δ-Tocotrienol feeding modulates gene expression of EIF2, mTOR, protein ubiquitination through multiple-signaling pathways in chronic hepatitis C patients

Abstract: Backgroundδ-Tocotrienol is a naturally occurring proteasome inhibitor, which has the capacity to inhibit proliferation and induce apoptosis in several cancer cells obtained from several organs of humans, and other cancer cell lines. Moreover, results of plasma total mRNAs after δ-tocotrienol feeding to hepatitis C patients revealed significant inhibition in the expression of pro-inflammatory cytokines (TNF-α, VCAM1, proteasome subunits) and induction in the expression of ICAM1 and IFN-γ after post-treatment. T… Show more

“…The vitamin E family, with powerful antioxidant and anticancer activities, , constitutes of tocotrienols and tocopherols. Each form has four isoforms, namely alpha-, beta-, gamma-, and delta-isoforms (Figure S1, Supporting Information), and the biological activities of the isomers are different in diversity and strength. Recently, tocotrienol has received wide attention because of its diverse pharmacological properties over tocopherols, such as gender differences in the antioxidant level and gene expression, , neuroprotection, cholesterol reduction, antiosteoporosis, and anticancer properties. Among the four isomers of tocotrienols, delta-tocotrienol has been investigated extensively for its most diverse biological activities for human health, in particular anticancer activities …”

“…We focused on the mTOR/AMPK/JNK/ERK/IκB-α pathway, which has an essential role in the activation of RA FLS [18]. In signaling pathways, the suppressive effects of tocotrienol are mediated through the mTOR/JNK/nuclear factor κB (NF-κB) pathway [22,23]. IL-17 activates mTOR, and tocotrienol decreased IL-17-activated mTOR phosphorylation.…”

“…Karoten berfungsi mengurangi radikal bebas, menurunkan masalah kurang vitamin A (Pignitter et al, 2016). Vitamin E (tokoferol dan tokotrienol) memiliki aktivitas sebagai hypochloesterolemic, anti-inflamatory, antikanker, antioksidan, neuroprotective, proteksi kulit dan kesehatan tulang (Qureshi et al, 2018). Tokotrienol juga berpotensi dalam pencegahan penyakit neurodegeneratif (Musa et al, 2017).…”
