β-Lactam TRPM8 Antagonists Derived from Phe-Phenylalaninol Conjugates: Structure–Activity Relationships and Antiallodynic Activity

Cristina Martín-Escura,
M. Ángeles Bonache,
Jessy A. Medina
et al.

Abstract: The protein transient receptor potential melastatin type 8 (TRPM8), a non-selective, calcium (Ca2+)-permeable ion channel is implicated in several pathological conditions, including neuropathic pain states. In our previous research endeavors, we have identified β-lactam derivatives with high hydrophobic character that exhibit potent and selective TRPM8 antagonist activity. This work describes the synthesis of novel derivatives featuring C-terminal amides and diversely substituted N′-terminal monobenzyl groups … Show more

“…Our behavioural results are partially consistent with our initial hypothesis, with respect to the ability to block environmental coldactivation of TRPM8 in vivo. The antinociceptive effects of VBJ103 are in line with prior studies demonstrating antinociceptive effects of TRPM8-selective antagonists in this model (Bertamino et al, 2018(Bertamino et al, , 2020Bonache et al, 2020;Chaudhari et al, 2013;Iraci et al, 2022;Martin-Escura et al, 2023Mizoguchi et al, 2016). There was no apparent sex difference in efficacy or detectable issues observed with repeat dosing.…”
