток в питательной среде начинает проявляться и антибиотикочувствительность. Мы выявили единственную подвидоспецифичную для subsp. mediasiatica нуклеотидную замену G/A в 290 положении гена blaB, кодирующего активную сериновую β-лактамазу. Эта замена приводит к аминокислотной замене Gly на Arg в 97 положении белка BlaB. Мы полагаем, что данная замена является наиболее вероятной причиной снижения активности этого фермента, обусловливая возможные конформационные изменения, приводящие либо к снижению сродства фермента к субстрату, либо к увеличению времени существования фермент-субстратного комплекса. Выявленная замена послужила основой для разработки аллель-специфичного ПЦР-теста, позволяющего определить принадлежность исследуемого штамма F. tularensis к подвиду mediasiatica.

Способ, при помощи которого происходит подавление защит-ных систем организма, не всегда очевиден, а механизм интоксикации изучен во многих случаях не полностью. Известно, что бактерии-возбудители токсикоинфекций, лишенные токсинов, в большинстве случаев авирулентны и не представляют существенной опасности или вызы-вают незначительную патологию, легко поддающуюся лечению [3,4], поэтому токсины таких микроорганизмов закономерно рассматривают как принципиальные факто-ры их вирулентности.Среди достаточно большого числа бактерий, проду-цирующих токсины, наибольшую опасность для жизни в результате интоксикации представляют возбудители ботулизма [5] и сибирской язвы [6]: высокая удельная токсичность продуцируемых токсинов обусловливает вы-сокую смертность в условиях интенсивной терапии и узкое «окно возможностей», при упущении которого вероятность летального исхода многократно возрастает [7,8]. В силу этого неспецифическая симптоматическая терапия указанных заболеваний может иметь ограничен-ный успех, а в случае массовой интоксикации смертность может многократно возрасти ввиду отсутствия доста-точного числа мест для проведения мероприятий реани-мации и интенсивной терапии [9]. В отличие от других упомянутых токсикоинфекций эффективные средства специфической терапии сибирской язвы и ботулизма до сих пор не созданы, а имеющаяся сибиреязвенная вакцина не удовлетворяет современным параметрам бе-зопасности применения и не может быть рекомендована для широкого использования. Между тем современная международная обстановка, глобализация экономики, логистики и миграционных потоков, а также растущая угроза биотерроризма диктуют необходимость создания эффективных мер защиты населения от этих патогенов.Достаточно давно стало очевидным, что наилучшим решением для предотвращения смертности и инвалид-ности у пациентов с указанными выше инфекциями является специфическая антитоксинная терапия и/или
Neutralization of the lethal toxin of Bacillus anthracis is an important topic of both fundamental medicine and practical health care, regarding the fight against highly dangerous infections. We have generated a neutralizing monoclonal antibody 1E10 against the lethal toxin of Bacillus anthracis and described the stages of receptor interaction between the protective antigen (PA) and the surface of eukaryotic cells, the formation of PA oligomers, assembly of the lethal toxin (LT), and its translocation by endocytosis into the eukaryotic cell, followed by the formation of a true pore and the release of LT into the cell cytosol. The antibody was shown to act selectively at the stage of interaction between Bacillus anthracis and the eukaryotic cell, and the mechanism of toxin-neutralizing activity of the 1E10 antibody was revealed. The interaction between the 1E10 monoclonal antibody and PA was found to lead to inhibition of the enzymatic activity of the lethal factor (LF), most likely due to a disruption of true pore formation by PA, which blocks the release of LF into the cytosol.
Aptamers are short single-stranded oligonucleotides which are selected via targeted chemical evolution in vitro to bind a molecular target of interest. The aptamer selection technology is designated as SELEX (Systematic evolution of ligands by exponential enrichment). SELEX enables isolation of oligonucleotide aptamers binding a wide range of targets of interest with little respect for their nature and molecular weight. A number of applications of aptamer selection were developed ranging from biosensor technologies to antitumor drug discovery. First aptamer-based pharmaceutical (Macugen) was approved by FDA for clinical use in 2004, and since then more than ten aptamer-based drugs undergo various phases of clinical trials. From the medicinal chemist’s point of view, aptamers represent a new class of molecules suitable for the development of new therapeutics. Due to the stability, relative synthesis simplicity, and development of advanced strategies of target specific molecular selection, aptamers attract increased attention of drug discovery community. Difficulties of the development of next-generation antibiotics basing on the conventional basis of combinatorial chemistry and high-throughput screening have also amplified the interest to aptamer-based therapeutic candidates. The present article reviews the investigations focused on the development of antibacterial aptamers and discusses the potential and current limitations of the use of this type of therapeutic molecules.
Live anthrax vaccine containing spores from attenuated strains STI-1 of Bacillus anthracis is used in Russia and former CIS (Commonwealth of Independent States) to prevent anthrax. In this paper we studied the duration of circulation of antibodies specific to spore antigens, the protective antigen (PA), the lethal factor (LF) and their domains (D) in donors’ blood at different times after their immunization with live anthrax vaccine. The relationship between the toxin neutralization activity level and the level of antibodies to PA, LF and their domains was tested. The effect of age, gender and number of vaccinations on the level of adaptive post-vaccination immune response has been studied. It was shown that antibodies against PA-D1 circulate in the blood of donors for 1 year or more after immunization with live anthrax vaccine. Antibodies against all domains of LF and PA-D4 were detected in 11 months after vaccination. Antibodies against the spores were detected in 8 months after vaccination. A moderate positive correlation was found between the titers of antibodies to PA, LF, or their domains, and the TNA of the samples of blood serum from the donors.
