β-glucan suppresses cell death of ASC deficient macrophages invaded by periodontopathic bacteria through the caspase-11 pathway

Abstract: β-glucan is an abundant cell wall component of fungi and yeast. Dectin-1, a β-glucan receptor, plays an important regulatory role in the natural immunity. In the present study, we investigated the effect of β-glucan on mouse macrophages that had been invaded by the periodontopathic bacterium, Aggregatibacter actinomycetemcomitans. Exposure to curdlan, a type of β-glucan, suppressed cell death and led to the accumulation of a sub-G1-phase population upon A. actinomycetemcomitans invasion under conditions of con… Show more

“…[ 63,64 ] Lastly, the most novel role for dectin‐1 involving trained immunity has been described for the regulation of cell death in murine macrophages and dendritic cells, where CARD9 has the dual role of enhancing TLR signaling through MAP kinase activation and directly promoting NF‐κB activation. “…The binding between dectin-1 and β-glucans results in the activation and recruitment of spleen tyrosine kinase (Syk). As a result, NFAT- and NF-κB-induced transcription induces cytokine and chemokine production [ 6 , 79 - 81 ]. Dectin-1 mRNA is widely expressed in several tissues, of which many are also expressed in monocytes of livers and a lungs [ 71 , 82 , 83 ].In the present study, the administration of 500 mg of Euglena /kg B.W.…”

“…They are considered to be the “front-line fighters” of the body's immune system, but invasion by germs or viruses is likely to cause apoptosis of macrophages, resulting in impaired immunity. Fermentum β-polysaccharide can bind to the Dectin-1 receptor and down-regulate the cell death induced by the invasion of macrophages by actinomycetes, ensuring the proper functioning of the body's immune system ( Inoue et al, 2019 ). It was shown that both water-soluble and granular β-polysaccharides recognize the Dectin-1 receptor, but that granular β-polysaccharides bind to the Dectin-1 receptor to better trigger the production of pro-inflammatory cytokines.…”
