Priming has been used to improve the performance of germination at the field, and potassium nitrate (KNO 3 ) is a promising compound for this purpose. The nitrate (NO 3 ) could be absorbed, being used in the metabolism of the embryo, through the enzyme nitrate reductase (NR). Besides, the priming could also activate the response of the antioxidant system, becoming the primed seeds more prepared for possible stresses. Thus, the aim of this study was to evaluate the metabolic effect of nitrate in tomato seed germination by the quantification of NR activity, and also evaluate the activity of some antioxidant enzymes as superoxide dismutase (SOD), catalase (CAT) and ascorbate peroxidase (APX). Tomato seeds were primed using solutions of polyethylene glycol (PEG 6000) -1,1MPa, 50 mM KNO 3 and PEG+KNO 3 . The variables analyzed were germination (germinability, mean germination time, mean germination rate, coefficient of variation of the germination time, uncertainty and synchrony) nitrogen, total proteins and enzymes. The germination data were analyzed using an ANOVA, comparing the averages by Scott-Knott test (P < 0.05). To analyze the nitrogen, protein and enzymatic activities, we used a Kruskal-Wallis test (P < 0.05). The results show an increase in the NR activity, as well as in the antioxidant enzymes. The germination time (t) and germination rate (v) primed in KNO 3 had a better performance compared to the other treatments. In conclusion, the observed benefits in tomato seeds primed with KNO 3 were related to the activity of the enzyme nitrate reductase in the production of nitrite/nitric oxide, which acted promoting a faster germination.
ABSTRACTThe aims of this study were to verify the effects of different tests in the overcoming dormancy, excellent germination terms, as light and temperature, and the possibility of re-induction of desiccation tolerance in Senna multijuga seeds. We evaluated two tests for overcoming dormancy: immersion in water with an initial temperature of 100ºC (hot water) and immersion in water at a constant temperature at 100ºC for 20 seconds
Em virtude disso, foi avaliado o efeito do eliciador exógeno quitosana no sistema antioxidante enzimático de jaborandi (Piper mollicomum ABSTRACT Elicitors are used in plant physiology studies for comprehension of defense mechanisms against herbivore attach and pathogen infections. For this reason, the exogenous chitosan effect was tested as an elicitor on Jaborandi (Piper mollicomum Kunth). The heat plume associated with fire has been hypothesized to cause sufficient water loss from trees to induce embolism and hydraulic failure. However, it is unclear whether the water transport path remains sufficiently intact during scorching or burning of foliage to sustain high water loss. We measured water uptake by branches of Magnolia grandiflora while exposing them to a range of fire intensities and examined factors influencing continued water uptake after fire. Burning caused a 22-fold mean increase in water uptake, with greatest rates of water loss observed at burn intensities that caused complete consumption of leaves. Such rapid uptake is possible only with steep gradients in water potential, which would likely result in substantial cavitation of xylem and loss of conductivity in intact stems. Water uptake continued after burning was complete and was greatest following burn intensities that killed leaves but did not consume them. This post-fire uptake was mostly driven by rehydration of the remaining tissues, rather than evaporation from the tissues. Our results indicate that the fire plume hypothesis can be expanded to include a wide range of burning conditions experienced by plants. High rates of water loss are sustained during burning, even when leaves are killed or completely consumed.
The loss of benefits after re-drying is one of the drawbacks of the seed priming technique. Different types of stresses have been used before re-drying to preserve the priming benefits. This process may be seen as promoting cross tolerance to increase the defense mechanisms that prevent loss of viability in seeds primed after drying. We tested the effect of some stresses to induce cross-tolerance and different drying conditions with the aim of maintaining priming benefits in melon seeds. The seeds were primed in an aerated KNO 3 solution (0.35M), -1.7MPa, 25 °C, in the dark for six days. The primed seeds were then submitted to slow drying, fast drying, cold shock + slow drying, cold shock + fast drying, heat shock + slow drying, heat shock + fast drying, PEG + slow drying, PEG + fast drying, ABA + slow drying, ABA + fast drying and no drying (planted directly after priming). We evaluated antioxidant enzyme activities (SOD, CAT and APX), germinability, mean time of germination (MTG) and mean rate of germination (MRG). A completely randomized design was used with three repetitions of 50 seeds in each treatment. Data were analyzed by ANOVA and means were compared by the Scott-Knott test (p ≤ 0.05). ABA increased SOD activity after drying and CAT activity was reduced by priming. APX activity was not observed. The stress submission prior to re-drying improved the MRG and reduced MTG. Therefore, the induction of the cross-tolerance mechanism could be effective to maintain priming benefits in melon seeds.Index terms: PEG; priming; dry seeds; antioxidant system.
Peltophorum dubium (Sprengel) Taubert seedlings were submitted to 60 days of flooding, with and without Ca + . During this period, growth, root anatomy and chlorophyll content were analyzed. The relative growth rates (RGR) of roots and shoots were higher in non-flooded plants. The flooded treatments, with and without Ca + , have affected the roots growth but did not affect the shoot and the stem diameter. The anatomy analysis showed thickening of the sclerenchyma in 15 and 60 days in the flooded treatments with and without Ca + , and the vessel diameter was smaller at 45 days. The chlorophyll content differed, however it was not statistically significant. The results indicate that Peltophorum dubium as candidate specie for reforestation in areas subjected to continuous flooding for up to 60 days, since no mortality was observed. Keywords: angico amarelo, calcium application, flooded environments.
