β3 Integrin–EGF receptor cross-talk activates p190RhoGAP in mouse mammary gland epithelial cells

Abstract: Filopodia formation is positively regulated by β3 integrin–EGFR cross-talk, which regulates p190RhoGAP activity and localization in normal mouse mammary gland epithelial cells.

“…Recent evidence suggests that cooperation between integrins and growth factor receptors alters downstream signaling and may contribute to pathological responses of cancer cells to soluble ligands (3,4). Indeed, we have shown that integrins form physical complexes with EGFR and TGF-␤ receptors giving rise to oncogenic signaling patterns and pathological responses to ligands (5)(6)(7).…”

“…Mouse NR6 fibroblasts are an NIH-3T3 variant lacking endogenous EGFR expression (24). Construction of NMuMG and NR6 cell lines expressing human WT-EGFR or EGFR with 679,680-LL converted to AA (EGFR-AA) were described previously (5,25). Cells were maintained at 37°C in a humidified atmosphere of 5% CO 2 and 95% air in Dulbecco's modified Eagle's medium (DMEM) supplemented with 10% fetal bovine serum (FBS).…”

“…are known to cross regulate each other in stiff environments, potentiating both integrin signaling and often increasing receptor potency [107,[109][110][111]. Her2 is acutely regulated by FAK, such that ECM stiffening is a powerful regulator of Her2 and of Her2 resistance [65,110,112].…”

“…In addition, FN activates a series of signaling molecules, including focal adhesion kinase (FAK), paxillin, and Src, thus regulating cell growth and differentiation. 20,21 The superior ability of FN to promote adhesion has been confirmed and has been applied to interface modification. The application of FN to the surface of nonglycolide polymers has been successful in a large number of experiments.…”
