We have used a torque magnetometer to measure de Haas - van Alphen oscillations in the magnetization of two-dimensional electrons in GaAs/AlGaAs heterostructures and multiple-quantum-well systems for temperatures ranging from 0.125 K to 4.2 K and in magnetic fields of up to 15 T. Our results indicate that for high magnetic fields the density of states can be described by a series of Lorentzian-broadened Landau levels with a broadening that is independent of the magnetic field, B, and Landau level index, n. However, at low magnetic fields the Lorentzian-broadened density of states becomes indistinguishable from a Gaussian one with a broadening that is proportional to . The high-field behaviour of the Landau level line-shape is shown to differ appreciably from the low-field case as reported by other workers using both magnetization and other experimental methods. The reliability of this and other experimental techniques is discussed.

The PCR was used to amplify a targeted region of the ribosomal DNA from 84 Candida isolates. Unique product sizes were obtained for Candida guilliermondii, Candida (Torulopsis) glabrata, and Candida pseudotropicalis. Isolates of Candida albicans, Candida tropicalis, Candida stellatoidea, Candida parapsilosis, and Candida krusei could be identified following restriction digestion of the PCR products.

De Haas-van Alphen ͑dHvA͒ oscillations are observed for Landau levels ͑LLs͒ with filling factors between 4 and 52, at temperatures in the range 50 mK to 1 K, in experiments on high-mobility GaAs/͑Al, Ga͒As heterojunctions. The oscillations become sawtooth-shaped at low filling factors, and theoretical fits to the data, assuming the two-dimensional electron gas to be a non-interacting Fermi system, show the shape of LLs to be close to a ␦ function. The small residual width ͑ϳ0.4 meV or less͒ fits equally well to either a Gaussian or a Lorentzian density-of-states model. In almost all cases, a constant background density of states has to be included to obtain a satisfactory fit. Weak odd-filling-factor dHvA peaks are detected at high fields, from which a g-factor enhancement of 15 can be inferred. Comparison of the scattering time derived from the fits before and after illumination, with the momentum relaxation time derived from transport, reveals a counterintuitive behavior in the bulk-modulation-doped sample.

Sixteen biopsies from patients with oral lichen planus (10), simple keratosis (3) and lichenoid reactions (3) were studied using monoclonal antibodies directed against lymphocyte markers. T lymphocytes predominated in the cellular infiltrates of the epithelium and lamina propria of all biopsies, and cells positive with Leu 3a and Leu 2a antibodies were also present at both sites in all specimens. However, many of the Leu 3a positive intraepithelial cells had a dendritic appearance and distribution consistent with their being Langerhans cells. Limited results from cell counts on three lichen planus specimens suggested that the majority of intraepithelial T cells were of the cytotoxic/suppressor phenotype (OKT3 and Leu 2a positive).

Confocal laser scanning microscopy permitted high resolution analysis of reconstituted human oral epithelium invasion by C. albicans and identified strain differences in the invasion process. Association between extensive hyphal morphology, direct epithelial penetration and high surface colonization were made with the 'highly invasive' strains.

The inflammatory cell infiltrate in biopsy material of chronic hyperplastic candidosis (CHC) from the oral mucosa was characterised using immunocytochemical techniques. Nine specimens were stained for human kappa and lambda immunoglobulin light chains, CD68 antigen (macrophages), lysozyme (macrophages, granulocytes), CD3 antigen (T-lymphocytes), CD20 antigen (B-lymphocytes) and leucocyte common antigen (LCA). In addition, these and a further 13 specimens were also examined for immunoglobulin (Ig)-containing cells (IgA, IgG and IgM). The density of the infiltrate varied considerably between cases; T-lymphocytes were the dominant cell type (53.9%), with fewer B-lymphocytes (8.2%) and macrophages (14.2%). Many Ig-containing cells were seen, and although IgG-containing cells predominated, (60.8%, SD +/- 9.0) there was a high proportion of IgA-containing cells (36.7%, SD +/- 9.1) with few IgM-containing cells (2.5%, SD +/- 3.0). Many neutrophils, together with smaller numbers of T-lymphocytes and macrophages, were seen in the epithelium. It is suggested that mucosal defence to Candida infection involves a cell-mediated reaction in which there is recruitment of macrophages and local production of immunoglobulin with a prominent IgA component.
